Patellofemoral Pain Syndrome (PFPS) is the most prevalent overuse injury in cycling, often linked to altered neuromuscular activation patterns of the quadriceps and hamstrings. While sensory feedback has successfully modified cyclists posture, there is a critical lack of evidence-based interventions targeting the underlying muscle activation imbalances associated with chronic knee pain. This study aimed to develop and validate a novel, real-time biofeedback system that utilizes electromyography (EMG) to trigger vibrotactile cues, aiming to shift muscle onset timing earlier in the pedal stroke to mitigate PFPS-related imbalances. As the global population ages, the demand for wearable assistive technologies continues to rise, driven by their potential to enhance mobility and independence in older adults. Effectively designed controllers for lower-limb exoskeletons to assist sit-to-stand (STS) and walking are crucial for delivering efficient, safe, and comfortable assistance during daily activities. Traditionally, controller optimization involves biomechanical modeling and user-specific customization. Gait impairments arise from systemic diseases, age-related degeneration, musculoskeletal dysfunctions, or neurological conditions. While traditional rehabilitation can be effective, they often face challenges such as high costs, inaccessibility, and low patient engagement. To address these challenges, my work introduces a virtual reality-based rehabilitation (VRBR) system, integrating real-time motion and electromyographic (EMG) muscle activation feedback with a gamified virtual environment for enhanced adaptability and engagement. The system includes a custom-designed hip-exoskeleton that provides adaptive spring-like assistance or resistance, supporting both mobility-impaired users and strength training. This dissertation addresses four specific aims that collectively attempted to advance the experimental and computational analysis of total knee arthroplasty (TKA) components. The first study focused on the development of a novel whole knee joint simulator capable of simultaneous tibiofemoral and patellofemoral knee loading. This simulator employs custom fixturing to facilitate dynamic, unconstrained, muscle-driven PF articulation alongside controlled TF contact mechanics. Validation against experimental measurements showed strong agreement, demonstrating the simulator's potential as a valuable tool for future TKA design and surgical technique investigations.

Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. This thesis explores the micromechanical environment induced when cyclically compressing hydrogels via finite element modeling and experimentally on the impact of loading on mesenchymal stem cells (MSCs) when encapsulated withing 3D hydrogel matrices. Degenerative joint diseases, characterized by cartilage degradation, present significant challenges due to cartilage's limited self-repair capacity. This research addresses the role of facemasks in helmet systems, focusing on their dynamic impact performance and material properties. A total of 150 facemasks from 30 types and five helmets were tested both quasi-statically and dynamically. The facemasks were grouped by style and helmet type for comparison. Dynamic testing involved three different locations and two speeds, with data collected for maximum angular velocity and linear acceleration. Quasi-static stiffness, performed at two locations, was compared with collected dynamic results to determine the strength of relationship. A concussion metric, HARM, was calculated from collected data and compared across style groups.

This dissertation focused on modeling specimen-specific soft tissue structures in the context of joint replacement surgery. The research addressed four key aspects. The first study involved developing a workflow for creating finite element models of the hip capsule to replicate its torque-rotational response. Experimental data from ten cadaveric hips were used to calibrate the models, resulting in improved accuracy and relevance for surgical planning and implant design. The second study tackled the challenge of expediting the calibration of mechanical properties of the hip capsule to match patient-specific laxities. Inactive adults often have decreased musculoskeletal health and increased risk factors for chronic diseases. However, there is limited data linking biomechanical measurements of generally healthy young adults to their physical activity levels assessed through questionnaires. Commonly used data collection methods in biomechanics for assessing musculoskeletal health include but are not limited to muscle quality (measured as echo intensity when using ultrasound), isokinetic (i.e., dynamic) muscle strength, muscle activations, and functional movement assessments using motion capture systems. Biomechanical analysis is widely used to assess human movement sciences, specifically using three-dimensional motion capture modelling. There are unprecedented opportunities to increase quantitative knowledge of rehabilitation and recreation for disadvantaged population groups. Specifically, 3D models and movement profiles for human gait analysis were generated with emphasis on post-stroke patients, with direct model translation to analyze equivalent measurements while horseback riding in use of the alternative form of rehabilitation, equine assisted activities and therapies (EAAT) or hippotherapy (HPOT). Developmental Dysplasia of the Hip is the dysplasia or dislocation of an unstable femoral head in the acetabulum of the pelvis. This is a result of an abnormal or underdeveloped growth of the acetabulum and triradiate cartilages and/or an out of place and uncentered femoral head in the acetabulum. This underdevelopment, if not diagnosed and treated at an early age, may lead to abnormal and pain and osteoarthritis later in life. Because of its extreme rarity, the genetic disease arthrogryposis multiplex congenita (AMC) and the needs of individuals with the diagnosis are often overlooked. AMC refers to the development of nonprogressive contractures in disparate areas of the body and is characterized by decreased flexibility in joints, muscle atrophy, and developmental delays. Colton Darst, a seven-year-old boy from Indianapolis, Indiana, was born with the disorder, and since then, he has undergone numerous surgical interventions and continues to receive orthopedic therapy to reduce his physical limitations. This paper presents a biomechanical model of the rat forelimb to test theories of determining viscoelastic muscle parameters. Several biomechanical models of rat hindlimbs have been developed and have explored the effects of multi-muscle control during locomotion. The forelimb model uses two ball-and-socket joints to model clavicle and scapula movement. A third ball-and-socket joint is used at the shoulder and two hinge joints are used at the elbow and wrist. Scapula motion is further constrained by muscle and spring elements. Each forelimb has 11 degrees of freedom, and 23 Hill-type muscles. The model has been created in Animatlab, which includes …

Prior researchers have observed the effect of simulated reduced-gravity exercise. However, the extent to which lower-body positive-pressure treadmill (LBPPT) walking alters kinematic gait characteristics is not well understood. The purpose of the study was to investigate the effect of LBPPT walking on selected gait parameters in simulated reduced-gravity conditions. Twenty-nine college-aged volunteers participated in this cross-sectional study. Participants wore pressure-measuring insoles (Medilogic GmBH, Schönefeld, Germany) and completed three 3.5-min walking trials on the LBPPT (AlterG, Inc., Fremont, CA, USA) at 100% (normal gravity) as well as reduced-gravity conditions of 40% and 20% body weight (BW). Two studies are discussed in this manuscript each preceded by a literature review of the topic. The first review and study explore agility movements and the effect that alternative upper designs in shoes might have on ground reaction force measures of performance. Machine learning is a promising approach to evaluate human movement based on wearable sensor data. A representative dataset for training data-driven models is crucial to ensure that the model generalizes well to unseen data. However, the acquisition of sufficient data is time-consuming and often infeasible. We present a method to create realistic inertial sensor data with corresponding biomechanical variables by 2D walking and running simulations. While no doubt valuable to safety, the post-hoc approach employed in these studies does little to explain the why and how behind the risk factors. Human movement is an instinctive yet challenging task that involves complex interactions between the neuromusculoskeletal system and its interaction with the surrounding environment. One key obstacle in the understanding of human locomotion is the availability and validity of experimental data or computational models. Corresponding measurements describing the relationships of the nervous and musculoskeletal systems and their dynamics are highly variable. Bone mineral density (BMD), among other factors, largely effect the initial stability of the cementless tibial tray component in a total knee replacement (TKR), where increased motion at the tray-bone interface hinders bony ingrowth. With a lack of bony ingrowth, the cementless implant will not experience long-term success. Understanding which factors influence initial stability yields insight into surgical technique considerations and help inform a surgeon’s implant choice.
