Deep-sea fishing presents significant challenges for individuals with upper-limb differences, particularly when using prosthetic devices that are not designed for recreational fishing activities. This project developed a custom prosthetic fishing rod attachment for a user who had lost her forearm and desired a safe, comfortable, and independent method of fishing. Through interviews with the sponsor and an iterative design process, a lightweight aluminum attachment was created to interface directly with the user's existing prosthetic quick-connect system.
The design process included concept generation, CAD modeling, rapid prototyping with 3D-printed models, manufacturing, and user-centered refinement. The final design consists of a welded …

Endovascular procedures require devices with widely varying mechanical properties: flexibility for navigating tortuous vessels and rigidity for stable therapeutic delivery. Suction-actuated variable stiffness sheaths address this challenge by incorporating axial wire string arrays that mechanically couple under suction to increase flexural rigidity. However, prototype devices achieve stiffening ratios below theoretical predictions, suggesting that interlayer mechanics and string positioning play a critical role. This study investigates whether interweaving polytetrafluoroethylene (PTFE) tape within the string array enhances flexural rigidity modulation and curvature stability. Prototype devices with varying wrap configurations were fabricated and evaluated using flexural testing, curvature stability testing, and mathematical modeling. …

Hybrid structures formed between single-stranded DNA (ssDNA) and single-walled carbon nanotubes (SWCNT) have emerged as promising components in future direct DNA sequencing technologies due to their unique morphological and electronic properties. The nanoscale configuration of these hybrids, including the DNA wrapping angle, pitch, and contour behavior, directly influences their potential for stable electronic readout. This thesis presents an experimental study of DNA-SWCNT hybrid morphology using a fixed, reproducible wrapping protocol designed to minimize environmental variability and isolate the influence of DNA nucleotide sequence on hybrid structure. As described in prior work, this protocol produces well-dispersed hybrids suitable for high-resolution imaging. …

Hamstring strain injuries are among the most common and recurring injuries in sports, occurring most often during the terminal swing phase of gait. Despite progress in rehabilitation, the incidence of injury has only increased, suggesting a need for a novel intervention. We designed a passive, assistive, wearable device with two specific criteria in mind: to offload the hamstrings during running, and do so without hindering normal kinematic patterns. The device consists of an adjustable elastic band anchored proximally at the waist and distally just above the ankle joint center. Persistent gait asymmetry following anterior cruciate ligament reconstruction (ACLR) is a recognized contributor to post-traumatic knee osteoarthritis, yet the independent role of pain in driving altered movement patterns remains poorly understood due to confounding factors inherent to clinical populations. This study used exercise-induced muscle damage (EIMD) as a controlled experimental model to isolate the effects of musculoskeletal pain on gait, independent of kinesiophobia, structural damage, and proprioceptive disruption. Eighteen healthy adults underwent unilateral eccentric calf-raise protocols to induce delayed onset muscle soreness (DOMS), with gait assessed at baseline, 48 hours, and 168 hours post-exercise using bilateral inertial measurement units. Contemporary research suggests that shear force is a major contributor to discomfort and pressure injury. However, the variation of shear forces during human-exoskeleton interaction dynamics is a highly unexplored field. The high cost of commercial triaxial force sensors may be a major factor in the notable lack of research in this field. Therefore, in this paper, we present a low-cost, 3D-printed triaxial force sensor designed specifically to measure the triaxial interaction forces between an exoskeleton and its user. Arteriovenous (AV) grafts are commonly used to provide vascular access for hemodialysis in patients with end-stage renal disease. Despite their widespread use, AV grafts are prone to complications such as stenosis and thrombosis. Early detection of these conditions remains challenging with current monitoring methods too costly or insufficient. This work presents the design, fabrication, and validation of an LC pressure sensor embedded within a model AV graft to enable real-time monitoring. Mechanical loading is known to regulate bone remodeling by driving interstitial fluid flow which stimulates cells and drives nutrient transport within the trabecular network. In microgravity, the absence of mechanical stimulation or loading suppresses convective flow processes, causing diffusion-driven nutrient mixing and renewal, and accelerated bone loss. This thesis investigates how oscillation frequency and trabecular bone density jointly control nutrient mixing and wall shear stress within trabecular cavities.

These manual processes can introduce variability, reduce efficiency, and increase the cost of biomedical research. To address these challenges, our lab is developing a modular robotic platform, iEye™, to automate key steps in cell biology workflows [1]. Cancer remains a predominant health challenge that is responsible for a significant portion of global morbidity and mortality. Epirubicin (EPI), a chemotherapeutic anti-cancer drug, has shown remarkable efficacy in combating various malignancies. However, it is known to have undesirable side effects on the heart, collectively referred to as cardiotoxicity. This research investigates the adverse effects of chemotherapy on cardiac contractility through an in vitro examination of the mechanics of healthy and infarcted animal heart tissues. Objective: Heart rate variability (HRV), quantified via the instantaneous frequency (IF) of arterial pulse signals, is a key indicator of cardiovascular (CV) regulation. This study presents a theoretical and experimental investigation of how motion artifacts (MA) influence IF-and thus HRVmeasured with a tactile sensor, emphasizing underlying mechanisms rather than population-level validation. Approach: Building on a previously developed single-degree-of-freedom (SDOF) model of the tissue-contact-sensor (TCS) stack between the sensor and the artery, MA is represented as low-frequency baseline drift and time-varying system parameters (TVSP) of the TCS stack. Patellofemoral Pain Syndrome (PFPS) is the most prevalent overuse injury in cycling, often linked to altered neuromuscular activation patterns of the quadriceps and hamstrings. While sensory feedback has successfully modified cyclists posture, there is a critical lack of evidence-based interventions targeting the underlying muscle activation imbalances associated with chronic knee pain. Background: Microgravity exposure alters cardiovascular loading, yet its impact on left atrial flow dynamics and thrombotic risk remains poorly understood. This study investigates how spaceflight-relevant microgravity-induced changes in cardiac outflow affect left atrial hemodynamics in healthy individuals and patients with atrial fibrillation.
Methods: Patient-specific left atrial models were generated for three healthy individuals and three AF patients. Computational fluid dynamics (CFD) simulations were performed using each patient’s baseline mitral outflow waveform and two modified waveforms representing short- and long-duration post-flight cardiac loading changes derived from echocardiographic observations. Anterior knee pain affects approximately 25% of adults and is associated with altered joint mechanics. Patellofemoral joint (PFJ) models vary widely in complexity, with no established gold standard. Musculoskeletal models efficiently estimate joint loading but do not capture tissue-level mechanics, whereas finite element (FE) models provide tissue-level predictions at greater computational cost. This thesis developed a workflow to evaluate relationships between musculoskeletal model-predicted PFJ loads and FE-predicted cartilage mechanics, examined the impact of altered quadriceps activation, and introduced improved computational models.

Split-thickness skin grafts are widely used to treat chronic wounds. Procedure design requires surgeons to predict how much a patch of the patient's own skin expands when it is meshed with rows of slits and stretched over a larger wound area. Accurate prediction of graft expansion remains a challenge, with current models overestimating the actual expansion, leading to suboptimal outcomes. Crimped fibers within ligaments allow for recruitment which means the viscoelastic properties of stress relaxation and creep are not inversely related. A feasibility study using ACLs and large-scale crimp models to study viscoelasticity, led to an in-depth analysis of the scapholunate interosseous ligament. Spinal ligaments play a crucial role in maintaining the mechanical stability of the lumbar spine. These dense, collagenous tissues not only resist excessive motion but also distribute loads between spinal components to protect neural structures. Traditionally, the mechanical response of ligaments has been modeled using linear elastic assumptions, which treat the tissue as having a constant stiffness regardless of loading history. While this simplifies analysis, it fails to capture the time-dependent behaviors, such as creep, stress relaxation, and hysteresis, that are characteristic of biological tissues.

This dissertation addresses four specific aims that collectively attempted to advance the experimental and computational analysis of total knee arthroplasty (TKA) components. The first study focused on the development of a novel whole knee joint simulator capable of simultaneous tibiofemoral and patellofemoral knee loading. This simulator employs custom fixturing to facilitate dynamic, unconstrained, muscle-driven PF articulation alongside controlled TF contact mechanics. Validation against experimental measurements showed strong agreement, demonstrating the simulator's potential as a valuable tool for future TKA design and surgical technique investigations.

Intraoperative neuromonitoring (IONM) plays a critical role in minimizing nerve damage during surgeries by providing real-time feedback on neural integrity. This study evaluated models associated with deep learning and machine learning models for electromyography classification of signal during intraoperative neuromonitoring (IONM). The CNNLSTM model achieved the highest accuracy (85.2%), outperforming traditional models like KNN (53%), RF (62%), and CNN (76%). This demonstrates the degree to which the CNN-LSTM model can gain insight into temporal and spatial dependencies throughout the EMG signals, which makes it optimal for real-time classification in IONM applications.
