Background: Erdafitinib is approved for locally advanced/metastatic urothelial cancer (LA/mUC). As enfortumab vedotin (EV) plus pembrolizumab enters frontline management, outcomes with erdafitinib pre- and post-EV are clinically relevant but not specifically evaluated in clinical trials.

Methods: UNITE is a multi-institutional retrospective study of patients with LA/mUC treated with novel targeted agents. All patients with FGFR3 alterations treated with EV only, erdafitinib then EV (Erda->EV), and EV then erdafitinib (EV->Erda) were included. Sequential treatment with EV and Erda was not required. Primary endpoints were observed response rates (ORR) and progression-free survival (PFS); secondary endpoint was overall survival (OS).

Background: Population-wide genomic screening (PGS) for genetic conditions such as hereditary breast and ovarian cancer syndrome, Lynch syndrome, and familial hypercholesterolemia presents opportunities to reduce morbidity and mortality among the 1%-2% of the population at elevated risk for these serious, preventable diseases. With decreasing sequencing costs and growing support from national bodies, there are increasing numbers of PGS programs in the United States. However, guidelines and strategies to support implementation are limited, especially regarding equitable access to PGS. Biliverdin reductase A (BVRA), the terminal enzyme in heme catabolism, generates the neuroprotective and lipophilic antioxidant bilirubin. Here, we identify a nonenzymatic role for BVRA in redox regulation. Through phylogenetic, genetic, biochemical, and enzymatic assays, we found that BVRA exerts critical nonenzymatic antioxidant activity. Transcriptomic analyses further revealed that BVRA physically and genetically interacts with nuclear factor erythroid-derived factor-like 2 (NRF2), a major transcriptional regulator of cellular redox signaling. ChIP-seq and RNA-seq analyses reveal that BVRA and NRF2 coordinate the expression of antioxidant genes, many of which are typically dysregulated in neurodegenerative conditions such as Alzheimer's disease. Sickle cell disease (SCD), an inherited blood disorder caused by a mutation in the β-globin gene, is characterized by sickle erythrocytes that are prone to hemolysis, leading to anemia and vaso-occlusion crises. In sickle erythrocytes, hemoglobin aggregation is followed by altered cation permeability and subsequent dehydration. Interventions that restore cation permeability can decrease hemolysis and ameliorate the symptoms associated with SCD. PIEZO1 is a nonselective mechanosensitive cation channel that regulates erythrocyte volume. Gain-of-function (GOF) mutations in PIEZO1 cause hemolytic anemia by increasing cation permeability, leading to erythrocyte dehydration in humans and mice. The ryanodine receptor 1 (RYR1) is the sarcoplasmic reticulum (SR) Ca2+ release channel required for both skeletal muscle contraction and Ca2+ leak. Mutations in RYR1 cause malignant hyperthermia susceptibility (MHS) and enhanced sensitivity to heat stroke (ESHS), which can result in death due to excessive skeletal muscle thermogenesis upon exposure to volatile anesthetics or heat. Here, we investigated the molecular and physiological functions of phosphorylation of RYR1 at Ser2902 by the kinase SPEG (striated muscle preferentially expressed protein). Directional cell migration by pulmonary arterial cells (PACs) is one of the important features of diseases involving arterial remodeling, such as pulmonary arterial hypertension (PAH), a disease that is often characterized by reduced arterial compliance and increased extracellular matrix (ECM) stiffening. However, there are no therapeutics that can halt the directional cell migration of PACs in PAH. Background: The Stroke Preclinical Assessment Network tested 6 therapeutic interventions initiated at the time of reperfusion after focal ischemic stroke in young mice, aging mice, obese mice, and spontaneously hypertensive rats. This randomized, controlled trial was conducted across 6 sites with concealed treatment and blinded neurobehavior assessments. The trial had an adaptive design with preset levels of efficacy and futility interrogated after each of 4 stages. The primary outcome was turning preference on the corner test at 1 month. Clinical trials support the iterative advancement of modern medicine. However, challenges in achieving population-representativeness or participant sampling commensurate with the burden of disease can limit the generalizability and reproducibility of trial results. Here, we present the recruitment strategies and cohort profile of the Engaging Adolescents in Decisions about Return of Genomic Research Results non-randomized clinical trial (NCT0448106), where traditional, targeted hybrid, and digital recruitment methods were implemented with quota sampling to enroll diverse adolescents (ages 13-17) and young adults (ages 18-21). Objectives: This study investigates the effectiveness of ankle distraction arthroplasty (ADA) using the Ilizarov frame in treating end-stage ankle osteoarthritis (OA) by evaluating changes in cartilage volume through semi-automatic MRI segmentation. Methods: The study involved 42 end-stage post-traumatic ankle OA patients from 2021 to 2023. Magnetic resonance imaging (MRI) scans were conducted before surgery and 12 months post-treatment. Changes in cartilage volume of the talus and tibia were measured using semi-automatic segmentation methods. statistical significance was assessed using the non-parametric Wilcoxon signed-rank test. OBJECTIVES: Clinical laboratory operations consume large amounts of electricity and water and generate considerable quantities of solid waste. In this study, we quantified and compared the estimated solid waste produced from performing one million basic metabolic panels (BMPs) across five major high-throughput chemistry analyzer systems commonly used in the United States. Additionally, we estimated the carbon emissions associated with the electricity and water usage of each analyzer to better understand their environmental impact.

Human noroviruses (HuNoVs), especially GII.4 strains, are the leading cause of acute viral gastroenteritis worldwide, yet no approved vaccines or antivirals exist. The pandemic GII.4 Sydney 2012 strain enters cells via membrane wounding and clathrin-independent carrier-mediated endocytosis, but it is unclear whether this entry mechanism is conserved across GII.4 variants. We compared early binding and entry of multiple GII.4 variants using wild-type and mutant GII.4 virus-like particles (VLPs) and modified human intestinal enteroid cultures. Only a subset of GII.4 variants, including GII.4 Sydney, form distinct, histo-blood group antigen (HBGA)-dependent capsid clusters on the cell surface. Biological tissues exhibit sharp phase transitions where cells collectively transition from disordered to ordered states at critical densities. We demonstrate through bio-chemo-mechanical modeling that this emergent behavior arises from a nonmonotonic dependence on nonlinear extracellular matrix (ECM) mechanics: mechanical communication between cells is optimized at intermediate stiffness values where cells can both generate sufficient forces and create strain-stiffened tension bands in the ECM. This balance establishes a critical cell spacing threshold for cell-cell communication ([Formula: see text]100 to 200 [Formula: see text]m) that is conserved across experimental observations for a broad range of cell types and collagen densities. Rapid diagnostic testing can improve pathogen detection and lead to targeted antibiotics. The BioFire FilmArray Pneumonia Panel (BFPP) is a multiplex PCR that has displayed strong concordance with traditional microbiologic techniques. However, most existing literature focuses on deep respiratory specimens, and there is sparse literature on performance in sputum specimens. Traumatic brain injury (TBI) is an environmental risk factor for dementia and long-term neurological deficits, posing a significant public health challenge. TBI-induced neuroinflammation involves both brain-resident microglia and peripheral monocyte-derived macrophages (MDMs). Previous research has shown that MDMs contribute to the development of long-term memory deficits, yet their long-term behavior following brain infiltration remains unclear. To address this, our study uses two complementary fate-mapping mouse lines, CCR2-creERT2 and Ms4a3-cre, for precise and lasting tracking of MDMs in vivo. Wearable Electroencephalography (EEG) is a noninvasive method for assessing brain activity. Outside of the context of a traditional medical or laboratory setting, this technology may provide insight into the study of brain activity during activity or sport. In golf, for example, similar research revealed associations in skilled golfers between activity in the alpha brain wave and conscious control of motor activity during putting. This study presents a longitudinal analysis of the qualitative experiences and quantitative outcomes associated with integrative, interdisciplinary teaching in the context of a specific nursing course, Pathophysiology (NURS 337), at a public, 4-year, state college (now university). Spanning an 8-year period, from 2014–2022, the research compares two distinct student cohorts: one exposed to collaborative interdisciplinary teaching methods, the other to traditional non-collaborative instructional approaches. The findings reveal that integrative, interdisciplinary teaching yielded significant benefits across multiple metrics. Streptococcus Agalactiae endocarditis is a rare clinical entity in newborns with a normal structured heart. It is generally characterized by acute onset, the presence of large vegetations, rapid valvular destruction and frequent complications, particularly embolization. Mortality is high with medical therapy alone. We are reporting a rare case of infective endocarditis (IE) with mitral valve vegetation in a 9-day old healthy newborn following Streptococcus Agalactiae sepsis, complicated by meningitis and micro-abscesses. Mitral valve (MV) vegetation was removed surgically; the patient received intravenous antibiotics for six weeks. Objective This poster presents a four-week literature review elective for medical students, co-designed and co-taught by library and medical faculty. We will share course materials, including session topics, readings, assignments, and feedback from both instructors and students.

Methods The elective was first offered in June 2025 and taught by two library faculty members, one basic science faculty member, and one clinical faculty member. Medication reconciliation errors are a significant clinical concern because they have the potential to result in serious or even life-threatening harm to patients. Implementing risk-reduction strategies through targeted education and systematic processes for medication reconciliation is crucial to enhancing patient safety and mitigating the risk of errors. Over 40% of medication errors occur during patient transitions, such as admissions, which are mainly due to inadequate medication reconciliation. Introduction:

Sleep plays a crucial role in memory consolidation, not only stabilizing newly encoded information but also potentially supporting forgetting. Yet it remains unclear how sleep prioritizes what is retained or discarded when multiple salience cues, such as emotional valence and top-down instructional goals, compete for consolidation.

Methods:

In two studies, we examined how emotional content and intentional memory instruction interact to shape memory performance across a 12 h interval that included either nocturnal sleep or wakefulness. Participants completed a directed forgetting paradigm with neutral and negatively valenced words, followed by immediate recognition and delayed free recall.

Alloplastic implants emphasize the importance of a comprehensive understanding of both clinical and histological aspects, which are attributed to enhancing patient satisfaction and surgical success rates. The study aims to assess the viability of different implant materials and their related risks over time. A systematic literature search was conducted across multiple databases, including PubMed, Cochrane, and EBSCOhost, to identify 79 records. After eliminating duplicates and applying specific inclusion and exclusion criteria, four new studies were included for evaluation, focusing on postoperative outcomes related to alloplastic implants in rhinoplasty. Background: Obesity is thought to increase cardiovascular disease (CVD) risk partly through dyslipidemia. Yet, obesity's effects on dyslipidemia are not uniform. Understanding the shared genetic basis between obesity and lipid traits can provide insight into this heterogeneity and its implications for CVD risk.

Methods: We examined local genetic correlations between three lipid measures [high-density lipoprotein cholesterol (HDL), low-density lipoprotein cholesterol (LDL), and triglycerides (TG)] and body mass index (BMI) using genome-wide association study summary statistics from European ancestry UK Biobank participants.
