Plasma vitamin A is not a reliable indicator of vitamin A status. Therefore, the feasibility of using the relative dose response (RDR) assay as a method for determining vitamin A status of calves was examined. In addition, effects of colostrum quality and season on vitamin A status and health of calves were determined. Blood was sampled from the jugular vein of 16 newborn calves to establish values over a 24 h period for immunoglobulin M (IgM) concentration, hematocrit, serum protein concentration, and leukocyte differentials. Liver biopsies were performed at birth, and successfully completed for 11 calves. Blood samples were obtained …

Milk samples were collected from 108,312 dairy cows during 1601 farm visits made between January 1991 and June 1995. The herd visits were made by personnel from the Central Laboratory of the Quality Milk Promotion Services at Cornell University (Ithaca, NY) to farms located in central New York and northern Pennsylvania. Dairy Herd Improvement Association records were available for 32,978 cows in 327 herds. Intramammary infections, as defined by positive milk cultures, were present in 48.5% of all cows and in 36.3% of cows in herds enrolled in the Dairy Herd Improvement Association. A series of germanium- or silicon-centred heteropolytungstates (polyoxometalates) with the Barrel, Keggin or double Keggin structure were evaluated in vitro for their effects against influenza A (IV-A) and B (IV-B) viruses. Their 50% effective concentrations (EC50) against recent isolates of IV-A (H1 N1 ) and IV-B ranged from 0.1 to 7.8 μM; against IV-A (H3N2), the EC50 concentrations were often 10-fold higher. Recent clinical isolates of IV-A were generally more susceptible to these antiviral effects than older, laboratory-adapted strains. A series of acyclic phosphonomethylether nucleosides were synthesized and then evaluated for inhibitory activity against respiratory viruses of clinical significance using CPE inhibition, neutral red uptake and virus yield reduction assays. Of the 20 compounds synthesized, none significantly inhibited influenza A or B viruses or respiratory syncytial virus strains A2, Long or 18537; the selective indices (SI) were less than 10. A new compound, GS-2128 (2R, 5R-9-[2,5-dihydro-5-(phosphonomethoxy)-2-furanyl]adenine; D4API), selectively inhibited adenovirus 5 (SI>10) as did GS-0577 (9-(3-hydroxy-2-phosphonylmethoxypropyl)-adenine; HPMPA) and GS-0504 [(S)-1-[3-hydroxy-2-(phosphonylmethoxypropyl)]-cytosine; HPMPC]. Advances in in vitro fertilization and in vitro culture techniques have allowed considerable progress in identifying physiological requirements of mammalian embryos. Parrish et al. (11) reported a major breakthrough on in vitro fertilization when his group identified heparin as an important factor for the capacitation of spermatozoa. Capacitation is necessary for fertilization of matured oocytes. During the precoculture era of embryos (prior to 1980), the development of early preirnplantation embryos was vary limited regardless of medium, medium supplement, gas atmosphere, osmolarity or pH used (7). Transgenic mice were produced by microinjection of a DNA construct composed of the bovine κ-casein (κ-CN) cDNA under the control of the goat β-CN 5' promoter elements and 3' flanking regions into pronuclear-stage embryos. The gene construct targeted the expression of bovine κ-CN RNA to the mammary gland and secretion of bovine κ-CN in the milk. In the three lines studied (BC-7, BC-31 and BC-67) the transgene was stably integrated and propagated as a Mendelian locus. Cryptosporidium parvum, a protozoan parasite infecting epithelial cells lining the intestinal tract of animals and humans, causes fulminate diarrhea and malabsorption of essential nutrients following damage to the cellular brush border. The present study was undertaken to evaluate the use of a pentose sugar (D-xylose) in a standardized tolerance test as an indicator of malabsorption due to cryptosporidiosis in the dexamethasone (DEX)-immunosuppressed adult C57BL/6N mouse model. One group containing 14 experimental mice (in addition to appropriate control groups) was immunosuppressed using daily intraperitoneal injections of DEX (125 μg/mouse). But there is always room for improvement and as grain prices rise it is crucial that maximum benefit is derived from the cheapest feed source - the pasture.With this in mind Ruth Dilley looks at the Dairy Farm Performance Program - a comprihensive farm database developed by Agriculture Western Australia.

An increase in the export of UHT (Ultra Heat Treated) milk to Asian markets is potentially worth millions of dollars to the dairy inmdustry. This increase however, relies on the shelf life of UHT products being extended to nine months. Spoilage of UHT milk can be caused by bacterial spores which originate on-farm and are resistant to processing. Expansion in the UHT market therefore relies on a continued reduction in the number of these spores getting into raw milk. Caroline Love, Ian Bell and Martin Robertson report on the sources of spores entering milk on-farm and the control methods available.

Milk glands, dissected out and collected from Glossina morsitans centralis (Machado) females, artificially inoculated at the third-instar larval stage with a virus suspension obtained from hypertrophied salivary glands of wild-caught virus-infected Glossina pallidipes (Austen), were processed for routine electron microscopy and examined for pathological changes. They were compared to milk glands dissected out from normal female G.m. centralis at the same stage of pregnancy cycle. Upon dissection there were notable physical differences between control and virus-infected milk glands. Histologically, some areas of the gland developed severe degeneration while other areas developed less severe pathological changes. Ultrastructural studies revealed the presence …

This manual on pasture establishment andmanagement is designed to help farmers grow productive pastures. It deals with annual and perennial pastures on dryland and irrigated farms in the above 700 mm rainfall zone and may be used by individual farmers or discussion groups. Input from discussion groups will improve futureeditions of the manual.Farmers frequently need to reseed their pastures. Pastures deteriorate and new, more productive species and cultivars come on to the market. Also improved techniques are developed to tackle land management problems like salinity, waterlogging and acidity. All these situations lead to a reseeding operation.

Transgenic mice produced in this investigation contained the proximal promoter elements, the genomic coding sequence and the 3' flanking region of bovine β-casein and the 5' regulatory elements of bovine α-lactalbumin. Specifically, the gene construct included the TATAA box, CAAT box and polyadenylation signal of bovine β-casein in addition to the 5' regulatory elements of the bovine α-lactalbumin gene. Four lines of transgenic mice were generated; they expressed bovine β-casein in their milk at concentrations of up to 10 mg mL-1. The present invention relates to a process for the removal of cholesterol from a processed or unprocessed dairy product, which process comprises: (a) obtaining a processed or unprocessed dairy product containing cholesterol (b) contacting the dairy product at a temperature of between about 35° and 80° C. with an amount of saponin effective to bind up to about 90% of the cholesterol present in the dairy product; (c) contacting a temperature of between about 35° and 80° C the cholesterol: saponin with an effective amount of diatomaceous earth; (d) separating the insoluble cholesterol: saponin/diatomaceous earth by filtration or centrifugation; and …

Most prople have heard of the Awassi, even if they have never seen one. This unique breed of fat tail sheep, imported into Astralia in 1987, was released from quarantine for commercial development nearly 12 months ago. Its supporters believe Awassis could form the basis of a high-value export industry while replacing imports. Developments over the next few years will test its potential and should result in the Awassi playing an important role in an increasingly diversified Australian farming scene.

Today's consumers are demanding food products that are "low-fat", "reduced fat", "high in -unsaturated fatty acids", and "low cholesterol". This trend in food product selection is at the advice of many health professionals. Current health recommendations are to lower the dietary intake of fat, especially saturated fat. In 1992, total dairy sales accounted for over 10% of total food sales (Milk Industry Foundation, 1993). Since dairy products are perceived as being hypercholesterolemic, a market exists for dairy products which are "lower in fat" and/or "lower in saturated fat". Mycotoxins are ubiquitous, mold-produced toxins that contaminate a wide variety of foods and feeds. Ingestion of mycotoxins cause a range of toxic responses, from acute toxicity to long-term or chronic health disorders. Some mycotoxins have caused outbreaks of human toxicoses, and at least one mycotoxin, aflatoxin B1, is a presumed human hepatocarcinogen. As part of a comprehensive effort to curtail the adverse health effects posed by mycotoxins, substantial research has been conducted to determine the mechanism of action of mycotoxins in animals. A preparative isoelectric focusing (IEF) method was applied to separate skim milk proteins using the Rotofor device in a pH 3-10 gradient containing 4 M urea/1% triton X-100. Each of the 20 fractions obtained from the Rotofor device was then analyzed by polyacrylamide gel electrophoresis (PAGE). Both urea-PAGE and SDS-PAGE were used to separate purified caseins and skim milk resulting in comparable two-dimensional pattern. The major bovine caseins (αs1, αs2, β, κ-casein) were resolved better on urea-PAGE. The αs1- and β -casein were focused at pH ~ 4.5 and 4.8, respectively, whereas αs2 …
