Despite the availability of the coronavirus disease 2019 (COVID-19) vaccination, uptake among pregnant women in Sub-Saharan Africa has been low. This scoping review aimed to identify and characterize determinants influencing COVID-19 vaccination decision-making behaviors among pregnant women in Sub-Saharan Africa. We searched five online databases for articles on COVID-19 vaccination among pregnant women in Sub-Saharan Africa. We identified studies published in English between March 2020 and April 2023 that assessed vaccine-specific issues, psychosocial constructs, and contextual factors associated with COVID-19 vaccination decision-making behaviors. Of the fourteen studies identified, over half (57.1%) were cross-sectional; three used qualitative research methods; and three …

Zea E-Books Collection
Microbe Hunters by Paul de Kruif was first published in 1926 by Harcourt, Brace and Company, New York. It dramatically recounts the breakthrough discoveries of the fundamental elements of bacteriology. It features exciting profiles of Antony Leeuwenhoek, Lazzaro Spallanzani, Louis Pasteur, Robert Koch, Émile Roux, Emil Behring, Élie Metchnikoff, Theobald Smith, David Bruce, Ronald Ross, Battista Grassi, Walter Reed, and Paul Ehrlich. Their development of germ theory and its scientific proofs led to the first effective treatments for human diseases like anthrax, rabies, diptheria, malaria, sleeping sickness, syphilis, and yellow fever. They also made discoveries that saved the dairy, wine, …

BACKGROUND: University students are an at-risk group for the development of eating disorders (EDs); however, many college campuses lack sufficient resources to provide ED specialty care. Students report unique reasons for not seeking ED treatment, including the desire to solve the problem on their own (eg, seeking help from friends, self-medicating, or waiting to see if their problems improve), inability to afford treatment, lack of time to participate in the treatment, fear of seeing their primary care physician, and lack of recognition of their issues as an ED. Mobile health (mHealth) apps may be a cost-effective, helpful adjunctive tool to …

Long noncoding RNAs (lncRNAs) are involved in glioma initiation and progression. Glioma stem cells (GSCs) are essential for tumor initiation, maintenance, and therapeutic resistance. However, the biological functions and underlying mechanisms of lncRNAs in GSCs remain poorly understood. Here, we identified that LINC00839 was overexpressed in GSCs. A high level of LINC00839 was associated with GBM progression and radiation resistance. METTL3-mediated m6A modification on LINC00839 enhanced its expression in a YTHDF2-dependent manner. Mechanistically, LINC00839 functioned as a scaffold promoting c-Src-mediated phosphorylation of β-catenin, thereby inducing Wnt/β-catenin activation. Combinational use of celecoxib, an inhibitor of Wnt/β-catenin signaling, greatly sensitized GSCs to …

Neurons in the suprachiasmatic nucleus (SCN) generate circadian changes in the rates of spontaneous action potential firing that regulate and synchronize daily rhythms in physiology and behavior. Considerable evidence suggests that daily rhythms in the repetitive firing rates (higher during the day than at night) of SCN neurons are mediated by changes in subthreshold potassium (K

Evaluating Vaccine Management Strategies For Edwardsiella Ictaluri Infections In Zebrafish (Danio Rerio), Brandy Malbrough
LSU Master's Theses
Aquaculture is a globally important industry that faces significant challenges due to infectious diseases, which can result in substantial financial losses. In 2011, a new strain of Edwardsiella ictaluri emerged as a major pathogen affecting zebrafish in the ornamental fish industry, leading to high mortality rates and posing a severe threat to the sector. Vaccines provide potential benefits for disease prevention in aquaculture, particularly live-attenuated vaccines (LAVs), which induce strong host immune responses. Bacterial attenuation for vaccine development has evolved from serial passage to more targeted genetic modification techniques. However, regulatory constraints limit the availability of licensed LAVs for aquaculture. …

Pharmacy Faculty Articles and Research
Proteoglycan 4 (PRG4) is a mucinous glycoprotein secreted by synovial fibroblasts and superficial zone chondrocytes, released into synovial fluid, and adsorbed on cartilage and synovial surfaces. PRG4′s roles include cartilage boundary lubrication, synovial homeostasis, immunomodulation, and suppression of inflammation. Gouty arthritis is mediated by monosodium urate (MSU) crystal phagocytosis by synovial macrophages, with NLRP3 inflammasome activation and IL-1β release. The phagocytic receptor CD44 mediates MSU crystal uptake by macrophages. By binding CD44, PRG4 limits MSU crystal uptake and downstream inflammation. PRG4/CD44 signaling is transduced by protein phosphatase 2A, which inhibits NF-κB, decreases xanthine oxidoreductase (XOR), urate production, and ROS-mediated IL-1β …

HPD Articles
Drug reaction with eosinophilia and systemic symptoms (DRESS) is a severe drug reaction that is triggered several weeks after the start of a new medication. This syndrome presents with a variety of clinical symptoms, specifically a manifestation involving a fever followed by a severe rash. A variety of medications are known to trigger DRESS, with the most common being anticonvulsants and allopurinol. Here, we discuss the case of a medication, apalutamide, that caused DRESS in our patient. Early recognition and abrupt discontinuation of the medication is required for the management of this syndrome and to minimize morbidity and mortality.

Cerebral Vasomotor Reactivity To Assess Brain Dysregulation In Post Covid Neurological Syndrome, Musharaf Mohiuddin
Health and Kinesiology Theses
Background: COVID-19 has wide-ranging physiological effects, with many patients complaining of persistent asthenia following recovery from the acute phase of the infection. The frequent term for this is Long Haul COVID (LHC). While we have tools to measure effects on general physiology in human subjects, a metric for cerebral dysregulation is lacking. Cerebral blood flow (CBF) is closely regulated in the healthy young person. Dysregulation has been well described in many conditions, including Posterior Reversible Encephalopathy Syndrome (PRES), and is associated with neurological deficits. Cerebral Vasomotor Reactivity was used as a tool to assess this dysregulation.
Methods: Transcranial Doppler (TCD) …

The COVID-19 pandemic disrupted hospital operations. Anecdotal evidence suggests financial performance likewise suffered, yet little empirical research supports this claim. This study aimed to explore the impact of the pandemic on the financial performance of the most prominent academic hospitals in the United States. Data from the 115 largest major teaching hospitals in the United States were extracted from the American Hospital Directory for three years (2019-2021). We hypothesized that the year and region would moderate the relationship between a hospital's return on assets (financial performance) and specific operational variables. We found evidence through descriptive statistics and multivariate moderated regressions …
Tissue Resident Regulatory T Cells And Immunopathology During Influenza Infections, Michael Mcgee
Tissue Resident Regulatory T Cells And Immunopathology During Influenza Infections, Michael Mcgee
LSU Doctoral Dissertations
Influenza (flu) virus infections are a leading cause of respiratory illness. Globally there are approximately 290,000-650,000 deaths and 3-5 million cases of severe illness during the average flu season. Severe flu infection is associated with the combination of a strong pro-inflammatory and weak anti-inflammatory response. Regulatory T cell subsets suppress pro-inflammatory responses during flu infection, with production of the immunomodulatory cytokine IL-10 playing a critical role in this process. Tissue resident regulatory T cell subsets are maintained within nonlymphoid tissues and are optimally positioned to suppress localized pro-inflammatory responses. However, knowledge of the signaling pathways regulating the development of tissue …

Predicted Structure And Functional Roles Of Alphaherpesvirus Tegument Protein Ul37 In Virion Morphogenesis And Innate Immunity, Therese Marie Collantes
LSU Doctoral Dissertations
Several members of the Family Herpesviridae are clinically significant pathogens, affecting humans and a wide range of animals. Alphaherpesviruses infect epithelial cells at the primary site of infection and subsequently invade adjacent sensory neurons. Infectious bovine rhinotracheitis (IBR) is an economically important disease caused by bovine herpesvirus 1 (BoHV-1), which belongs to the subfamily Alphaherpesvirinae, genus Varicellovirus. To date, vaccination is the only preventive measure against IBR. Understanding determinants of herpesvirus replication and innate immune regulation is important to help further develop vaccines that are efficacious and incapable of establishing latency in the host nervous system. The alphaherpesvirus …

Older Adults With A History Of Psychiatric Conditions Experience More Cognitive Decline Than Older Adults Without This History, Maria T. Brown, Miriam Mutambudzi
Population Health Research Brief Series
Having psychiatric conditions (such as bipolar disorder, schizophrenia, and major depressive disorder) throughout one’s life can lead to faster rates of cognitive decline as one ages. This data slice explores the associations between psychiatric history and cognitive function among U.S. White, Black, and Hispanic adults ages 65 and older from 1995-2014. Findings demonstrate that having a history of psychiatric problems is related to lower cognitive functioning and a faster rate of cognitive decline for all three groups, but Black and Hispanic adults experience steeper declines compared with White adults.

Acute myeloid leukemia (AML) with retinoic acid receptor γ (RARG) rearrangement has clinical, morphologic, and immunophenotypic features similar to classic acute promyelocytic leukemia. However, AML with RARG rearrangement is insensitive to alltrans retinoic acid (ATRA) and arsenic trioxide (ATO) and carries a poor prognosis. We initiated a global cooperative study to define the clinicopathological features, genomic and transcriptomic landscape, and outcomes of AML with RARG rearrangements collected from 29 study groups/institutions worldwide. Thirty-four patients with AML with RARG rearrangements were identified. Bleeding or ecchymosis was present in 18 (54.5%) patients. Morphology diagnosed as M3 and M3v accounted for 73.5% and …

An epidemic of obesity has affected large portions of the world, increasing the risk of developing many different age-associated diseases, including cancer, cardiovascular disease, and diabetes. In contrast with the prevailing notion that "a calorie is just a calorie," there are clear differences, within and between individuals, in the metabolic response to different macronutrient sources. Recent findings challenge this oversimplification; calories from different macronutrient sources or consumed at different times of day have metabolic effects beyond their value as fuel. Here, we summarize discussions conducted at a recent NIH workshop that brought together experts in calorie restriction, macronutrient composition, and …

Epithelial-to-mesenchymal transition (EMT) underlies immunosuppression, drug resistance, and metastasis in epithelial malignancies. However, the way in which EMT orchestrates disparate biological processes remains unclear. Here, we identify an EMT-activated vesicular trafficking network that coordinates promigratory focal adhesion dynamics with an immunosuppressive secretory program in lung adenocarcinoma (LUAD). The EMT-activating transcription factor ZEB1 drives exocytotic vesicular trafficking by relieving Rab6A, Rab8A, and guanine nucleotide exchange factors from miR-148a-dependent silencing, thereby facilitating MMP14-dependent focal adhesion turnover in LUAD cells and autotaxin-mediated CD8

Acute myeloid leukemia (AML) can be treated with either high- or low-intensity regimens. Highly sensitive assays for measurable residual disease (MRD) now allow for a more precise assessment of response quality. We hypothesized that treatment (Rx) intensity may not be a key predictor of outcomes, assuming that an optimal response to therapy is achieved. We performed a single-center retrospective study including 635 patients with newly diagnosed AML responding to either intensive cytarabine/anthracycline-based chemotherapy (IA; n = 385) or low-intensity venetoclax-based regimens (LOW + VEN; n = 250) and who had adequate flow cytometry-based MRD testing performed at the time of …

Outcomes for children and adolescents with relapsed and refractory Hodgkin lymphoma (HL) are poor, with ∼50% of patients experiencing a subsequent relapse. The anti-CD30 antibody-drug conjugate brentuximab vedotin improved progression-free survival (PFS) when used as consolidation after autologous stem cell transplantation (ASCT) in adults with high-risk relapsed/refractory HL. Data on brentuximab vedotin as consolidative therapy after ASCT in pediatric patients with HL are extremely limited, with data of only 11 patients reported in the literature. We performed a retrospective analysis of 67 pediatric patients who received brentuximab vedotin as consolidation therapy after ASCT for the treatment of relapsed/refractory HL to …

Single-cell nanopore sequencing of full-length mRNAs transforms single-cell multi-omics studies. However, challenges include high sequencing errors and dependence on short-reads and/or barcode whitelists. To address these, we develop scNanoGPS to calculate same-cell genotypes (mutations) and phenotypes (gene/isoform expressions) without short-read nor whitelist guidance. We apply scNanoGPS onto 23,587 long-read transcriptomes from 4 tumors and 2 cell-lines. Standalone, scNanoGPS deconvolutes error-prone long-reads into single-cells and single-molecules, and simultaneously accesses both phenotypes and genotypes of individual cells. Our analyses reveal that tumor and stroma/immune cells express distinct combination of isoforms (DCIs). In a kidney tumor, we identify 924 DCI genes involved in …

Background: Nearly 80% of patients with pancreatic cancer suffer from glucose intolerance or diabetes. Pancreatic cancer complicated by diabetes has a more immunosuppressive tumor microenvironment (TME) and is associated with a worse prognosis. The relationship between glucose metabolism and programmed cell death-Ligand 1 (PD-L1) is close and complex. It is important to explore the regulation of high glucose on PD-L1 expression in pancreatic cancer and its effect on infiltrating immune effectors in the tumor microenvironment. Methods: Diabetic murine models (C57BL/6) were used to reveal different immune landscape in euglycemic and hyperglycemic pancreatic tumor microenvironment. To report the activity of venetoclax in patients with relapsed mantle cell lymphoma (MCL), we identified 81 patients treated with venetoclax monotherapy (n = 50, 62%) or in combination with a Bruton tyrosine kinase inhibitor (BTKi) (n = 16, 20%), an anti-CD20 monoclonal antibody (n = 11, 14%), or other active agents at 12 US academic medical centers. Patients had high-risk disease features including Ki67 >30% (61%), blastoid/pleomorphic histology (29%), complex karyotype (34%), and TP53 alterations (49%), and received a median of 3 prior treatments including BTKis in 91%. Venetoclax alone or in combination resulted in an overall response rate …

This report identifies a novel variant form of the inherited bleeding disorder Glanzmann thrombasthenia, exhibiting only mild bleeding in a physically active individual. The platelets cannot aggregate ex vivo with physiologic agonists of activation, although microfluidic analysis with whole blood displays moderate ex vivo platelet adhesion and aggregation consistent with mild bleeding. Immunocytometry shows reduced expression of αIIbβ3 on quiescent platelets that spontaneously bind/store fibrinogen, and activation-dependent antibodies (ligand-induced binding site-319.4 and PAC-1) report β3 extension suggesting an intrinsic activation phenotype. Genetic analysis reveals a single F153Sβ3 substitution within the βI-domain from a heterozygous T556C nucleotide substitution of ITGB3 exon …
