Multi-scale calcium (Ca2+) dynamics, exhibiting wide-ranging temporal kinetics, constitutes a ubiquitous mode of signal transduction. We report a novel endoplasmic-reticulum (ER)-targeted Ca2+ indicator, R-CatchER, which showed superior kinetics in vitro (koff≥2×103 s−1, kon≥7×106 M−1 s−1) and in multiple cell types. R-CatchER captured spatiotemporal ER Ca2+ dynamics in neurons and hotspots at dendritic branchpoints, enabled the first report of ER Ca2+ oscillations mediated by calcium sensing receptors (CaSRs), and revealed ER Ca2+-based functional cooperativity of CaSR. We elucidate the mechanism of R-CatchER and propose a principle to rationally design genetically encoded Ca2+ indicators with a single Ca2+-binding site …

BACKGROUND: Intensive care unit acquired weakness is a serious problem, contributing to respiratory failure and reductions in ambulation. Currently, there is no pharmacological therapy for this condition. Studies indicate, however, that both beta-hydroxy-beta-methylbutyrate (HMB) and eicosapentaenoic acid (EPA) increase muscle function in patients with cancer and in older adults. The purpose of this study was to determine whether HMB and/or EPA administration would increase diaphragm and quadriceps strength in mechanically ventilated patients.
METHODS: Studies were performed on 83 mechanically ventilated patients who were recruited from the Medical Intensive Care Units at the University of Kentucky. Diaphragm strength was assessed as …

The Earth–ionosphere cavity resonator is occupied primarily by the electromagnetic radiation of lightning below 100 Hz. The phenomenon is known as Schumann resonances (SR). SR intensity is an excellent indicator of lightning activity and its distribution on global scales. However, long-term measurements from high latitude SR stations revealed a pronounced in-phase solar cycle modulation of SR intensity seemingly contradicting optical observations of lightning from satellite, which do not show any significant solar cycle variation in the intensity and spatial distribution of lightning activity on the global scale. The solar cycle-modulated local deformation of the Earth–ionosphere cavity by the ionization of …

The fission of singlet excitons into triplet pairs in organic materials holds great technological promise, but the rational application of this phenomenon is hampered by a lack of understanding of its complex photophysics. Here, we use the controlled introduction of vacancies by means of spacer molecules in tetracene and pentacene thin films as a tuning parameter complementing experimental observables to identify the operating principles of different singlet fission pathways. Time-resolved spectroscopic measurements in combination with microscopic modelling enables us to demonstrate distinct scenarios, resulting from different singlet-to-triplet pair energy alignments. For pentacene, where fission is exothermic, coherent mixing between the …

Singlet fission (SF), the photophysical process converting an excited singlet exciton into two triplet excitons, is a promising approach to boost solar cell efficiencies. It is controlled by various parameters such as intermolecular interactions, energetics, entropy and vibronic coupling and a controlled modification of these parameters is key to a fundamental understanding. Blends of organic semiconductors present an interesting alternative to established methods of chemical functionalization and their potential for the study of SF pathways will be discussed using acene blends as example. Mixed thin films of SF chromophores and weakly interacting, high-bandgap spacer molecules allow one to study the …

Efficient and selective CO2 electroreduction into chemical fuels promises to alleviate environmental pollution and energy crisis, but it relies on catalysts with controllable product selectivity and reaction path. Here, by means of first-principles calculations, we identify six ferroelectric catalysts comprising transition-metal atoms anchored on In2Se3 monolayer, whose catalytic performance can be controlled by ferroelectric switching based on adjusted d-band center and occupation of supported metal atoms. The polarization dependent activation allows effective control of the limiting potential of CO2 reduction on TM@In2Se3 (TM = Ni, Pd, Rh, Nb, and Re) as well as the reaction paths and final products on …

Increasing climatic pressures such as drought and flooding challenge agricultural systems and their management globally. How agricultural soils respond to soil water extremes will influence biogeochemical cycles of carbon and nitrogen in these systems. We investigated the response of soils from long-term agricultural field sites under varying crop rotational complexity to either drought or flooding stress. Focusing on these contrasting stressors separately, we investigated soil heterotrophic respiration during single and repeated stress cycles in soils from four different sites along a precipitation gradient (Colorado, MAP 421 mm; South Dakota, MAP 580 mm; Michigan, MAP 893 mm; Maryland, MAP 1192 mm); …

Allelopathic Influence Of Eragrostis Curvula Water Extract On Seed Germination And Seedling Growth Of Two Other Species, R. W. Coelho, R. E. Schmidt
IGC Proceedings (1977-2023)
Weeping lovegrass (Eragrostis curvula Schrad. Nees) is a very agressive tropical species and does not grow well with other species. This study was set up to investigate the allelopathic effect of various extract concentrations of weeping lovegrass on seed germination of white clover (Trifolium repens L.) and bentgrass (Agrostis stolonifera L.) and the seedling development of white clover. Labotatory and growth chamber studies were conducted. Extract concentrations of 13.6, 27.2, 40.8, 54.4, and 62.0 g/100mL of water were studied. Treatments were arranged in a completely ramdomized design with three replications. The allelopathic effect of weeping lovegrass was …

Two cultivars of Guinea grass (P. maximum, Jacq.), Mombaça and Tanzânia, were cultivated in five litre pots, as single plants, in a green house. Plants were subjected to three defoliation levels (100, 200 and 400 mm height) for five successive monthly harvests. The experimental design was a 3x2 randomised block with five replicates. Three types of tiller (main, primary and secondary) were selected, at random, on each plant and leaf elongation rate (LER) above cutting height was measured, weekly, between harvests. There were significant differences (P < 0.05) in LER between tiller types in cv. Mombaça in all periods, but only in period two in cv. Tanzânia. In the last period tillers were classified, at random, as old (over four months age) mature (between two and four months) and young (between one and two months). Old tillers had a lower (P < 0.05) LER than the other two categories in both cultivars and lower (P < 0.05) SLA (specific leaf area) in Mombaça.
