This thesis explores the micromechanical environment induced when cyclically compressing hydrogels via finite element modeling and experimentally on the impact of loading on mesenchymal stem cells (MSCs) when encapsulated withing 3D hydrogel matrices. Degenerative joint diseases, characterized by cartilage degradation, present significant challenges due to cartilage's limited self-repair capacity. Innovative approaches, including stem cell-based therapies and engineered biomaterials, have emerged as promising strategies for cartilage repair and regeneration. The advancement of additive manufacturing technologies has opened new avenues for fabricating biocompatible and structurally functional bone tissue scaffolds, essential in treating osseous fractures, defects, and diseases. This research aims to develop mechanically strong, dimensionally precise, and patient-specific porous bone tissue scaffolds that provide both structural integrity and biological functionality. Through three interconnected studies, several key challenges in the fabrication of these structures using the PME additive manufacturing process are addressed. An accurate representation of cellular mechanobiology necessitates the inclusion of subcellular elements characterized by minute masses and dimensions. These minute objects yield multiscale dynamic models with disproportionate terms, which require inordinate amounts of computational time to simulate. The computational requirements limit the time span of the simulation to time histories shorter than one second, even when employing supercomputers. This work presents a high-speed approach to simulating the mechanobiology of stem cells. Biomanufactured 3D-organoids show promise for modeling and treating various conditions, particularly insulin-dependent diabetes mellitus (type-1 diabetes). Pancreatic islet organoids serve as sustainable sources of insulin-producing β-cells, potentially restoring insulin regulation in patients and providing a long-term solution for diabetes. Hydrogel droplet printing is an emerging technique in regenerative medicine due to its advantageous tunability and biocompatibility; however, challenges of consistency, quality control, and scalability persist. This paper describes the design, manufacturing, and testing of a novel controllable hypoxic incubator with fully functional oxygen gas control and temperature control in a humid environment. On the current market, a majority of the few hypoxic incubators use pre-mixed gas that does not offer precise control over gas concentration. Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. Myocardial infarction (MI) is one of the diseases with the highest mortality rate. Following MI, myocardium experiences abrupt changes in its loading condition due to the presence of infarct. In response to such changes, myocytes undergo adaptations to maintain homeostasis. However, maladaptation can happen and lead to remodelling, in which the left ventricle (LV) gradually loses its function and eventually turns into heart failure. In this work, the approach to the manipulation of alginate artificial cell soft-microrobots, both individually and in swarms is shown. Fabrication of these artificial cells were completed through centrifugation, producing large volumes of artificial cells, encapsulated with superparamagnetic iron oxide nanoparticles; these artificial cells can be then externally stimulated by an applied magnetic field. The construction of a Permeant Magnet Stage (PMS) was produced to manipulate the artificial cells individually and in swarms. The stage functionalizes the permanent magnet in the 2D xy-plane. It is widely accepted that cells behave differently responding to the stiffness of their extracellular matrix (ECM). Such observations were made by culturing cells on hydrogel substrates of tunable stiffness. However, it was recently proposed that cells may sense how strongly they are tethered to ECM, not the local stiffness of ECM. To investigate both hypotheses, we developed a method to fabricate Janus polyacrylamide (PAAM) gels. Muscle mechanical behavior potentially plays an important role in some of the most common bladder disorders. These include overactive bladder, which can involve involuntary contractions during bladder filling, and impaired contractility or underactive bladder, which may involve weak or incomplete contractions during voiding. Actin-myosin cross-bridges in detrusor smooth muscle (DSM) are responsible for contracting and emptying the bladder. The total tension produced by muscle is the sum of its preload and active tensions. Experimental results show that collagen fibers exhibit stress relaxation under tension and a highly anisotropic distribution. To further develop the earlier model of Stender [1], the collagen constituent was updated to reflect its intrinsic viscoelasticity and anisotropic distribution, and integrated with an existing mixture model with glycosaminoglycans and ground substance matrix. A two-term Prony series expansion of the quasi-linear viscoelastic model was chosen to model the viscoelastic properties of the collagen fibers. Material parameters were determined by using the simplex method to minimize the sum of squared errors between model results and experimental stress relaxation data of tissue in tension. …
