Mechanical loading is known to regulate bone remodeling by driving interstitial fluid flow which stimulates cells and drives nutrient transport within the trabecular network. In microgravity, the absence of mechanical stimulation or loading suppresses convective flow processes, causing diffusion-driven nutrient mixing and renewal, and accelerated bone loss. This thesis investigates how oscillation frequency and trabecular bone density jointly control nutrient mixing and wall shear stress within trabecular cavities.

Mathematical Modelling and Numerical Simulation with Applications

Effective carbon dioxide (CO₂) storage is essential for mitigating climate change amid increasing global greenhouse gas emissions. This study investigates the influence of geomechanics on CO₂ storage performance within carbon capture and storage (CCS), focusing on structural, residual, and solubility trapping mechanisms using a fully coupled modeling framework. Two numerical models, with and without geomechanical effects, are developed to evaluate impacts on reservoir behavior, CO₂ migration, and trapping efficiency. Each mechanism is analyzed separately and within an integrated framework to assess their combined contributions. Background: Microgravity exposure alters cardiovascular loading, yet its impact on left atrial flow dynamics and thrombotic risk remains poorly understood. This study investigates how spaceflight-relevant microgravity-induced changes in cardiac outflow affect left atrial hemodynamics in healthy individuals and patients with atrial fibrillation.

Methods: Patient-specific left atrial models were generated for three healthy individuals and three AF patients. Computational fluid dynamics (CFD) simulations were performed using each patient’s baseline mitral outflow waveform and two modified waveforms representing short- and long-duration post-flight cardiac loading changes derived from echocardiographic observations. Gait impairments arise from systemic diseases, age-related degeneration, musculoskeletal dysfunctions, or neurological conditions. While traditional rehabilitation can be effective, they often face challenges such as high costs, inaccessibility, and low patient engagement. To address these challenges, my work introduces a virtual reality-based rehabilitation (VRBR) system, integrating real-time motion and electromyographic (EMG) muscle activation feedback with a gamified virtual environment for enhanced adaptability and engagement. The system includes a custom-designed hip-exoskeleton that provides adaptive spring-like assistance or resistance, supporting both mobility-impaired users and strength training. Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. The focus of this dissertation is insect blood (Hemolymph). Hemolymph is analyzed by taking a materials science approach. Rheological characterization of hemolymph has been performed for the first time. Owing to a minute amount of available material, a new protocol was established where Magnetic Rotational Spectroscopy (MRS) with ferromagnetic nanorods was employed. The challenge was examining the microliter droplets' viscosity in less than a few minutes. This challenge was successfully resolved.

This dissertation aimed to (1) quantify morphological variations in the pediatric hip joint and (2) evaluate the sensitivity of an infant musculoskeletal model (MSM) to these variations, considering hip joint center estimation errors. A shape statistical model (SSM) of decedent infant femurs from the Ortolani collection was created using ShapeWorks, capturing key morphological features, such as variations in the femoral neck-shaft and anteversion angles. Seven synthetic femurs were generated from the SSM to create SSM-informed MSMs, which were systematically evaluated through kinematics and kinetics analyses in OpenSim. Total Knee Arthroplasty (TKA) is a widely performed surgical procedure aimed at alleviating pain and restoring function in patients with severe knee osteoarthritis. Despite its general success, disparities in postoperative outcomes have been observed across different racial and ethnic groups, with minority populations often experiencing less favorable results. Stroke is a leading cause of long-term disability, affecting thousands of individuals annually and significantly impairing their mobility, independence, and quality of life. Traditional methods for assessing motor impairments are often costly and invasive, creating substantial barriers to effective rehabilitation. This thesis explores the use of DeepLabCut (DLC), a deep-learning-based pose estimation tool, to extract clinically meaningful kinematic features from video data of stroke survivors with upper-extremity (UE) impairments.

Inactive adults often have decreased musculoskeletal health and increased risk factors for chronic diseases. However, there is limited data linking biomechanical measurements of generally healthy young adults to their physical activity levels assessed through questionnaires. Commonly used data collection methods in biomechanics for assessing musculoskeletal health include but are not limited to muscle quality (measured as echo intensity when using ultrasound), isokinetic (i.e., dynamic) muscle strength, muscle activations, and functional movement assessments using motion capture systems. While marker-based motion capture remains the gold standard in measuring human movement, accuracy is influenced by soft-tissue artifacts, particularly for subjects with high body mass index (BMI) where markers are not placed close to the underlying bone. Obesity influences joint loads and motion patterns, and BMI may not be sufficient to capture the distribution of a subject’s weight or to differentiate differences between subjects. Over the last couple of decades, numerous piezoelectric footwear energy harvesters (PFEHs) have been reported in the literature. This paper reviews the principles, methods, and applications of PFEH technologies. First, the popular piezoelectric materials used and their properties for PEEHs are summarized. Then, the force interaction with the ground and dynamic energy distribution on the footprint as well as accelerations are analyzed and summarized to provide the baseline, constraints, potential, and limitations for PFEH design. Traumatic brain injury (TBI) continues to have the greatest incidence among athletes participating in American football. The headgear design research community has focused on developing accurate computational and experimental analysis techniques to better assess the ability of headgear technology to attenuate impacts and protect athletes from TBI. The meshless method is an incredibly powerful technique for solving a variety of problems with unparalleled accuracy and efficiency. The pharmacokinetic problem of transdermal drug delivery (TDDD) is one such topic and is of significant complexity. The locally collocated meshless method (LCMM) is developed in solution to this topic. First, the meshless method is formulated to model this transport phenomenon and is then validated against an analytical solution of a pharmacokinetic problem set, to demonstrate this accuracy and efficiency. In most everyday activities, we head towards a specific goal by updating our choices for a more direct path. However, there are specific clinical tasks where taking the direct path is more challenging. Clinical investigations of optimizing a prosthesis involve the assessment of multiple parameter settings through trial and error rather than goal-directed optimization. We investigate if a human-in-the-loop optimization algorithm can guide manual alterations to a prosthesis-simulating device to reduce the ground reaction force on the contralateral limb. Designing earplugs adapted for the widest number of earcanals requires acoustical test fixtures (ATFs) geometrically representative of the population. Most existing ATFs are equipped with unique sized straight cylindrical earcanals, considered representative of average human morphology, and are therefore unable to assess how earplugs can fit different earcanal morphologies. Combining vibrating mesh nebulizers with additional new technologies leads to substantial improvements in pharmaceutical aerosol delivery to the lungs across therapeutic administration methods. In this dissertation, streamlined components, aerosol administration synchronization, and/or Excipient Enhanced Growth (EEG) technologies were utilized to develop and test several novel devices and aerosol delivery systems. Producing analytical solutions that can predict the post-collision motion of the colliding bodies require consistent modeling of the dynamics of the colliding bodies. Biomimicry is an approach in science and engineering to overcome human challenges by designing materials and systems modeled after nature. Selected applications of biomimicry include catheters, hearing devices, and artificial appendages such as arms, legs, and fingers. The inspiration for this study is the hydrofoil-like structured pectoral fin of the harbor porpoise whale that is ultimately useful for studying fluid-structure interactions, drag, lift, and laminar-to-turbulent transitions.
