This study aims to review the current practices and perceptions of dentists regarding the likelihood of dental professionals incorporating blood glucose monitoring into patient care protocol of patients with diabetes mellitus and prediabetes in dental offices. A survey link was shared by DentalOffices, a dental resource center. The survey included a series of questions which aimed to review dentists' current attitudes, practices, and considerations for implementation of in-office blood glucose testing as it relates to the treatment of patients with diabetes mellitus. A Mutational Analysis Of Thatisin: How Single Point Mutations Impact The Biosynthetic Process Of A Group 16 Graspetide, John F. Boynton Jr

Abstract:

Graspetides are a family of Ribosomally synthesized and Post-translationally modified Peptide (RiPP) natural products named for their ATP Grasp Macrocyclases. We identified several Graspetide Biosynthetic Gene Clusters containing common double glycine cleavage motifs and attempted characterization of the mature Graspetides. One of the resulting peptides, Thatisin, has a topology with two interlocking macrolactones installed by a single ATP Grasp Ligase named ThtC. Here we use single and double point mutations to observe the impact or variation on the installation of these macrocycles. Introduction

The marginalization of intersecting racial, gender, and rural identities compounds the structural vulnerabilities (e.g., discrimination, stigma, health disparities) experienced by Black women in South Carolina’s rural areas. Discriminatory experiences can deteriorate mental health; however, resilience can potentially reduce the ill effects of discrimination by decreasing associated psychological distress. This study examines the mediating role of resilience in the association between discrimination and psychological distress among rural, Black women in South Carolina in the post-pandemic era.

Purpose: Cancer immunotherapy has transformed oncology, but underserved populations face persistent disparities in access and outcomes. This review explores how artificial intelligence (AI) can help mitigate these barriers. Methods: We conducted a narrative review based on peer-reviewed literature selected for relevance to artificial intelligence, cancer immunotherapy, and healthcare challenges, without restrictions on publication date. We searched three major electronic databases: PubMed, IEEE Xplore, and arXiv, covering both biomedical and computational literature. Transcription factors such as Tbx5, Gata4, Mef2c and Pitx2 are required during cardiac development, and in adult cardiac homeostasis. We demonstrate that the gene dosage and modulation of these factors are mediated in vivo by the miR-200 family. Inhibition of a single miR-200 family member within the cluster results in defects of the left ventricle and cardiomyocyte maturation during development. Inhibition of the entire miR-200 family results in a ventricular septal defect and embryonic lethality by embryonic day (E)16.5. This quality improvement project focused on increasing the use of continuous glucose monitors (CGMs) in primary care for patients with type 2 diabetes. Its goal was to enhance providers' ability to prescribe and manage CGMs effectively. A review of existing literature and a provider survey identified key barriers, such as limited time and insufficient provider education on CGMs. The survey emphasized educational needs related to initiating and prescribing CGMs. Educational materials, including a PowerPoint presentation on prescribing, monitoring, and management, were developed and shared with clinic staff. Immune checkpoint blockade (ICB) is a commonly used treatment modality for cancer with a growing list of oncologic indications. Ototoxicity is a potential immune-related adverse event of ICB treatment, but the risk of hearing loss after ICB remains unknown. This retrospective chart review sought to identify individuals who received ICB and had available audiometric data before and after treatment in order to identify clinically meaningful changes in hearing. This single center, institutional chart review examined hearing function data in patients who underwent audiometry before and after ICB treatment. BACKGROUND: Hyams grading is considered prognostic in olfactory neuroblastoma (ONB), but grading criteria are subjective and predate modern classification and surgical approach. We evaluate the application of granular grading criteria to an endoscopic surgery predominant cohort with extended follow-up.

METHODS: 78 ONB patients were identified (1994-2019) with original diagnoses dating to 1979. Original Hyams grade, Modified Hyams grade incorporating more detailed criteria and other histologic features (i.e. divergent differentiation, clear cell/oncocytic change, and spindling) were assessed if feasible, evaluated for distribution by grade and correlated with outcomes.

During the COVID pandemic, schools were widely believed to be a venue that facilitated disease transmission. While some worried Black and Hispanic students would disproportionately face greater risk of COVID exposure at school (school COVID risk) – given well-documented racially disparate school environments and resources – little empirical evidence exists documenting or refuting potential disparities in school COVID risk. This review aimed to explore the potential of several legume species indigenous to Indonesia, from the Leguminosae family, as sources of food and medicine. Painful diabetic neuropathy (PDN), a complication of type 1 and type 2 diabetes, also occurs in obesity and prediabetes, with dyslipidemia as a modifiable risk factor. The distal-to-proximal neuropathy pattern and length-dependent intraepidermal nerve fiber loss suggest peripheral contributions. We hypothesized that elevated neurotoxic lipid intermediates—diacylglycerols (DAG), free fatty acids (FFA), and acylcarnitines (CARs)—in the dermal interstitial fluid (ISF) drive metabolism-associated hyperalgesia in prediabetic and diabetic rats. Asthma is a chronic inflammatory disease of the lungs characterized by complex pathophysiological mechanisms and diverse endotypes, primarily categorized as T helper type 2 cell (Th2)-high and Th2-low asthma. Despite significant advances in understanding asthma pathogenesis, the mechanisms governing the pulmonary immune system’s response to external challenges have not been fully understood. To address these gaps, this comprehensive project was designed to investigate the interplay between fungal infections and specific CD4+ T cell populations in asthma. The purpose of this study was to determine where in the brain there may be a difference in brain activity in patients during their first episode of psychosis (FEP) compared to healthy controls (HCs) while performing an auditory oddball task. Time-frequency analysis allows for spectral decomposition where the signal measured by magnetoencephalography (MEG) is used to characterize changes in neural oscillations and their synchronization over time and region. Previous EEG studies report reduced alpha (8-12Hz, 70-160ms) power but most studies focused on patients with chronic schizophrenia and with limited source localization.

Elizabethkingia species, particularly E. anophelis and E. meningoseptica, are gram negative bacilli known to be associated with neonatal infections. Although these bacteria are rare, they are highly concerning pathogens associated with severe sepsis, bacteraemia and meningitis in neonates and immunocompromised individuals. Managing Elizabethkingia infections presents significant challenges because of their multidrug resistance, including resistance to most β-lactam agents. They have been implicated in both nosocomial and community outbreaks worldwide, often with high mortality rates. The purpose of this study was to identify the factors contributing to dental hygienists’ consideration of changing careers, retiring early, or feeling burned out. A 26-question survey was emailed to members of the American Dental Hygienists’ Association, with 742 responses meeting inclusion criteria. Participants evaluated aspects of their careers, including job satisfaction, wages and benefits, personal growth, work-related stress, burnout, work-life balance, and retirement plans. Results revealed that one-third of respondents planned to leave the profession, citing burnout, physical strain, and limited career growth as reasons. Organic acids, as natural metabolites, play crucial roles in human metabolism and health. Tumor Necrosis Factor (TNF), a pivotal mediator in immune regulation and inflammation, is a key therapeutic target. We evaluated ten organic acids as TNF modulators using in silico molecular docking, followed by detailed ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) profiling and molecular dynamics (MD) simulations for three lead candidates: gallic, aconitic, and crocetin acids. Purpose:

Dynamic Temporal and Tactile Cueing (DTTC) is an evidence-based treatment for children with childhood apraxia of speech. In clinical practice, speech-language pathologists (SLPs) frequently use an eclectic approach incorporating aspects of DTTC, for reasons that are not well understood. This research note describes a qualitative investigation of the experiences of SLPs and caregivers who participated in protocolized research DTTC for a 24-session treatment efficacy study. Specifically, we sought to understand the perspectives of these SLPs and caregivers regarding (a) DTTC treatment, as provided during the study, and (b) participating in a treatment research study.

Blood stem cells are among the body's longest-living cells despite being highly vulnerable to proteotoxic damage, which accelerates their aging. To maintain protein homeostasis (proteostasis), hematopoietic stem cells (HSCs) employ mechanisms such as reduced translation rates, high chaperone activity, autophagy, and selective protein degradation. These strategies mitigate protein misfolding, maintain quiescence, and preserve regenerative potential. Disruptions in proteostasis can lead to the elimination of impaired HSCs through differentiation or apoptosis, ensuring the integrity of the stem cell pool. Clay masks are cosmetic formulations designed to adhere closely to the skin, effectively lifting contaminants, cleansing the face, and absorbing impurities. We made a clay mask containing active ingredients such as niacinamide, alpha arbutin, ceramide, and hyaluronic acid. A careful selection of ingredients is essential for ensuring safety and mitigating potential dermatological issues. This study aimed to evaluate the physical characteristics, stability, primary irritation potential, and moisturizing efficacy of this clay mask formulation. Despite the success of targeted inhibitors in cutaneous melanoma, therapeutic responses are limited by the aged tumor microenvironment and drug-tolerant residual cells. Given the similarities between drug tolerance and cellular dormancy, we studied the dormancy marker, nuclear receptor subfamily 2 group F member 1 (NR2F1), in response to BRAF-V600E inhibitors (BRAFi) plus MEK inhibitors (MEKi) in BRAF-mutant melanoma models. Transcriptomic analysis of melanoma patient samples treated with BRAFi + MEKi showed increased NR2F1. NR2F1 was highly expressed in the drug-tolerant invasive cell state of minimal residual disease in patient-derived and mouse-derived xenografts on BRAFi + MEKi. Despite the critical role of DNA methylation, clinical implementations harnessing its promise have not been described in acute myeloid leukemia. Utilizing DNA methylation from 3314 leukemia patient samples across 11 harmonized cohorts, we describe the Acute Leukemia Methylome Atlas, which includes robust models capable of accurately predicting AML subtypes. A genome-wide prognostic model as well as a targeted panel of 38 CpGs significantly predict five-year survival in our pediatric and adult test cohorts. To accelerate rapid clinical utility, we develop a specimen-to-result protocol that uses long-read nanopore sequencing and machine learning to characterize patients' whole genomes and epigenomes. Background: Helicobacter pylori infects approximately half of the global population, leading to gastric and duodenal ulcers. Despite the availability of antibiotics, challenges such as patient reluctance, high treatment costs, and antibiotic resistance limit their effectiveness, making vaccination a promising alternative. This study used immunoinformatics to identify candidate epitopes for a multiepitope vaccine construct against H. pylori.

Material and methods: The protein variability server was utilized for conservation analysis. The epitopes were screened for antigenicity, allergenicity, toxicity, cross-reactivity, and population coverage. Introduction: SIRPα+ macrophages can mediate resistance to lenalidomide and rituximab (R2) in patients with B-cell non-Hodgkin lymphoma (B-NHL). Evorpacept (ALX148) is a novel CD47 blocker that abrogates interactions between lymphoma cells and SIRPα+ macrophages.

Methods: Adult patients with B-NHL who had received at least 2 prior lines of systemic therapy were included in this single arm phase I study (NCT05025800).
