The development of reliable hydrogel-based scaffolds for extrusion bioprinting remains limited by the poor structural fidelity of low-viscosity bioinks and the lack of robust, high-throughput quality evaluation methods. This dissertation can resolve such challenges by developing the combination of optimized hydrogel formulations, cryogenic-assisted bioprinting, and artificial intelligence (AI)-based scaffold evaluation to the use of tissue engineering and preclinical cancer modelling applications. About 40% of the US construction workforce experiences high-level fatigue, which leads to poor judgment, increased risk of injuries, a decrease in productivity, and a lower quality of work. Excessive fatigue from working in unpleasant working conditions, long working hours, or heavy workloads can aggravate fatigue's adverse effects, leading to work-related musculoskeletal disorders (WMSDs) and productivity loss. Mechanically robust solid-state nanopores have the potential to be the next generation DNA sensing platforms. However, mass production and limited base-calling accuracy are the hurdles for solid-state nanopore based DNA sensing. In order to solve these problems, a polymer dual-nanopore device fabricated via high throughput nanoimprint lithography (NIL) was proposed to sequence DNA by time-of-flight (ToF) measurement. As a proof of concept, this study presents mononucleotides discrimination via ToF measurement using polymer in-plane dual-nanopore device.

Neuromuscular electrical stimulation (NMES) is a technology where skeletal muscles are externally stimulated by electrodes to help restore functionality to human limbs with motor neuron disorder. This dissertation is concerned with the model-based feedback control of the NMES quadriceps muscle group-knee joint dynamics. A class of nonlinear controllers is presented based on various levels of model structures and uncertainties. The two main control techniques used throughout this work are backstepping control and Lyapunov stability theory.
