Development Of An On-Line Computer System For Enzyme Kinetics Studies, Jacqueline R. Thweatt

The PDP-11/10 minicomputer was interfaced to the Beckman 25 ultraviolet/visible spectrophotometer and this on-line computer system was applied to enzyme kinetics studies. This interface involved hardware equipment to locate, amplify, and transmit the analog signal to an analog-to-digital converter, and software to acquire and store the digital data. Data processing programs were used to calculate kinetic parameters and statistical information from the acquired data.
The Varian Cary 219 ultraviolet/visible spectrophotometer can be readily interfaced to the Apple II Plus microcomputer by hardware equipment which can be purchased from the manufacturer. Various software may also be purchased for use in enzyme …

Human serum high density lipoprotein subclasses, HDL2 and HDL3 , were isolated by preparative salt density gradient ultracentrifugation and further analyzed by electrophoresis on a 4 to 15% concentration gradient polyacrylamide gel.
The separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation for 272,000 g at 15°c in a swinging bucket rotor. High resolution concentration gradient gel electrophoresis was found to be particularly suitable for the separation of lipoproteins. The isolation of HDL by precipitation methods, heparin-manganese and dextran-magnesium have also been evaluated in this study, and were found not to completely separate …

The purpose of this study was to survey Old Dominion University dental hygiene master's degree graduates and their employers to assess the overall quality of the graduates' education. Individuals who graduated between May 1976 and December 1981 (N =29) were sent a self-designed questionnaire to obtain evaluative data about the ODU dental hygiene master's degree program. The graduates were asked to identify strengths and weaknesses of the ODU master's degree program, the role competencies required for their current position, and the degree to which they were prepared for their current job by the master's degree program. The cloning of DNA fragments derived from the (beta)-like globin gene cluster of normal and (beta)('+)-thalassemic individuals is described. These and other cloned human DNA fragments are employed to define the strucure and transcription of a family of interspersed, repetitive RNA polymerase III transcription units in human DNA. These data allow unequivocal identification of these transcription units with the Alu family of interspersed, repetitive DNA sequences. Comparison of these sequences to those of other primate and rodent interspersed repeats reveals that the primate sequence resembles in many respects a dimer of the rodent sequence. The literature on esophageal speech has identified the problem of extraneous air intake noise, suggested its possible etiology, and provided practical advice for clinical management. Documentation on the efficacy of specific methodology is lacking in the literature. Such documentation would be simplified if objective criteria were used to rate the severity of intake noise. The present study was prompted by the lack of basic data regarding listener evaluation of intake noise.

The present study was designed to investigate the perceptual and spectrographic features of vocal effort in the speech of severely to profoundly hearing-impaired children and their normal hearing agemates. Recorded vowel and speech samples were obtained from ten normal hearing children, ten severely to profoundly hearing-impaired children attending Oral/Aural educational programs, and eight severely to profoundly hearing-impaired children attending Total Communication programs. The degree of perceived vocal effort for vowels and speech was evaluated, using a nine point equal-appearing-interval scale. A woman with medullary carcinoma of the thyroid and a past history of breast cancer was found to have metastatic bone lesions. It was initially assumed that this was due to recurrent breast carcinoma, but bone biopsy revealed medullary carcinoma of thyroid origin. Although clinical bone metastasis is a rare event in patients with medullary carcinoma, quite clearly such an occurrence is possible. Medullary carcinoma should be considered in the differential diagnosis of patients with metastatic bone lesions.

The differential diagnosis of hemoptysis in patients with mitral stenosis includes many primary pulmonary problems besides those peculiar to mitral stenosis. We describe the case of a 40-year-old woman with a 40-pack-year smoking history whose presenting symptom of hemoptysis was wrongly ascribed to pulmonary embolus and infarction, based on a presumably positive angiogram, and to chronic bronchitis, based on endoscopic findings at fiberoptic bronchoscopy, as provided by the referring physician. The features differentiating between cardiac and primary pulmonary disease are described, and the pathophysiology of hemoptysis in mitral stenosis is reviewed. Creatine kinase (CK) and lactate dehydrogenase (LD) are isoenzymes that have been measured in clinical laboratories for over 20 years; their separation has proved valuable in the diagnosis of myocardial infarction and other cardiac-related diseases. Although, historically, electrophoresis was the preferred method to measure both isoenzymes, there has been controversy over the best method for separating the CK isoenzyme. lon exchange chromatography has been used successfully to isolate CK isoenzymes, and the procedure was adapted to determine CK-MB activity in serum. Hypokalemic periodic paralysis is an uncommon disease that has rarely been reported in blacks. Historical, clinical, and pathological findings in three black patients, in addition to those previously reported in the literature, suggest that the spectrum of the disease is identical in both blacks and whites. Considering the disease as either different or unknown in the black population is probably not justified.
