Heterocyclic compounds play an important role in pharmaceutical drug development. Several natural products and biologically active compounds contain heterocyclic motifs in them. Multicomponent coupling reactions offer an excellent platform for the synthesis of diverse libraries of heterocyclic compounds. We have been working on the synthesis of novel heterocyclic small molecules utilizing reactions such as Baylis-Hillman reaction, Passerini reaction, Click reaction, reductive amination aldol condensation, etc.

Mindfulness, as a state, trait, and training, is linked with myriad positive mental and physical health outcomes. Understanding the individual characteristics potentially influencing links between mindful traits, mindfulness training, and physical health, is therefore important, yet remains under-addressed. Utilizing data from the ongoing Serenity Study (NCT02371317), the current project examines if (1) at baseline, higher trait mindfulness relates to lower BP consistently as a function of demographics, (2) Mindfulness-Based Stress Reduction (MBSR) training lowers BP consistently across demographic subgroups and initial levels of trait mindfulness, and (3) if change in trait mindfulness following MBSR training correlates with change in BP …

BACKGROUND: Hypophosphatasia (HPP) is a rare, inherited, metabolic bone disease caused by deficient tissue-non-specific isoenzyme of alkaline phosphatase activity that manifests as a broad range of signs/symptoms, including bone mineralization defects and systemic complications. The burden of disease is poorly characterized, particularly in children. This study aimed to characterize the patient-reported burden of disease among children with HPP using two survey instruments: the HPP Impact Patient Survey (HIPS) and the HPP Outcomes Study Telephone interview (HOST).

Fragile X syndrome (FXS) is characterized by hallmark features of gaze avoidance, reduced social approach, and social anxiety. The development of therapeutics to manage these symptoms has been hindered, in part, by the lack of sensitive outcome measures. This study investigated the utility of a novel eye tracking paradigm for indexing social avoidance-related phenotypes. Adolescent/young adult-aged males with FXS (n=24) and typical development (n=23) participated in the study. Objective: People with serious mental illness (SMI) experience significant disparities in morbidity and mortality from preventable and treatable medical conditions. Women with SMI have low mammography screening rates. SMI, poverty, and poor access to care can have a significant effect on a woman’s opportunity to learn about and discuss breast cancer screening with health care providers. This study examines the feasibility pilot outcomes of mammography decision support and patient navigation intervention (DSNI) for women with SMI living in supportive housing settings. Neuropilin-1 (NRP1) is an essential transmembrane receptor with a variety of cellular functions. Here, we identify two human NRP1 splice variants resulting from the skipping of exon 4 and 5, respectively, in colorectal cancer (CRC). Both NRP1 variants exhibit increased endocytosis/recycling activity and decreased levels of degradation, leading to accumulation on endosomes. This increased endocytic trafficking of the two NRP1 variants, upon HGF stimulation, is due to loss of N-glycosylation at the Asn150 or Asn261 site, respectively. Moreover, these NRP1 variants enhance interactions with the Met and β1-integrin receptors, resulting in Met/β1-integrin co-internalization and co-accumulation on endosomes. This provides persistent …

Background
In 2014, Vietnam was the first Southeast Asian country to commit to achieving the World Health Organization’s 90–90-90 global HIV targets (90% know their HIV status, 90% on sustained treatment, and 90% virally suppressed) by 2020. This pledge represented further confirmation of Vietnam’s efforts to respond to the HIV epidemic, one feature of which has been close collaboration with the U.S. President’s Emergency Plan for AIDS Relief (PEPFAR). Starting in 2004, PEPFAR supported community outreach programs targeting high-risk populations (people who inject drugs, men who have sex with men, and sex workers). The "ED Stories" project is a novel method for promoting individual wellness and interpersonal relationships on a virtual platform in an emergency medicine residency program. We engaged multiple individuals across all levels of medical training on a virtual platform to facilitate discussion regarding difficult situations, patient interactions, and tips on how to succeed in challenge circumstances in the future. The platform was outlined as a "safe space," free of judgment and criticism. We engaged 54 participants with a culmination of 131 interactions; there were 14 original posts with a total of 22 comments. We believe that this virtual platform allowed …

Pediatric brain cancer patients are at a high risk for radiation-induced cognitive impairment due to white matter changes in the brain. Half of six-month radiotherapy survivors develop significant changes in white matter. Previous research has shown that a mouse model can be used to show similar cognitive and behavioral deficits in human patients. The purpose of this work is to evaluate the effectiveness of two drug therapies, Donepezil and 3,3-Diindolylmethane (DIM), that could be used to either protect the brain from radiation injury or cure the cognitive injury and behavioral deficits that result from whole-brain irradiation. Atrazine and glyphosate are the two most common agricultural herbicides used in the United States. Both herbicides can move in the environment, which results in contamination of drinking water sources. Graphene nanoplatelets (GNPs) are an emerging nanoparticle with potential uses for the remediation of environmental contamination. The first aim of this study was to determine binding interactions between atrazine and GNPs to mimic a mixture composition. To determine binding interactions, GNPs with different functional groups (none, carboxylated, or aminylated) were evaluated. Surfing is a popular sport, but little is known about the extent to which recreational surfers experience fluid loss from this activity. The principal objective of this research was to estimate fluid loss during a surfing session through changes in pre- to post-session urine color (Ucol), urine osmolality (Uosm), and body mass (BM). According to the World Health Organization (WHO), every child should learn to swim as a recognised life skill. Which swimming skill or stroke to learn first is not the most important question. What is important is to teach children to learn tasks or techniques which allow them to they feel safe. Emphasizing foundational principles of motor development and focusing on safety in and around the water are paramount. Beginning in 2016 school swimming lessons in Flanders (Belgium) were given a new focus in all educational institutions. This club began out of a desire to educate high school students about the opportunities and careers available in the science, technology, engineering, and mathematics fields. Often, students feel that the prospect of entering a STEM field is daunting and out of their grasp. Recently, a commercially available starting ‘ledge’ designed to reduce foot slippage during the execution of the backstroke start was introduced in competitive swimming. For the purpose of identifying potential safety consequences, the present study investigated the effect of ledge use on head depths, speeds, and distances in backstroke starts of athletes with no prior or only novice familiarity of the ledge. Competitive backstroke starts were performed with and without ledges by high school-aged (14.5 to 19.2 yr, N = 61) swimmers in 1.52 m of water during a closed testing session. Gene-annotation enrichment is a common method for utilizing ontology-based annotations in gene and gene-product centric knowledgebases. Effective utilization of these annotations requires inferring semantic linkages by tracing paths through edges in the ontological graph, referred to as relations. However, some relations are semantically problematic with respect to scope, necessitating their omission or modification lest erroneous term mappings occur. To address these issues, we created the Gene Ontology Categorization Suite, or GOcats—a novel tool that organizes the Gene Ontology into subgraphs representing user-defined concepts, while ensuring that all appropriate relations are congruent with respect to scoping semantics. Bacterial invasion of synovial joints, as in infectious or septic arthritis, can be difficult to treat in both veterinary and human clinical practice. Biofilms, in the form of free-floating clumps or aggregates, are involved with the pathogenesis of infectious arthritis and periprosthetic joint infection (PJI). Infection of a joint containing an orthopedic implant can additionally complicate these infections due to the presence of adherent biofilms. Because of these biofilm phenotypes, bacteria within these infected joints show increased antimicrobial tolerance even at high antibiotic concentrations. The advent of Tissue-on-a-Chip (TiOC) devices has provided a novel way for researchers to approach biological study and drug development. As a platform that enables human in vivo conditions to be accurately replicated ex vivo, TiOC can accelerate both fundamental biological research to answer basic questions regarding tissue behavior and function (e.g., cell-cell interactions in the tumor microenvironment (TME)) and translational research that includes testing of standard and novel therapeutics. The reported work focuses on development of processes and technologies common to two classes of TiOC. Field immobilization of native or invasive wild pigs (Sus scrofa) is challenging. Drug combinations commonly used often result in unsatisfactory immobilization, poor recovery, and adverse side effects, leading to unsafe handling conditions for both animals and humans. We compared four chemical immobilization combinations, medetomidine–midazolam–butorphanol (MMB), butorphanol–azaperone–medetomidine (BAM™), nalbuphine–medetomidine–azaperone (NalMed-A), and tiletamine– zolazepam–xylazine (TZX), to determine which drug combinations might provide better chemical immobilization of wild pigs. We achieved adequate immobilization with no post-recovery morbidity withMMB. Adequate immobilization was achieved with BAM™; however, we observed post-recovery morbidity. A large number of Epstein Barr virus (EBV) immortalized lymphoblastoid cell lines (LCLs) have been generated and maintained in genetic/epidemiological studies as a perpetual source of DNA and as a surrogate in vitro cell model. Recent successes in reprograming LCLs into induced pluripotent stem cells (iPSCs) has paved the way to generate more relevant in vitro disease models using this existing bioresource. However, the latent EBV infection in the LCLs make them a unique cell type by altering expression of many cellular genes and miRNAs.
