IL-22, a signature cytokine for type 3 lymphoid cells, including T helper 17/22 (Th17/22) and type 3 innate lymphoid cells (ILC3), mediates epithelial homeostasis and protective pathogen responses in barrier tissues. Upon dysregulation, IL-22 can drive chronic inflammatory diseases, yet little is known about transcriptional elements modulating its expression. Here, we identify two enhancers, E22-1 and E22-2, with distinct capacities for regulating Il22 expression in type 3 lymphoid cells. Both enhancers are necessary for protection from Citrobacter rodentium infection and for the onset of IL-22-mediated psoriasis. E22-2 is specifically required for IL-22 expression in ILC3s, while E22-1 functions in both …

Objectives: Cancer is still one of the leading causes of death worldwide and a high percentage of patients with cancer face emotional challenges such as guilt and self-blame. Early maladaptive schemas (EMSs) can make these feelings even stronger. This study aimed to explore how guilt and self-blame relate to EMSs in individuals with cancer, while taking overall psychological distress into account. Methods: This cross-sectional study was conducted at Rasoul-e-Akram and Firoozgar hospitals in Tehran, Iran, from October 2022 to June 2023. Background and objectives: Re-irradiation with stereotactic radiosurgery (SRS) is an increasingly used treatment for recurrent high-grade glioma (HGG). We analyzed the patterns of failure after salvage SRS to better identify which patients would most benefit.

Methods: We reviewed all patients who received SRS for recurrent HGG over 12 years at our institution. Pattern of failure was defined by the distance of recurrence from the SRS isodose line. Overall-survival (OS), progression-free survival (PFS), and local-failure-free survival (LFS) were estimated using Kaplan-Meier methods, and regression model analysis was performed to analyze clinical predictors of failure.

Background: Tenosynovial giant cell tumour (TGCT) is a rare, locally aggressive neoplasm that affects otherwise healthy adults. There are few systemic treatment options, highlighting an unmet need. We report the results of part 1 of the MANEUVER trial, which aimed to evaluate the efficacy and safety of pimicotinib, a highly selective, potent, colony-stimulating factor-1 receptor inhibitor, in patients with TGCT.

Semi-automated drug delivery systems, such as closed-loop insulin pumps, smart infusion devices, and patient-controlled analgesia (PCA) tools, are transforming healthcare by improving dosing precision and empowering patients with greater autonomy. However, their safety and efficacy depend critically on trust calibration—the dynamic alignment between patient trust in automation and actual system reliability (Hoff & Bashir, 2015; Lee & See, 2004). Miscalibrated trust can lead to misuse or disuse, jeopardizing patient safety in high-stakes contexts (Fallon et al., 2010).
