Cochlear implant surgery is a delicate procedure performed by Otolaryngologists (ENTs) to implant an electronic device into the inner ear to provide a sense of sound for people who are profoundly deaf or hard of hearing. The current practices of training involve cadavers and 3D-printed models. Cadavers are commonly used but are expensive, single-use, and do not provide visual and haptic feedback, which are essential for medical students. 3D printed models are less commonly used and are hard to fabricate and not as realistic. Insects tend to have very high aerodynamic performance characteristics like thrust and lift generation compared to their size. Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. People with disabilities, particularly wheelchair users, face limitations in participating in outdoor activities like paddleboarding. Central Cal Adaptive, a nonprofit organization, aims to bridge this gap by offering adapted paddleboarding experiences. However, their current paddleboard presents challenges such as portability, range of motion, stability, and safety. Our team, comprising Madeline Evenson, Remy Lacchia, Katie Kellum, and Shelbi Sullaway, has collaborated with Central Cal Adaptive to refine the existing paddleboard design. The team’s goal is to enhance the paddleboard's functionality to better cater to wheelchair users' needs, ensuring lightweight, portable, and safe features for an improved outdoor experience.

Alzheimer’s disease (AD) is a progressive neurodegenerative disease with the clinical symptoms of gradual cognitive decline and memory loss. Biologically, abnormal accumulation of Aβ (Aβ) plaque and tau tangles in the brain are the core pathological hallmarks in AD. Although inflammatory response is not specific to AD, emerging evidence suggests that the disrupted innate immune system within the central nervous system, particularly microglial activation, might act as the bridge linking Aβ aggregation to pathological phosphorylation and aggregation of tau. Genetics plays a critical role in understanding the molecular mechanisms underlying neurodegenerative disorders and pathogen evolution in infectious diseases. For example, identifying genetic variants associated with a disease phenotype uncovers functional pathways that could lead to potential drug targets and therapeutic interventions. The focus of this dissertation is insect blood (Hemolymph). Hemolymph is analyzed by taking a materials science approach. Rheological characterization of hemolymph has been performed for the first time. Owing to a minute amount of available material, a new protocol was established where Magnetic Rotational Spectroscopy (MRS) with ferromagnetic nanorods was employed. The challenge was examining the microliter droplets' viscosity in less than a few minutes. This challenge was successfully resolved.

The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. The reconstruction of craniomaxillofacial bone defects remains clinically challenging. To date, autogenous grafts are considered the gold standard but present critical drawbacks. These shortcomings have driven recent research on craniomaxillofacial bone reconstruction to focus on synthetic grafts with distinct materials and fabrication techniques. Among the various fabrication methods, additive manufacturing (AM) has shown significant clinical potential. AM technologies build three-dimensional (3D) objects with personalized geometry customizable from a computer-aided design. The combination of outsole, midsole and insole layers of new shoes provide proper cushioning to the foot during the daily activities of life. Subsequently, the wear-based degeneration or damage to one or more of these sole layers can reduce the cushioning effect to the plantar surface of the foot. This dissertation aimed to (1) quantify morphological variations in the pediatric hip joint and (2) evaluate the sensitivity of an infant musculoskeletal model (MSM) to these variations, considering hip joint center estimation errors. A shape statistical model (SSM) of decedent infant femurs from the Ortolani collection was created using ShapeWorks, capturing key morphological features, such as variations in the femoral neck-shaft and anteversion angles. Seven synthetic femurs were generated from the SSM to create SSM-informed MSMs, which were systematically evaluated through kinematics and kinetics analyses in OpenSim. This dissertation explores the use of cold atmospheric pressure plasma (CAP) technology in eradicating bacterial biofilms from tissue surfaces, using Ps. fluorescens on chicken tissue to model human chronic wounds. It introduces a novel image-guided technique for precise biofilm removal, employing trypan blue staining coupled with advanced computer vision analysis, enhancing the CAP treatment's precision and efficacy. The study demonstrates the CAP device's ability to effectively disrupt biofilms through a non-thermal mechanism, based on its electrical characteristics and operational parameters. Total shoulder arthroplasty (TSA) is often considered a surgery of last resort for patients with debilitating pathologies of the glenohumeral joint such as osteoarthritis, humeral head fracture, and advancing rotator cuff tears. TSA replaces the articulation between the humeral head and the glenoid fossa, with the goals of relieving pain and restoring function. Traditional methods for assessing motor impairments are often costly and invasive, creating substantial barriers to effective rehabilitation. This thesis explores the use of DeepLabCut (DLC), a deep-learning-based pose estimation tool, to extract clinically meaningful kinematic features from video data of stroke survivors with upper-extremity (UE) impairments.

This thesis explores the micromechanical environment induced when cyclically compressing hydrogels via finite element modeling and experimentally on the impact of loading on mesenchymal stem cells (MSCs) when encapsulated withing 3D hydrogel matrices. Degenerative joint diseases, characterized by cartilage degradation, present significant challenges due to cartilage's limited self-repair capacity. Innovative approaches, including stem cell-based therapies and engineered biomaterials, have emerged as promising strategies for cartilage repair and regeneration. Bio-inspired design has been used by many engineers to solve difficult problems or to make manufacturing processes more efficient. Biomimetics is the study of implementing the structure or function of biological substances, materials, mechanisms, and processes onto artificial ones that mimic the original. The goal of the BIASD tool is to provide bio-inspiration for engineers by studying the fault-adaptive strategies that biological systems use, rather than just their structure or function.
