Wnt ligands belong to a family of secreted glycoproteins in which binding to a range of receptors/co-receptors activates several intracellular pathways. WNT5A, a member of the Wnt family, is classified as a non-canonical Wnt whose activation triggers planar cell polarity (PCP) and Ca+2 downstream pathways. Aberrant expression of WNT5A has been shown to play both protective and harmful roles in an array of conditions, such as inflammatory disease and cancer. Oncology Articles

Due in part to racial disparities and underrepresentation in clinical studies, optimal therapies for Black patients with multiple myeloma remain undefined. This final analysis of GRIFFIN by race showed that the addition of daratumumab (D) to lenalidomide/bortezomib/dexamethasone (RVd) provides clinical benefit among both Black and White transplant-eligible newly diagnosed patients compared with RVd alone. However, Black patients were more likely to discontinue ≥1 drug due to treatment-emergent adverse events. In summary, these findings suggest a benefit of D-RVd front-line therapy among Black and White patients and underscore the importance of equitable treatment access for all patients.

In healthcare, teams are essential in solving today’s toughest challenges. However, narrow disciplinary perspectives have limited our current understanding of how teams function in various healthcare contexts. Researchers often rely on traditional survey methods as their primary tool, which can prevent them from gathering comprehensive data. To overcome this limitation, sharing stories and narratives visually allows participants to create representations of their perceptions of team experiences and complex encounters using a variety of approaches. Neurocutaneous syndromes comprise a diverse group of conditions affecting both the nervous system and the integumentary system. Within this spectrum, we present a rare and compelling case of cerebrum polycystica vera associated with ectodermal dysplasia in a 30-year-old male. The patient's clinical journey includes a sequence of seizures progressing to left-sided weakness, accompanied by compelling dermatological manifestations. Extensive neuroimaging studies, including MRI and CT scans, reveal the presence of multiple cysts in the brain, highlighting the distinct nature of this novel neurocutaneous syndrome. Myxomas are rare, benign but locally aggressive neoplasms that are of mesenchymal origin. These tumors are usually diagnosed only when they have grown enough to cause symptoms because of their indolent nature, hence imaging plays a large role in the diagnosis of these tumors. Imaging findings may include well-circumscribed multinodular masses with destruction of adjacent bony structures. Treatment of these tumors is complete resection and close follow up. Background and objective: Bilateral thalamic lesions present a significant diagnostic and therapeutic challenge due to their critical role in sensory, motor, and cognitive functions. The objective of this study was to study the causes and presentations of bilateral thalamic lesions among patients presenting to a tertiary care hospital. Methods: This retrospective study reviewed 40 patients aged 18–80 years admitted to Aga Khan University Hospital between 2006 and 2023 with MRI-confirmed bilateral thalamic lesions. Immigration from the Middle East and North Africa (MENA) has diversified the U.S. non-Hispanic White population. Analyzing health disparities within this group is a complex task due to data limitations across most federal and state data collection systems. This study investigates disparities in the risk of giving birth to a low-birth-weight infant among foreign-born non-Hispanic White MENA and non-MENA mothers and by MENA mothers’ nationality. Since 2011, Oxford County Wellness Collaborative (OCWC) has been transforming personal, environmental, and economic health by building strong relationships among people and organizations. OCWC contracted with the Data Innovation Project at the Catherine Cutler Institute of Health and Social Policy to synthesize previously collected or reported data and collect primary data to conduct a retrospective evaluation. The evaluation team used this data to convey past successes, better understand the outcomes of the collaborative to date, and provide recommendations for OCWC’s next steps in furthering its impact.

Swallowing disorders are prevalent in tracheostomized patients and are associated with a high incidence of aspiration. This narrative review discusses the primary causes and subsequent management strategies for dysphagia in this population. Factors such as reduced laryngeal elevation, compromised hypopharyngeal and laryngeal sensations, impaired vocal cord function, loss of glottic airflow and subglottic pressure, weakened cough reflex, increased respiratory secretions, and disuse muscle atrophy have been identified as major contributors. These challenges can lead to severe complications, including aspiration pneumonia and respiratory failure. An injury to the brachial plexus can lead to the development of a pseudomeningocele, a buildup of cerebrospinal fluid (CSF) in the subarachnoid space caused by tears in the affected root sleeves. External forces acting on the brachial plexus represent the main mechanism of injury. The resulting stretching and tearing of the meninges outside the nerve root led to the pseudomeningocele. Although often initially asymptomatic, a pseudomeningocele can eventually cause pain, paresthesia, muscle weakness, and paralysis. The nerve conduction studies (NCS), electromyography (EMG), and magnetic resonance imaging (MRI) can be used to identify the injury, and treatment can involve either …

Genetic prediction holds immense promise for translating genetic discoveries into medical advances. As the high-dimensional covariance matrix (or the linkage disequilibrium (LD) pattern) of genetic variants often presents a block-diagonal structure, numerous methods account for the dependence among variants in predetermined local LD blocks. Moreover, due to privacy considerations and data protection concerns, genetic variant dependence in each LD block is typically estimated from external reference panels rather than the original training data set. This paper presents a unified analysis of blockwise and reference panel-based estimators in a high-dimensional prediction framework without sparsity restrictions.
