Background: Prior studies evaluating the relationship between psychopharmacotherapy (PPT), and postpartum hemorrhage (PPH) have yielded inconsistent findings. Clarifying this potential relationship is important for effective counseling and risk stratification.
Objectives: Our primary objective was to evaluate the association between prenatal exposure to PPT (any drug class) and the occurrence of PPH requiring transfusion of packed red blood cells (PPH+pRBC) after systematically adjusting for known hemorrhage risk factors at the time of admission for delivery. Secondary objectives were to evaluate the association between individual PPT drug classes and PPH+pRBC, and the association between treatment intensity of mental health condition and PPH+pRBC. …

Introduction
As our population continues to age and the number of cancer survivors increases, the physical, emotional, and financial burden on family caregivers will increase exponentially. While burden among cancer caregivers is recognized to impact both caregiver and patient health and quality of life, our current comprehensive geriatric assessments (CGAs) do not routinely include evaluations of caregivers’ distress, burden, or functioning. As we continue to work to better understand the needs of older adults with cancer, it is critical that this circle of concern includes the needs of their caregivers.

Simple Summary
Pancreatic cancer (PanCa) is one of the deadliest forms of cancer with limited therapeutic options. The available conventional therapies are highly toxic and often show resistance. Accumulating evidence suggests that dysregulated ribosome biogenesis has been linked to the survival and aggressive phenotypes of many tumor types, including PanCa. Thus, targeting ribosome biogenesis could be a novel approach for suppressing the growth of PanCa. The current study aimed to investigate the anti-cancer efficacy and underlying molecular mechanisms of honey against PanCa. Our results demonstrated that honey induces apoptosis and inhibits the growth and invasive potential of PanCa cells by …

Characterizing The Influence Of Metabolism On Steady State Water Exchange In Yeast As A Potential Mri Diagnostic Biomarker, Samantha Mumford
Dissertations and Theses
Under conditions of ionic equilibrium with the environment a cell maintains a constant volume yet rapidly exchanges water molecules with the extracellular space. The rate constant for steady state water efflux, kio, can be measured using contrast enhanced magnetic resonance (CE-MR). The active water cycling (AWC) hypothesis is puts forward that kio is primarily influenced by cellular ATP levels and H+ ATPase activity at the cell membrane. Due to the correlation between kio and ATP levels, kio is thought to be a probable MRI biomarker for metabolic diseases such as cancer. Yet, the molecular …

We introduce a novel approach for predicting running performance, designed to apply across a wide range of race distances (from marathons to ultras), elevation gains, and runner types (front-pack to back of the pack). To achieve this, the entire running logs of 15 runners, encompassing a total of 15,686 runs, were analyzed using two approaches: (1) regression and (2) time series regression (TSR). First, the prediction accuracy of a long short-term memory (LSTM) network was compared using both approaches. The regression approach demonstrated superior performance, achieving an accuracy of 89.13% in contrast, the TSR approach reached an accuracy of 85.21%. …

Summary: The surgical delay technique can be used effectively in autologous breast reconstruction when there is unfavorable flap vascular anatomy or when the reconstruction necessitates a larger volume of donor tissue to obtain optimal results. The length of time between surgically delaying the flap to pedicle division and inset of the flap often varies based on surgeon preference but is typically approximately a week or longer. The authors present a case in which a 24-hour surgical delay was successfully used to augment deep inferior epigastric perforator flaps for autologous reconstruction. This technique is beneficial as it does not allow time …

Whole genome sequences (WGS) enable discovery of rare variants which may contribute to missing heritability of coronary artery disease (CAD). To measure their contribution, we apply the GREML-LDMS-I approach to WGS of 4949 cases and 17,494 controls of European ancestry from the NHLBI TOPMed program. We estimate CAD heritability at 34.3% assuming a prevalence of 8.2%. Ultra-rare (minor allele frequency ≤ 0.1%) variants with low linkage disequilibrium (LD) score contribute ~50% of the heritability. We also investigate CAD heritability enrichment using a diverse set of functional annotations: i) constraint; ii) predicted protein-altering impact; iii) cis-regulatory elements from a cell-specific chromatin …

The presence of somatic mutations, including copy number variants (CNVs), in the brain is well recognized. Comprehensive study requires single-cell whole genome amplification, with several methods available, prior to sequencing. Here we compare PicoPLEX with two recent adaptations of multiple displacement amplification (MDA): primary template-directed amplification (PTA) and droplet MDA, across 93 human brain cortical nuclei. We demonstrate different properties for each, with PTA providing the broadest amplification, PicoPLEX the most even, and distinct chimeric profiles. Furthermore, we perform CNV calling on two brains with multiple system atrophy and one control brain using different reference genomes. We find that 20.6% …

Recognition of viral infection often relies on the detection of double-stranded RNA (dsRNA), a process that is conserved in many different organisms. In mammals, proteins such as MDA5, RIG-I, OAS, and PKR detect viral dsRNA, but struggle to differentiate between viral and endogenous dsRNA. This study investigates an shRNA targeting DDX54's potential to activate PKR, a key player in the immune response to dsRNA. Knockdown of DDX54 by a specific shRNA induced robust PKR activation in human cells, even when DDX54 is overexpressed, suggesting an off-target mechanism. Activation of PKR by the shRNA was enhanced by knockdown of ADAR1, a …

Alzheimer's disease (AD) and Alzheimer's disease-related dementias (ADRDs) are broad-impact multifactorial neurodegenerative diseases. Their complexity presents unique challenges for developing effective therapies. This review highlights research presented at the 2024 Society for Neuroscience meeting which emphasized the gut microbiome's role in AD pathogenesis by influencing brain function and neurodegeneration through the microbiota-gut-brain axis. This emerging evidence underscores the potential for targeting the gut microbiota to treat AD/ADRD.

Cow behavior is a crucial indicator for monitoring health, reproductive status, and welfare in livestock management. However, methods that rely on wearable devices often face significant challenges, including high costs, maintenance difficulties, and potential impacts on animal welfare. To address these limitations, this study explored the potential of using YOLOv8, a cutting-edge computer vision model, for non-invasive monitoring of cow behavior. The research methodology involved four key steps: data collection, preliminary data processing, model training, and validation. The findings revealed that YOLOv8 is capable of accurately detecting and localizing key cow behaviors—lying, standing, eating, and ruminating—achieving a mean average precision …

Aberrant preterm infant gut microbiota assembly predisposes to early-life disorders and persistent health problems. Here, we characterize gut microbiome dynamics over the first 3 months of life in 236 preterm infants hospitalized in three neonatal intensive care units using shotgun metagenomics of 2,512 stools and metatranscriptomics of 1,381 stools. Strain tracking, taxonomic and functional profiling, and comprehensive clinical metadata identify Enterobacteriaceae, enterococci, and staphylococci as primarily exploiting available niches to populate the gut microbiome. Clostridioides difficile lineages persist between individuals in single centers, and Staphylococcus epidermidis lineages persist within and, unexpectedly, between centers. Collectively, antibiotic and non-antibiotic medications influence gut …

BACKGROUND: Smoking is the leading behavioural risk factor for mortality globally, accounting for more than 175 million deaths and nearly 4·30 billion years of life lost (YLLs) from 1990 to 2021. The pace of decline in smoking prevalence has slowed in recent years for many countries, and although strategies have recently been proposed to achieve tobacco-free generations, none have been implemented to date. Assessing what could happen if current trends in smoking prevalence persist, and what could happen if additional smoking prevalence reductions occur, is important for communicating the effect of potential smoking policies.
METHODS: In this analysis, we use …

Crafting Lornoxicam-Infused Microsponges For Elevated Efficacy In Inflammatory Disorders, Vishal Yadav, Pooja Kachare
Bulletin of Faculty of Pharmacy Cairo University
Introduction: Lornoxicam (LRX), a member of the Oxicam class of non-steroidal anti-inflammatory drugs (NSAIDs), exhibits potent analgesic, anti-inflammatory, and antipyretic properties. Nevertheless, its effectiveness is hampered by high dosage requirements, a short half-life, and low and pH dependent water solubility, necessitating further research for enhanced therapeutic outcomes.
Methodology: This study introduces floating microsponges of LRX to enhance drug release duration and reduce dosing frequency. Microsponges were formulated using quasi-emulsion solvent diffusion with ethyl cellulose and polyvinyl alcohol (PVA) as sustained-release polymers. A 32 factorial design assessed the effects of ethyl cellulose (X1) and PVA (X2) …

Mycobacterium tuberculosis (MTB) is a bacterium known to target, infect, and destroy lung cells as well as connective tissues within the body. This bacterium is prevalent worldwide and has infected over a quarter of the current world population, becoming one of the most successful pathogens in history. Due to its extreme transmission rates as an airborne pathogen, MTB strains have been treated with antibiotics such as rifampicin and isoniazid, which inhibit bacterial infection in the human body. These first-round drugs remained as successful mechanisms for slowing and killing the pathogen, notably through rifampicin's inhibition of RNA-polymerase and isoniazid’s ability to …
