Complementary Spectroscopic Assays For Investigating Protein−Ligand Binding Activity: A Project For The Advanced Chemistry Laboratory, David Mascotti, Mark Waner

A protein−ligand binding, guided-inquiry laboratory project with potential application across the advanced undergraduate curriculum is described. At the heart of the project are fluorescence and spectrophotometric assays utilizing biotin-4-fluorescein and streptavidin. The use of the same stock solutions for an assay that may be examined by two distinct spectroscopic techniques offers an opportunity to discuss not only protein−ligand binding but also to compare instrumental techniques and to discuss the underlying physical principles that are the origins of the assays. In addition to critically evaluating analytical techniques, students are pushed to develop quantitative pipetting, estimation, and experimental-design skills to collect and …

Police agencies have been accused of suffering from an acute form of technophilia. Rather than representing some dreadful disorder, this assessment reflects the strong imperative, both in police agencies and the wider community, that police must have access to the latest technologies of surveillance and crime detection.
The last decade has witnessed the proliferation of low-cost surveillance technologies, some developed specifically for law enforcement purposes. Technology once the preserve of the military or secret intelligence agencies is now within the reach of ordinary general duties police officers. In England, Wales and Northern Ireland, the Police and Criminal Evidence Act 1984 (the PACE) contained powers for the taking of fingerprints, and samples in the form of deoxyribonucleic acid (DNA). In 2001, Section 64(1A) of the PACE was substituted with Section 82 of the Criminal Justice and Police Act. The change to legislation meant that a suspect of a crime would have their fingerprints and samples permanently stored on the police national computer (PNC) even after having been acquitted. Species selection can make the difference between successful revegetation projects and costly failures. Candidate native species for revegetating burned arid lands in the southwestern United States must meet at least two criteria. These species must: (1) be competitive in postfire environments typically dominated by exotic grasses, and (2) be able to become established reliably by seeding or planting. In response to Manager’s Request Task 3 (reestablishment of native vegetation after fires on arid lands) in the 2006 JFS announcement for proposals, this research tackled the problem of selecting native species with the greatest chance of revegetation success by conducting synergistic …

Measurement Of Single- And Double-Spin Asymmetries In Deep Inelastic Pion Electroproduction With A Longitudinally Polarized Target, H. Avakian, Gerard P. Gilfoyle, Et. Al.

We report the first measurement of the transverse momentum dependence of double-spin asymmetries in semi-inclusive production of pions in deep-inelastic scattering off the longitudinally polarized proton. Data have been obtained using a polarized electron beam of 5.7 GeV with the CLAS detector at the Jefferson Lab (JLab). Modulations of single spin asymmetries over the azimuthal angle between lepton scattering and hadron production planes φ have been measured over a wide kinematic range in Bjorken x and virtual photon squared four-momentum Q2. A significant nonzero sin2φ single spin asymmetry was observed for the first time indicating strong spin-orbit …

The Lamprey River Nomination Committee sought the assistance of the Piscataqua Region Estuaries Partnership (PREP) to complete an extensive and detailed information package to nominate the Lamprey River and its major tributaries to the NH Rivers Management and Protection Program. The nomination package was due at DES on June 1, 2010, was reviewed and accepted by the State Rivers Management Committee in September and forwarded to the Commissioner of DES. The nomination is now working through the NH legislature with hearings anticipated during the spring 2011 session. If passed, the bill should be signed by Governor Lynch in July of …

Knowledge of many life history parameters are essential to properly assess and manage fish species. Although the snowy grouper, Epinephelus niveatus, is a commercially valuable fish, which is harvested throughout the Gulf of Mexico, little is known about its age, growth, and reproduction from this region. In this study, snowy grouper from the northern and eastern regions of the Gulf of Mexico were examined primarily using commercially-derived samples that were collected between 1984 and 2004. A total of 1,200 snowy grouper with fork lengths between 242 and 1,190 mm were collected. Sectioned saggital otoliths were used to determine the age …

Corrole Synthesis And Catalytic Applications Toward Cobalt(Iii)-Catalyzed Epoxidation, N-H Insertion, And Cyclopropanation, Chung Sik Kim

A variety of bromocorroles, useful precusors for Pd-based cross coupling, were sucessfully synthesized in moderate to good yields. Chiral corroles were also synthesized through use of chiral amides in the aforementioned cross coupling reactions. Cobalt complexes bearing π-acceptor (CNtBu) and σ-donor (PPh3) ligands were also prepared in excellent yields. In this dissertation, cobalt (III) corrole complexes were applied to three different reaction areas: epoxidation, N-H insertion, and cyclopropanation. Cobalt(III) corroles were found to be efficient catalysts for epoxidation reactions. The epoxidation reactions can be carried out using ethyl phenyldiazoacetate, an acceptor/donor diazo compound, as a carbenoid precusor.The reaction provided highly …

Altered DNA methylation may lead to suboptimal fetal programming, increasing the risk of adverse pregnancy outcomes such as small for gestational age (SGA); however, few studies have examined the associations between DNA methylation, prenatal exposures, and fetal outcomes. Cross-sectional data from a larger, ongoing study were used to assess the impact of prenatal smoking on gene specific methylation of umbilical cord blood derived DNA and to investigate the association between gene-specific methylation and risk of SGA. Understanding emerging viruses is critical for disease monitoring and prediction; however, surveys of novel viruses are hindered by the lack of a universal assay for viruses. Viral metagenomics, consisting of viral particle purification and shotgun sequencing, is a powerful technique for discovering viruses in a wide variety of sample types. Epithelial ovarian cancer (EOC) is a leading cause of morbidity and mortality among women in the United States, and the etiology is incompletely understood. Common, low penetrant genetic variants such as single nucleotide polymorphisms (SNPs) likely contribute to a significant proportion of EOC. We examined whether SNPs in two understudied yet biologically important types of genes, mitochondrial-related and miRNA-related genes, may contribute to EOC susceptibility using data from a large, homogeneous study population of 1,815 EOC cases and 1,900 controls (frequency-matched on age-group and race/ethnicity) genotyped through stage 1 of an ongoing genome-wide association study. This paper presents the real possibility that commercial mobile tracking and monitoring solutions will become widely adopted for the practice of non traditional covert policing within a community setting, resulting in community members engaging in covert policing of family, friends, or acquaintances. This paper investigates five stakeholder relationships using scenarios to demonstrate the potential socio-ethical implications that tracking and monitoring people will have on society at large. In the presence of missing response, reweighting the complete case subsample by the inverse of nonmissing probability is both intuitive and easy to implement. However, inverse probability weighting is not efficient in general and is not robust against misspecification of the missing probability model. Calibration was developed by survey statisticians for improving efficiency of inverse probability weighting estimators when population totals of auxiliary variables are known and when inclusion probability is known by design.
