Mycobacterium tuberculosis and M. smegmatis form drug-tolerant biofilms through dedicated genetic programs. In support of a stepwise process regulating biofilm production in mycobacteria, it was shown elsewhere that lsr2 participates in intercellular aggregation, while groEL1 was required for biofilm maturation in M. smegmatis. Here, by means of RNA-Seq, we monitored the early steps of biofilm production in M. bovis BCG, to distinguish intercellular aggregation from attachment to a surface. Genes encoding for the transcriptional regulators dosR and BCG0114 (Rv0081) were significantly regulated and responded differently to intercellular aggregation and surface attachment. Diets are commonly prescribed to induce weight loss and reduce the risk of many diseases. Because a standard daily calorie restriction diet is often hard to maintain, other diet alternatives such as fasting diets were created. Fasting diets have been found to be effective, though they may not be any more effective than typical diets. Before beginning a fasting diet, it is best to seek professional medical advice to assess individual risks and circumstances. This fact sheet helps readers learn more about the safety, health, and effectiveness of fasting diets.

From ancient times, Dǎngshēn (Codonopsis javanica) has been used in Chinese traditional medicine. In this study we investigated the anti-hyperinsulinemia and antioxidant properties of C. javanica root extracts in a rat model of insulin resistance (IR), induced by chronic fructose feeding. Twenty-four Sprague–Dawley rats were randomized into control, fructose-treated (10%, w/v), and fructose then C. javanica (Fru + Cod)-treated groups. After 8 weeks fructose feeding, increased fasting serum insulin levels (2.6 ± 0.45 μg/L) and insulin area under the curve confirmed the IR (p < 0.001). However, C. javanica treatment to fructose-fed rats significantly attenuated the hyperinsulinemia with correspondingly improved glucose tolerance. Weight gain in Fru + Cod group was comparably (p < 0.01) lower than in the fructose-fed group. Furthermore, IR-induced increased hepatic lipid peroxidation, as demonstrated by elevated malondialdehyde levels, were significantly (p < 0.001) alleviated by C. javanica treatment. These findings reveal that chronic fructose intake may facilitate IR and oxidative damage, which could be eradicated by improved antioxidant status. Accordingly, we found that C. javanica treatment significantly improved the antioxidant enzyme activities, including superoxide dismutase, glutathione peroxidase and glutathione reductase in the liver. These findings that fructose-induced hyperinsulinemia and associated oxidative stress could be attenuated by C. javanica root extracts. © 2013

What Kind Of An Illusion Is The Illusion Of Self, Karsten J. Struhl
Comparative Philosophy
Both early and later forms of Buddhism developed a set of arguments to demonstrate that the self is an illusion. This article begins with a brief review of some of the arguments but then proceeds to show that these arguments are not themselves sufficient to dispel the illusion. It analyzes three ways in which the illusion of self manifests itself – as wish fulfillment, as a cognitive illusion, and as a phenomenal illusion (what might be called the “I” sense). With respect to this last, the article reviews some recent developments in cognitive neuropsychology and neuroscience to discuss the way …

The cancer microenvironment is known for its complexity, both in its content as well as its dynamic nature, which is difficult to study using two-dimensional (2D) cell culture models. Several advances in tissue engineering have allowed more physiologically relevant three-dimensional (3D) in vitro cancer models, such as spheroid cultures, biopolymer scaffolds, and cancer-on-a-chip devices. Although these models serve as powerful tools for dissecting the roles of various biochemical and biophysical cues in carcinoma initiation and progression, they lack the ability to control the organization of multiple cell types in a complex dynamic 3D architecture. BACKGROUND: Single-cell RNA sequencing is a powerful tool for characterizing cellular heterogeneity in gene expression. However, high variability and a large number of zero counts present challenges for analysis and interpretation. There is substantial controversy over the origins and proper treatment of zeros and no consensus on whether zero-inflated count distributions are necessary or even useful. While some studies assume the existence of zero inflation due to technical artifacts and attempt to impute the missing information, other recent studies argue that there is no zero inflation in scRNA-seq data.

For most vertebrates, an abundance of oxygen is necessary for the production of ATP and the maintenance of cellular homeostasis. The absence of oxygen, even for brief periods, quickly leads to ATP depletion which can lead to irreparable damages to sensitive organs, such as the brain and heart. However, certain vertebrates demonstrate an extraordinary ability to thrive and recover fully from periods of no oxygen (anoxia). The reintroduction of fire to landscapes where it was once common is considered a priority to restore historical forest dynamics, including reducing tree density and decreasing levels of woody biomass on the forest floor. However, reintroducing fire causes tree mortality that can have unintended ecological outcomes related to woody biomass, with potential impacts to fuel accumulation, carbon sequestration, subsequent fire severity, and forest management. In this study, we examine the interplay between fire and carbon dynamics by asking how reintroduced fire impacts fuel accumulation, carbon sequestration, and subsequent fire severity potential. There have been multiple national calls for curricular reform in college-level science, technology, engineering, and mathematics (STEM), including a need to instill democratic skills in students. Democratic skill building can be embedded in STEM classrooms through intentional "deliberative pedagogies" which include skills in: communication, collaboration, and application. We developed and implemented a deliberative pedagogy, Deliberative Democracy (DD), across introductory majors and non-majors biology courses and a majors chemistry course. In two separate studies, I took a longitudinal, qualitative research approach to understand introductory biology and chemistry students' experiences and perceptions of DD. High variability in historical fire patterns characteristic to mixed-severity fire regimes is expected to have contributed to a structurally heterogeneous landscape throughout much of the forested ecosystems of the western United States. After more than a hundred years of fire exclusion in the region, many forests have shifted to a more homogeneous structure, which raises concern regarding these forests' ability to sustain expected increases in fire activity with a warming climate. The magneto-optical method is used to study the dynamics of the oscillatory motion of domain boundaries in a Tb3Fe5O12 garnet ferrite in the temperature range 200-295K, including the magnetic compensation point of this ferrimagnet Fs ≈ 249K. To interpret the experimental results obtained, a theoretical model of magnetic resonance of domain boundaries is proposed, according to which when the temperature approaches the compensation point, the mass of the domain boundaries tends to infinity, and the resonance frequency tends to zero.

In this article, we studied the inverse problem of finding the integral kernel for the multidimensional integro-differential heat equation. In the space of continuous functions with weight, the existence and uniqueness theorem for the stated problem is proved for an optional interval of function definition. In this paper, we study the problem of determining the core in a multidimensional heat equation for an arbitrary interval.

The solubility of carbon dioxide (CO2) in the moisture and protein components of cheese matrices and the influence of changing pH, salt and temperature levels remains unclear. In this study, model casein matrices were prepared, by renneting of micellar casein concentrate (MCC), with modulation of salt and pH levels by adding salt and glucono delta-lactone, respectively, to the MCC solutions prior to renneting. Different moisture-to-protein levels were achieved by freeze-drying, incubation of samples at different relative humidities, or by applying varying pressures during gel manufacture.
