The development of reliable hydrogel-based scaffolds for extrusion bioprinting remains limited by the poor structural fidelity of low-viscosity bioinks and the lack of robust, high-throughput quality evaluation methods. This dissertation can resolve such challenges by developing the combination of optimized hydrogel formulations, cryogenic-assisted bioprinting, and artificial intelligence (AI)-based scaffold evaluation to the use of tissue engineering and preclinical cancer modelling applications. To assess the rheological behavior, printability, mechanical properties, and biocompatibility of the alginate – gelatin (Alg–Gel) system, a novel system of hydrogel was developed and characterized using Alg–Gel hydrogel system. Hydrocephalus is developed when the flow of cerebrospinal fluid is obstructed in the ventricles and a pressure build-up is generated within the brain. Ventricular shunts are used to remove excess fluid from the brain, but these shunts have a common problem of failure due to the shunt being obstructed by the build-up of astrocytes. To address this, two sets of 27 designs of ventricular shunts were identified and analyzed with parameters that could potentially reduce obstruction risks. While marker-based motion capture remains the gold standard in measuring human movement, accuracy is influenced by soft-tissue artifacts, particularly for subjects with high body mass index (BMI) where markers are not placed close to the underlying bone. Obesity influences joint loads and motion patterns, and BMI may not be sufficient to capture the distribution of a subject’s weight or to differentiate differences between subjects. Subjects in need of a joint replacement are more likely to have mobility issues or pain, which prevents exercise. Obesity also increases the likelihood of needing a total joint replacement. Traumatic brain injury (TBI) continues to have the greatest incidence among athletes participating in American football. The headgear design research community has focused on developing accurate computational and experimental analysis techniques to better assess the ability of headgear technology to attenuate impacts and protect athletes from TBI. Despite efforts to innovate the headgear system, minimal progress has been made to innovate the faceguard. It has been demonstrated that although many varieties of upper limb prosthetics exist, commercially available prosthetics are outdated and unsatisfactory. Ineffectiveness and limitations have led to some prosthesis wearers having to own multiple devices, whereas others have given up on them entirely. Even though ample research has been conducted to design and test new hand designs, the industry appears to rest in an overall stagnated state.

Blalock-Taussig (BT) shunts are used for defects that affect the flow of blood from the right ventricle, through the pulmonary artery, and to the lungs. Arteriovenous (AV) fistula is one type of vascular access which is a surgically created vein used to remove and return blood during hemodialysis. Quantification of plantar tissue behavior of the heel pad is essential in developing computational models for predictive analysis of preventive treatment options such as footwear for patients with diabetes. Simulation based studies in the past have generally adopted heel pad properties from the literature, in return using heel-specific geometry with material properties of a different heel.
