In interwar America, the relatively young profession of dentistry was developing rapidly. As the government, health institutions, and social organizers focused more intently on notions of national wellness, questions about how to achieve a healthy smile—and what a healthy smile should look like—proliferated. Groups like the National Dental Association worked to establish rigorous standards of technique, define ethical behavior, and encourage dentists to develop business acumen, yet the surprising proliferation of blues songs about dubious dentists tricking unassuming clients, or about fraught love affairs allegorized by disastrous dental exams, highlights the equivocal nature of many developments in early dentistry.

Natural polysaccharides, such as chitosan, offer promising avenues for drug delivery due to their cytocompatibility and ability to interact with cell surfaces. However, the endothelial glycocalyx, a glycan-rich extracellular matrix, presents a barrier that must be navigated for effective intracellular delivery. This study investigates how cationic poly(glucosamine)-based polymers, functionalized with guanidinium or ammonium groups, interact with key glycocalyx components including hyaluronan (HA) and heparan sulfate (HS). We demonstrate that these cationic polymers form tunable biomolecular condensates with glycans, with stronger binding observed for sulfated glycans, HS and heparin, than unsulfated HA. The Dental Clinical Humanities Certificate Program was implemented 10 years ago at the UTHealth Houston School of Dentistry to expose dental students to dental humanities content and engage them in educational experiences exploring oral health care through humanities lenses, providing insight into the human condition and patient-centered approaches. With more than 30 students having graduated from the program, we conducted a retrospective analysis of the exit surveys previously distributed at the end of each academic year. Modern-day research ethics training often begins with a discussion of the medical research atrocities committed by Nazi health care professionals and the resulting 10-principle Nuremberg Code, which is often considered to be the beginning of modern-day medical research regulations. It is occasionally mentioned that Nazi dentists also conducted unethical research in the concentration camps, and a few were brought to trial. In this manuscript, I compare two studies (one medical and one dental) conducted after World War II that violated the Nuremberg Code.

As new information becomes available, they may need to update their understanding of the situation and possibly revise their preferences. During a decision, as a preference develops for one alternative course of action, a nonconscious goal of maintaining consistency between that preference, however tentative, and the new information can lead to an interpretation of that information so as to support the current preference. Buprenorphine (BUP) is widely used in the treatment of neonatal opioid withdrawal syndrome (NOWS). However, the most compounded formulation contains 30% ethanol, despite regulatory and clinical concerns regarding ethanol exposure in pediatric patients. Thus, this research aimed to develop an ethanol-free sublingual (SL) gel formulation of BUP that would be safe, stable, and suitable for NOWS. Multiple polymers were screened as gelling agents, with hydroxypropyl methylcellulose (HPMC) emerging as the ideal base polymer for the formulation due to its optimal pH, rheological characteristics, and stability. Mechanisms responsible for skeletal muscle kidney crosstalk have not been defined. We have determined that a circulating mediator, signal regulatory protein α (SIRPα), impairs intracellular insulin-mediated functions. To elucidate the effect of myokine SIRPα on diabetic kidney disease (DKD), flox mice and muscle-specific (m-specific) SIRPα-KO mice were subjected to an obesity-induced model of diabetes, high-fat diet (HFD; 60%) or insulin-deficient hyperglycemia model, streptozotocin (STZ), and were subsequently exposed to anti-SIRPα monoclonal antibodies. In the obesity-induced diabetic mice, serum SIRPα increased. Purpose

The role of adjuvant fractionated radiotherapy (aFRT) after gross total resection (GTR) of WHO-2 meningiomas remains unclear. We aimed to estimate the effect of aFRT on recurrence risk and survival following GTR and subtotal resection (STR).

Methods

We analyzed 1452 patients with WHO-2 from our international, multicenter database (followed between 1989 and 2019). Outcomes were recurrence (10-year follow-up) and death (5-year follow-up). Risk estimates were obtained using competing risks and survival analysis. This case study describes the integration of a board-certified music therapist into two specialty dental clinics at the University of Rochester Medical Center’s Eastman Institute for Oral Health. Initiated in collaboration with the Eastman Performing Arts Medicine Center, the effort reflects a humanistic and biopsychosocial approach to dentistry that leverages music therapy to reduce anxiety, improve patient experience, and support better clinical outcomes.

Before implementation, monthly interdisciplinary planning meetings were held, bringing together dental faculty, research and administrative staff, and the music therapy team. Activation of cGAS-STING signaling in cancer cells requires cytosolic DNA produced by intrinsic or treatment-induced DNA damage. However, clinical efforts to exploit this pathway to improve immunotherapy have yielded limited success, highlighting gaps in understanding the link between DNA damage and immunotherapy. Here, we identify ubiquitination-directed cytosolic DNA degradation as a critical determinant for cGAS-STING activation following DNA damage. Mechanistically, the cytosolic DNA exonuclease TREX1 is degraded by the E3 ubiquitin ligase SPOP but is reversely stabilized by the deubiquitinase USP7. Cancer-associated SPOP mutations or USP7 overexpression elevate TREX1 levels, promoting cytosolic DNA degradation and impairing cGAS-STING-mediated immune activation. Notably, …

The co-evolution of different cell subsets in the progression of precursor lesions to lung adenocarcinoma (LUAD) is incompletely understood. We generated spatial transcriptomic maps of 56 human precursor lesions and LUADs from 25 patients and of an independent cohort of 36 lesions from 19 patients, analyzing a total of 486,519 spots and 5.4 million cells. We identify region-specific programs that distinguish precursors from LUADs. Spatially resolved clonal architectures reveal patient-specific heterogeneity in evolution of precursors to LUADs. Rapid and comprehensive analysis of complex proteomes across large sample sets is vital for unlocking the potential of systems biology. We present a high-throughput mass spectrometry (MS) proteomics method that integrates narrow-window data-independent acquisition (nDIA) with short-gradient micro-flow chromatography, enabling profiling of >240 samples per day. This optimized MS approach identifies 6,201 and 7,466 human proteins with 1- and 2-min gradients, respectively. As a practical application, we analyzed 507 samples composed of 13 different tissues from mice treated with the enzyme-drug L-asparaginase (ASNase) or its glutaminase-free Q59L mutant, generating a quantitative profile of 11,472 proteins following drug treatment.
