Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. People with disabilities, particularly wheelchair users, face limitations in participating in outdoor activities like paddleboarding. Central Cal Adaptive, a nonprofit organization, aims to bridge this gap by offering adapted paddleboarding experiences. However, their current paddleboard presents challenges such as portability, range of motion, stability, and safety. Our team, comprising Madeline Evenson, Remy Lacchia, Katie Kellum, and Shelbi Sullaway, has collaborated with Central Cal Adaptive to refine the existing paddleboard design. The team’s goal is to enhance the paddleboard's functionality to better cater to wheelchair users' needs, ensuring lightweight, portable, and safe features for an improved outdoor experience.

Alzheimer’s disease (AD) is a progressive neurodegenerative disease with the clinical symptoms of gradual cognitive decline and memory loss. Biologically, abnormal accumulation of Aβ (Aβ) plaque and tau tangles in the brain are the core pathological hallmarks in AD. Although inflammatory response is not specific to AD, emerging evidence suggests that the disrupted innate immune system within the central nervous system, particularly microglial activation, might act as the bridge linking Aβ aggregation to pathological phosphorylation and aggregation of tau. Genetics plays a critical role in understanding the molecular mechanisms underlying neurodegenerative disorders and pathogen evolution in infectious diseases. For example, identifying genetic variants associated with a disease phenotype uncovers functional pathways that could lead to potential drug targets and therapeutic interventions. In addition, tracking the genetic evolution of pathogens enables early detection and warning of infectious disease outbreaks. The focus of this dissertation is insect blood (Hemolymph). Hemolymph is analyzed by taking a materials science approach. Rheological characterization of hemolymph has been performed for the first time. Owing to a minute amount of available material, a new protocol was established where Magnetic Rotational Spectroscopy (MRS) with ferromagnetic nanorods was employed. The challenge was examining the microliter droplets' viscosity in less than a few minutes. This challenge was successfully resolved.

These systems now permeate many industries, and have enhanced traditional processes through autonomous interpretation of visual data. In the field of orthopaedics, the increasing prevalence of imaging highlights a need for similar tools. In many cases, however, direct translation of available frameworks fails to resolve the complex problems currently facing this field. The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. Electrostatic artificial muscles benefit from a microfluidic approach due to the non-linearity of electrostatic attraction creating disproportionate benefits when miniaturized. When designed …

The reconstruction of craniomaxillofacial bone defects remains clinically challenging. To date, autogenous grafts are considered the gold standard but present critical drawbacks. These shortcomings have driven recent research on craniomaxillofacial bone reconstruction to focus on synthetic grafts with distinct materials and fabrication techniques. Among the various fabrication methods, additive manufacturing (AM) has shown significant clinical potential. AM technologies build three-dimensional (3D) objects with personalized geometry customizable from a computer-aided design. These layer-by-layer 3D biomaterial structures can support bone formation by guiding cell migration/proliferation, osteogenesis, and angiogenesis. The combination of outsole, midsole and insole layers of new shoes provide proper cushioning to the foot during the daily activities of life. Subsequently, the wear-based degeneration or damage to one or more of these sole layers can reduce the cushioning effect to the plantar surface of the foot. This reduction of cushioning effect can impact the distribution of stress concentration regions along the foot plantar surfaces and lead to foot pathologies. This dissertation aimed to (1) quantify morphological variations in the pediatric hip joint and (2) evaluate the sensitivity of an infant musculoskeletal model (MSM) to these variations, considering hip joint center estimation errors. A shape statistical model (SSM) of decedent infant femurs from the Ortolani collection was created using ShapeWorks, capturing key morphological features, such as variations in the femoral neck-shaft and anteversion angles. Seven synthetic femurs were generated from the SSM to create SSM-informed MSMs, which were systematically evaluated through kinematics and kinetics analyses in OpenSim. Understanding and predicting the hydrodynamic interactions of micron-scale droplets is crucial in a wide range of industrial and real-life applications, including microfluidics, pharmaceutics, drug delivery, food science, and enhanced oil recovery. These multi-phase and multi-scale phenomena are further complicated by the presence of core droplets of an immiscible fluid within shell droplets, known as compound droplets. This dissertation explores the use of cold atmospheric pressure plasma (CAP) technology in eradicating bacterial biofilms from tissue surfaces, using Ps. fluorescens on chicken tissue to model human chronic wounds. It introduces a novel image-guided technique for precise biofilm removal, employing trypan blue staining coupled with advanced computer vision analysis, enhancing the CAP treatment's precision and efficacy. The study demonstrates the CAP device's ability to effectively disrupt biofilms through a non-thermal mechanism, based on its electrical characteristics and operational parameters. The shoulder is a complex joint with a wide range of motion, including flexion, extension, abduction, adduction, and internal and external rotation. In volleyball, repetitive overhand movements can cause shoulder overuse injuries. Total shoulder arthroplasty (TSA) is often considered a surgery of last resort for patients with debilitating pathologies of the glenohumeral joint such as osteoarthritis, humeral head fracture, and advancing rotator cuff tears. TSA replaces the articulation between the humeral head and the glenoid fossa, with the goals of relieving pain and restoring function. Two types of total shoulder implants are available: Anatomic Total Shoulder Arthroplasty (aTSA) and Reverse Total Shoulder Arthroplasty (rTSA). Despite its general success, disparities in postoperative outcomes have been observed across different racial and ethnic groups, with minority populations often experiencing less favorable results. Stroke is a leading cause of long-term disability, affecting thousands of individuals annually and significantly impairing their mobility, independence, and quality of life. Traditional methods for assessing motor impairments are often costly and invasive, creating substantial barriers to effective rehabilitation. This thesis explores the use of DeepLabCut (DLC), a deep-learning-based pose estimation tool, to extract clinically meaningful kinematic features from video data of stroke survivors with upper-extremity (UE) impairments.

Purpose: The mechanical behavior of heart tissue plays a crucial role in elucidating cardiac physiology and pathophysiology. Accurate measurement of the mechanical properties of cardiac and arterial tissues is indispensable for advancing computer simulations and developing medical devices. This study aimed to assess the passive mechanical properties of cardiac and arterial tissue in a calf model using a paired comparison method.

Advancements in robotics and bioengineering aim to emulate biological muscle systems through robotic actuators, blending mechanical strength with biological adaptability. A lesser-explored aspect is mimicking the pulse-like control characteristic of biological muscles, which contract in response to action potentials from motoneurons, with muscle contractile force relying heavily on the timing between these potentials. This study explores the effect of pulse lengths and the gaps between pulses on braided pneumatic actuators (BPAs), which mimic the nonlinearity and dynamic response of biological muscles. This thesis explores the micromechanical environment induced when cyclically compressing hydrogels via finite element modeling and experimentally on the impact of loading on mesenchymal stem cells (MSCs) when encapsulated withing 3D hydrogel matrices. This research addresses the role of facemasks in helmet systems, focusing on their dynamic impact performance and material properties. A total of 150 facemasks from 30 types and five helmets were tested both quasi-statically and dynamically. The facemasks were grouped by style and helmet type for comparison. Dynamic testing involved three different locations and two speeds, with data collected for maximum angular velocity and linear acceleration. Quasi-static stiffness, performed at two locations, was compared with collected dynamic results to determine the strength of relationship. A concussion metric, HARM, was calculated from collected data and compared across style groups.

Aging is a risk factor for myriad diseases and is often associated with the accumulation of senescent cells. Cellular senescence, a process of irreversible cell cycle arrest, is associated with various changes in cellular morphology and function. While senescence will occur naturally in a time-dependent manner, it can be prematurely induced by various extrinsic and intrinsic factors. Most senescence studies model cell behavior in two-dimensional (2D) microenvironments; however, these models are not physiologically relevant.
