Nucleocytoplasmic trafficking is a highly regulated process that allows the cell to control the partitioning of proteins and nucleic acids between the cytosolic and nuclear compartments. The Ebola virus minor matrix protein VP24 (eVP24) hijacks this process by binding to a region on the NPI-1 subfamily of karyopherin alpha (KPNA) nuclear importers. This region overlaps with the activated transcription factor STAT1 binding site on KPNAs, preventing STAT1 nuclear localization and activation of antiviral gene transcription. The differentiation and suppressive functions of regulatory CD4 T cells (Tregs) are supported by a broad array of metabolic changes, providing potential therapeutic targets for immune modulation. In this study, we focused on the regulatory role of glycolytic enzymes in Tregs and identified phosphoglycerate mutase (PGAM) as being differentially overexpressed in Tregs and associated with a highly suppressive phenotype. Pharmacologic or genetic inhibition of PGAM reduced Treg differentiation and suppressive function while reciprocally inducing markers of a pro-inflammatory, T helper 17 (Th17)-like state. Individuals infected with coronavirus disease 2019 (COVID-19) have experienced torrents of problems associated with the pandemic, impairing their health, mental, social, and economic conditions. This study aimed to determine the psychological reactions and emotions of patients with COVID-19 infection and the social consequences during hospitalization. The current study used a qualitative and phenomenological design at a training and research hospital in Istanbul, Turkey. The study sample included 17 patients (41.6 ± 16.5 years) who had been discharged after COVID-19 treatment. Modern walking often involves secondary tasks that may increase fall risk, especially in older adults. About one-third of adults aged 65+ fall annually, with dual-tasking further elevating this risk. This study examines how cognitive and motor dual-tasks affect gait variability. Ten healthy young adults completed baseline and three dual-task conditions: (1) walking with a Stroop test, (2) walking while carrying a tray with water, and (3) both tasks combined. Gait parameters—including step length, step width, stride length, and double support time—were analyzed. This study examines the combined effects of green noise and texting on gait to understand how multitasking and auditory stimuli affect walking stability. Green noise, associated with relaxation, was the environmental stimulus; texting the cognitive distractor. We hypothesized texting would reduce cadence and stride length, with green noise either stabilizing or disrupting gait. Ten participants walked on a treadmill under five conditions: normal walking (NW), no texting/sound (WNTNS), sound only (WNTS), texting only (WTNS), and texting with sound (WTS). Introduction: Leadless intracardiac pacemakers (LICPs) reduce lead-related complications but may be malpositioned in patients with unrecognized structural defects. We report a 69-year-old man with high-grade AV block whose LICP, intended for right ventricular placement, was found in the left ventricle via an undiagnosed ostium secundum ASD after presenting with multifocal embolic strokes 16 months later. Case Presentation: Stroke workup for acute right-sided symptoms revealed the LICP in the LV and a large, previously undiagnosed ASD with bidirectional shunting and severe pulmonary hypertension. Embolic infarcts were attributed to either paradoxical embolism or device-related thromboembolism. Due to high surgical risk, …

Allegedly named after a dog show judge, our late dog Rodi was never a Recovery-Oriented Dog Intervention (RODI). Through collaborative-duo-autoethnography, we explore how our mundane co-existence with Rodi has influenced our perspectives on health struggles, recovery, ontology, and what really matters when life becomes difficult, and push comes to shove. We have shared texts describing our memories of how Rodi entered and influenced our lives. Through our collaborative writing we have discovered similarities and differences. Drawing inspiration from philosophy, posthumanism, and new materialism, we argue that our four-legged companion not only changed our everyday lives, but also our perspectives on …

Unresolved tissue damage is a common feature of Inflammatory Bowel Disease (IBD) that facilitates disease progression. Here, we showed that high animal fat diets (HFD), an environmental risk factor associated with IBD pathogenesis, suppress intestinal macrophage production of critical tissue repair responses after damage. This includes reduced IL-23 production, which drives downstream production of IL-22, which is needed for barrier repair. Indicating that dietary lipids interfere with responses to microbial molecules needed to induce barrier protective functions, we found oleic acid could directly suppress macrophage Il23a induction after lipopolysaccharide (LPS) treatment. Colorectal liver metastasis (CRLM) poses a significant challenge in oncology due to its high incidence and poor prognosis in unresectable cases. Current treatments, including surgical resection, systemic chemotherapy, and liver-directed therapies, often fail to effectively target hypoxic tumor regions, which are inherently more resistant to these interventions. This review examines the potential of a novel therapeutic strategy combining irreversible electroporation (IRE) ablation and Clostridium novyi-nontoxic (C. novyi-NT) bacterial therapy. Military personnel deployed to Iraq and Afghanistan were exposed to emissions from open-air burn pits, where plastics, metals, and medical waste were incinerated. These exposures have been linked to deployment-related respiratory diseases (DRRD) and may also impact neurological health via the lung-brain axis. To investigate molecular mechanisms, adult male rats were exposed to filtered air, naphthalene (a representative volatile organic compound), or a combination of naphthalene and carbon black (surrogate for particulate matter; CBN) via whole-body inhalation (six hours/day, three consecutive days). Background: Orthodontic miniscrew anchorage induced a paradigm shift in orthodontics, by avoiding unwanted movement of anchoring teeth. Failure of miniscrew anchorage (premature loosening of the miniscrew) is known to be related to root proximity after insertion. Objective: This study aimed to evaluate root proximity of orthodontic miniscrew implants using CBCT and to more precisely investigate the correlation between root proximity and miniscrew failure. Materials and methods: CBCT images of 47 miniscrews, placed buccally between maxillary first molars and second premolars, were used in this retrospective study. Introduction: Ingrown toenails are one of the most common nail conditions and are usually treated with chemical matricectomy using phenol (CMP). However, this procedure is often contraindicated in patients with diabetes mellitus or peripheral vascular disease. Orthonyxia, or nail braces application, is a non-invasive alternative treatment for ingrown toenails. The nail braces are spring wires with one side shaped into a hook that is placed under one side of the nail, and the other side attached to a plastic pad that is glued to the nail plate. Atherosclerosis, a major cause of mortality and morbidity in the United States (US), is driven by complex factors, including the cumulative burden of low-density lipoprotein cholesterol (LDL-C) and other atherogenic lipids and lipoproteins. Lipid management in US primary care aims to prevent atherosclerotic cardiovascular disease (ASCVD) events through screening and LDL-C lowering to risk-based goals. However, lipid screening and ASCVD treatment is often inadequate, with few patients meeting LDL-C goals in clinical practice. During mouse embryonic brain development, the Evf2 ultraconserved enhancer (UCE) lncRNA guides the Dlx5/6UCE to ~129 sites across chr6. However, previous work identified only 4 transcriptionally regulated targets associated with Evf2-Dlx5/6UCE-gene guidance, raising questions about the significance of Evf2-regulated Dlx5/6UCE-gene interactions. Here, single-cell transcriptomics reveal far greater alignment between Evf2-Dlx5/6UCE-gene guidance and transcriptional regulation than previously reported. Evf2 divides chr6 into short-range ( < 10 Mb distant), activated genes, and long/super-long-range (10-129 Mb distant), repressed genes, identifying seizure regulating genes in the embryonic subventricular zone that predict adult phenotypes. Evf2-regulated Dlx5/6UCE-gene distances and directions (closer to or further from gene targets) can be decoupled from gene target transcriptional effects. Evf2 regulates Evf2-ribonucleoprotein (RNP) binding in a combinatorial manner to key regulatory sites, including chr6 Evf2-Dlx5/6UCE-gene guided sites, Evf1/2 RNA-directly bound sites (RBSs), and inter-chromosomal HiC looping interactions. RBSs divide chromosomes into multi-megabase domains enriched for Evf2-regulated RNP recruitment, transcription factor motifs, and HiC looping interactions. Together with Evf2-controlled homeobox motif recognition at Evf2-RNP recruitment sites and transcription factor motif enrichment in RBSs with DNA identity, this work supports direct roles for Evf2 in enhancer-gene guidance and transcriptional regulation, with the potential for both site-specific and chromosomal domain specific RNP recruitment.

BACKGROUND: A key cardiac magnetic resonance (CMR) challenge is breath-holding duration, difficult for cardiac patients.
AIM: To evaluate whether artificial intelligence-assisted compressed sensing CINE (AI-CS-CINE) reduces image acquisition time of CMR compared to conventional CINE (C-CINE).
METHODS: Cardio-oncology patients (
RESULTS: In 89 participants (58.5 ± 16.8 years, 42 males, 47 females), total AI-CS-CINE acquisition and reconstruction time (37 seconds) was 84% faster than C-CINE (238 seconds). C-CINE required repeats in 23% (20/89) of cases (approximately 8 minutes lost), while AI-CS-CINE only needed one repeat (1%; 2 seconds lost). The primary objective of this study is to examine the relationship between stigma and physical disability, to discover the impact that perceived stigma has on several factors in a person’s life including overall health, well-being, and quality of life. A qualitative research design was used. Six semi-structured interviews were conducted to examine the relationship between stigma and physical disability, and to discover the impact of stigma on overall health and well-being. Rising youth public health disparities can be attributed to the social determinants of health that influence health and wellness. Youth sport providers can be influential in creating healthier environments that facilitate positive youth development and social emotional learning, thus positively impacting life-long health behaviors choices and public health. Eccentric muscle actions are integral to human movement, rehabilitation, and performance training due to their characteristic high force output (overload) and low energy cost and perceived exertion. Despite the growing use of eccentric devices, a gap in the research exists exploring multi-muscle activation profiles during multi-joint eccentric-only, isokinetic exercise. This study aimed to quantify and compare surface electromyographic (EMG) activity of four leg muscles—vastus lateralis (VL), tibialis anterior (TA), biceps femoris (BF), and medial gastrocnemius (GM)—during a standardized (isokinetic) submaximal eccentric multi-joint exercise using the Eccentron dynamometer. Background/Objectives: Dietary modulation of the gut microbiome is a promising strategy for mitigating gastrointestinal diseases, such as inflammatory bowel disease (IBD) and colitis-associated colorectal cancer (CAC). Cocoa powder is rich in polyphenols, including (-)-epicatechin and (+)-catechin, which have been associated with beneficial effects on gut health and microbiome modulation. Due to their clinical heterogeneity, nonspecific symptoms, and the limitations of existing biomarkers and imaging modalities, metabolic brain diseases (MBDs), such as mitochondrial encephalopathies, lysosomal storage disorders, and glucose metabolism syndromes, pose significant diagnostic challenges. This review examines the growing potential of cell-free DNA (cfDNA) derived from cerebrospinal fluid (CSF) epigenetic profiling as a dynamic, cell-type-specific, minimally invasive biomarker approach for MBD diagnosis and monitoring.
