Mechanical loading is known to regulate bone remodeling by driving interstitial fluid flow which stimulates cells and drives nutrient transport within the trabecular network. In microgravity, the absence of mechanical stimulation or loading suppresses convective flow processes, causing diffusion-driven nutrient mixing and renewal, and accelerated bone loss. This thesis investigates how oscillation frequency and trabecular bone density jointly control nutrient mixing and wall shear stress within trabecular cavities.

This dissertation investigates the hemodynamics of pulsating blood flow in curved vessels through direct numerical simulations (DNS), with the overarching aim of uncovering fluid dynamic mechanisms linked to the onset and progression of aortic dissection in humans. The work is structured in two parts: (i) fundamental studies in idealized geometries and (ii) preliminary investigations in patient-specific anatomies. In the first part, curved pipe models representing the aortic arch are used to isolate the effects of pulsation frequency, amplitude ratio, and curvature ratio on transitional and turbulent flow dynamics. Hydrocephalus is developed when the flow of cerebrospinal fluid is obstructed in the ventricles and a pressure build-up is generated within the brain. Ventricular shunts are used to remove excess fluid from the brain, but these shunts have a common problem of failure due to the shunt being obstructed by the build-up of astrocytes. To address this, two sets of 27 designs of ventricular shunts were identified and analyzed with parameters that could potentially reduce obstruction risks. Blalock-Taussig (BT) shunts are used for defects that affect the flow of blood from the right ventricle, through the pulmonary artery, and to the lungs. Arteriovenous (AV) fistula is one type of vascular access which is a surgically created vein used to remove and return blood during hemodialysis. Plastic grafts used in the above two reconstructions may result in areas of non-physiologic flow in the grafts leading to risk of stenosis (blocked area) and thrombosis, which is the single major cause for access morbidity. This research utilizes CFD to analyze blood flow through pathways representative of central shunts, commonly used as part of the Fontan procedure to treat cyanotic heart disease. In the first part of this research, a parametric study of steady, Newtonian blood flow through parabolic pathways was performed to demonstrate the effect that flow pathway curvature has on wall shear stress distribution and flow energy losses. In the second part, blood flow through two shunts obtained via biplane angiograms is simulated. Pressure boundary conditions were obtained via catheterization. Vorticella convallaria is a genus of protozoa living in fresh water. It has a bell-shaped zooid equipped with adoral ciliary bands and a contractile stalk tethering the zooid to a substrate. Vorticella is regarded as a biological spring because its contractile stalk can pull the zooid towards the substrate at a remarkably high speed and then relaxes to its extended state much more slowly. However, reasons for Vorticella’s stalk contraction are still unknown. Pharmaceutical aerosols provide a number of advantages for treating respiratory diseases that include targeting high doses directly to the lungs and reducing exposure of other organs to the medication, which improve effectiveness and minimize side effects. Therapeutic proteins are used to successfully treat hemophilia, Crohn's Disease, diabetes, and cancer. Recent product recalls have occurred because of sub-visible particle formation resulting from the inherent instability of proteins. It has been suggested that particle formation is associated with late stage processing steps of filling, shipping, and delivery. Previous work at the University of Denver demonstrated that fluid cavitation can generate a large number of sub-visible protein particles in ultra clean formulations, but that mitigation can be achieved with fluid property manipulation.
