Using Phylogenetic Analysis To Detect Market Substitution Of Atlantic Salmon For Pacific Salmon: An Introductory Biology Laboratory Experiment, Erica T. Cline, Jennifer Gogarten

We describe a laboratory exercise developed for the cell and molecular biology quarter of a year-long majors' undergraduate introductory biology sequence. In an analysis of salmon samples collected by students in their local stores and restaurants, DNA sequencing and phylogenetic analysis were used to detect market substitution of Atlantic salmon for Pacific salmon. This allowed students to apply molecular methods such as polymerase chain reaction (PCR) and DNA sequencing to a socially relevant issue.

Expressing barnacle cement proteins genes such as the BA5 gene in plants may enable individual study and analysis. This technique is effective since barnacle cement is difficult to work with as a whole in the lab setting. The BA5 gene extracted from Balanus amphitrite is transferred to tobacco leaf tissue using Agrobacterium tumefaciens.

Defeating Cytoplasmic Sequestration Of P53 In Human Breast Cancer Cells; Is Mortalin Involved?, Sarah Yunes

Cytoplasmic sequestration of p53, possibly caused by p53 interacting with mortalin, can prevent p53 from functioning in DNA repair and apoptosis, causing aberrant growth. This project treated SKBR3 breast cancer cells with MKT-077, a dye that is a competitive binder to mortalin to see if it would result in the release of p53 from the cytoplasm and restoration of p53 function. Treatment resulted in partial translocation of a protein suspected to be p53 to the nucleus and apoptosis initiated at the mitochondria.

Understanding the spatial scale of gene flow can yield valuable insight into the ecology of an organism and guide conservation strategies. Fine-scale genetic structure is uncommon in migratory passerines because of their high vagility and presumed high dispersal abilities. Aspects of the behavior and ecology of some migratory species, however, may promote structure on a finer scale in comparison to their mobility. We investigated population genetic structure in the Saltmarsh Sparrow (Ammodramus caudacutus), a migratory passerine that breeds along the northeastern coast of the United States, where it is restricted exclusively to a narrow strip of patchily distributed tidal marsh …

Optimizing Greenhouse Corn Production: How Can Two-Spotted Spider Mites Be Controlled?, Robert Eddy, Daniel T. Hahn

Purdue Methods for Corn Growth

This publication describes our experiences controlling mites while conducting experiments to optimize greenhouse corn production. Controlled studies were not conducted, but we recommend cultural practices of water spray-offs of leaf undersides and the stripping lower leaves as the crop ages. Also recommended are applications of beneficial insect Phytoseiulus persimilis at very high rates. The rates are an update of our previous publication. Photos of the pest and cultural practices are provided.

This publication describes studies conducted to determine best pot size for optimizing greenhouse corn production. We suggest a 2.2-gallon container without large drainage holes extending up the side of the pot. This represents an update of our previous recommendation of a smaller pot, after observing instability in row planting configurations. Photos are included of corn in various size pots.

The Helicobacter pylori cag pathogenicity island (cag PAI) encodes a type IV secretion system that is more commonly found in strains isolated from patients with gastroduodenal disease than from those with asymptomatic gastritis. Genome-wide organization of the transcriptional units in H. pylori strain 26695 was recently established using RNA sequence analysis (Sharma et al., 2010). Here we used quantitative reverse-transcription polymerase chain reaction of open reading frames and intergenic regions to identify putative cag PAI operons in H. pylori; these operons were analyzed further by transcript profiling after deletion of selected promoter regions. Implicit in cellular mechanisms essential for learning and memory, NMDA receptors are heterotetrameric members of the family of ionotropic glutamate receptors. Activation of the glutamate-specific receptor subunits results in the opening of ion channels, triggering an excitatory pathway, which can be moderated by certain endogenous and exogenous compounds. Previous research with the endogenous neurosteroids pregnenolone sulfate (PS) and 3α-hydroxy-5-β-20-one sulfate (PregS) have indicated that the binding of these neurosteroids to NMDA receptors containing the four A-D isoforms of the GluN2 subunit affects current flow through them1. The evolution of gene regulatory networks accounts for much of the diversity we observe in the animal kingdom. In particular, the Pax/Six/Eya/Dac gene network has been known to play critical roles in development of all bilaterians and cnidarians. However, this network has not been characterized in Porifera. These studies focus on tracing the evolutionary history of the PSED network back to the most basal organism in the metazoan phylogeny through demonstrating the presence of a direct gene regulatory network between PaxB and Six1/2 orthologs in sponges. Residential composting has increased in popularity in recent years, along with the untested claim that the addition of animal-based kitchen scraps will increase the number of scavenging wildlife visitors. This study represents the first experimental test of that claim. Using three compost piles consisting respectively of no kitchen scraps (CON), a mix of animal and vegetable scraps (MIX), or only vegetable scraps (VEG) that were monitored with heat-in-motion sensitive cameras, we studied the visitation pattern of attracted wildlife. Current synthesis of cobalt (II) complexes of sulfonated binucleating Schiff base ligands requires the reaction be run under nitrogen.1 This research introduces a novel process in which the complex can be prepared in open air. The cobalt (II) complexes are prepared with synthesized 5- sulfosalicylaldehyde, and then analyzed by IR and UV-Vis spectrophotometry.

Ailanthus altissima (Simaroubaceae) is an invasive tree from China that has spread over much of North America. A number of characteristics contribute to its success, notably tolerance of nutrient-poor soils, rapid growth rates, prolific production of wind dispersed seeds, escape from predation, and the production of allelopathic compounds. That Ailanthus altissima frequently co-occurs with legumes in nutrient poor soils alludes to a possible relationship between Ailanthus altissima and nitrogen fixing legumes. Legumes increase the biospheres supply of bio-available nitrogen through facilitating the fixation of atmospheric nitrogen within structures on roots called nodules. Understanding the visual capabilities and correlated behavior of prosimians provides vital information regarding the evolution of color vision. Feeding behavior in particular is often modified based on color vision status in various primate taxa. The present study examined feeding behavior under various light conditions in Eulemur rubriventer, a lemur species with a color vision polymorphism in which only females can have trichromatic vision. Behavioral observations were conducted for a single E. rubrventer group over a 13 day study period in April 2012. Diversity Of Tanaidacea (Crustacea: Peracarida) In The World's Oceans - How Far Have We Come?, Gary Anderson, Magdalena Blazewicz-Paszkowycz, Roger Bamber

Tanaidaceans are small peracarid crustaceans which occur in all marine habitats, over the full range of depths, and rarely into fresh waters. Yet they have no obligate dispersive phase in their life-cycle. Populations are thus inevitably isolated, and allopatric speciation and high regional diversity are inevitable; cosmopolitan distributions are considered to be unlikely or non-existent. Options for passive dispersion are discussed. Tanaidaceans appear to have first evolved in shallow waters, the region of greatest diversification of the Apseudomorpha and some tanaidomorph families, while in deeper waters the apseudomorphs have subsequently evolved two or three distinct phyletic lines.
