The innate immune system plays an important role in the early defense against pathogens. To better understand the chicken’s innate inflammatory responses, we concurrently examined the cellular responses at the injection site and in the blood following intradermal (i.d.) injections of lipopolysaccharide (LPS) or peptidoglycan (PGN) into the pulp of growing feathers (GFs). Both LPS from Gram-negative bacteria and PGN from Gram-positive bacteria are potent stimulators of innate inflammatory responses. The time-course study conducted revealed similarly elevated levels of heterophils and macrophages in injected GF-pulps during the first 24 h for both LPS and PGN. However, PGN also stimulated rapid …

There is a demand for novel pathogen mitigation strategies in antibiotic-free poultry production, which has triggered the development of various probiotic application methods. One application method is the in ovo (in egg) administration of probiotics on incubation day (d) 18, where the unhatched chick consumes a probiotic before hatch and prior to being exposed to environmental pathogens. Therefore, the objective of this study was to evaluate the influence of in ovo L. animalis, B. licheniformis, or L. animalis + B. licheniformis (106 cfu/50 mL L each) administration on hatch performance (N = 21 egg flats/treatment, 30 …

Objective, robust, and repeatable assessments of fear responses of poultry can lead to improvements in research techniques, the validity of test results, and ultimately bird welfare. The objective of this study was to examine the effect of 2 different holding methods on broiler fear responses during the inversion test, a standardized method of assessing fear in poultry. On D15 and D34 mixed-sex broilers (N = 80) were inverted and held either by their shanks (N = 40) or feet (N = 40) at arm's length away from the experimenter for 30 s. The frequency of wing flapping, head movement, attempts …

Dietary CP reductions in broilers can result in increased FCR when 6 or 7 feed-grade amino acids are allowed to enter formulation, and a partial explanation may be a dietary limitation of His. Histidine requirement data are sparce in the current body of literature, especially beyond 3 wk post-hatch. Therefore, a His dose-response experiment (expressed as a percentage of Lys) was conducted with male Cobb 500 broilers from 15 to 35 d post-hatch. A His deficient test diet was formulated to contain a digestible His to digestible Lys (dHis:dLys) ratio of 28.0%. Graded increments of L-His were added to the …

Heat stress (HS) and water scarcity are significant challenges to sustainable poultry production worldwide. It is, therefore, critical to identify effective strategies to prevent, withstand, or adapt to these challenges. After four generations of divergent selection for water efficiency, the present study was undertaken to determine the effect of HS on meat quality and muscle myopathy incidences in high (HWE)- and low (LWE)-water efficient broilers. Day-old male chicks (240 chicks/line) were allotted randomly by line and body weight-matched groups to 12 controlled-environmental chambers (2 pens/chamber). At d29, birds were exposed to 2 environmental conditions …

Bacterial chondronecrosis with osteomyelitis (BCO) lameness is a leg disorder in broilers leading to huge economic losses, food safety concerns, and animal welfare issues in the poultry industry. The active metabolite of vitamin D3, 1,25-dihydroxyvitamin D3, plays pivotal roles in mineral homeostasis, bone health, and immune systems which are essential for protection against BCO. Thus, we hypothesized that supplementation with a Solanum glaucophyllum-derived source of 1,25-dihydroxyvitamin D3 (1,25(OH)2D3-glycosides) would be an effective measure to control lameness. Here, we report the optimum concentration of 1,25(OH)2D3-glycosides …

In the food industry, assessing the quality of poultry carcasses during processing is a crucial step. This study proposes an effective approach for automating the assessment of carcass quality without requiring skilled labor or inspector involvement. The proposed system is based on machine learning (ML) and computer vision (CV) techniques, enabling automated defect detection and carcass quality assessment. To this end, an end -to - end framework called CarcassFormer is introduced. It is built upon a Transformer -based architecture designed to effectively extract visual representations while simulta- neously detecting, segmenting, and classifying poultry carcass defects. Our …

Previous studies with broiler breeders indicate a P retention threshold when fed daily dietary levels of non-phytate P (NPP) exceeding 320 mg. Fibroblast growth factor 23 (FGF23) is a hormone secreted by osteocytes which modulates P retention and could be the biological agent which controls the P threshold in breeders. To evaluate the relationship between FGF23 and the P retention threshold, a 4 -wk study with 32 -wk -old breeders was conducted with 6 dietary treatments with daily NPP intake of 216 to 576 mg/d/h with increments of 80 mg/kg diet. The goals were 1) to …

Further reductions of crude protein in broiler diets may cause essential amino acids traditionally considered to be supplied at adequate levels to become limiting. Requirement data for histidine are currently scarce and this amino acid is uniquely able to be stored within the body. Thus, the objectives of this study were to evaluate female Cobb 500 broiler growth performance and carcass characteristic responsiveness to increasing digestible histidine to digestible lysine ratios (dHis:dLys) during a 22- to 42-day feeding period and determine if dHis:dLys in preceding feeds influence the former responses. Starter diets were formulated to contain dHis:dLys of 33% or …

Ensuring good water quality is one of the key management strategies to promote optimal broiler health and performance. A few studies have assessed the microbial status of drinking water for poultry, but there is insufficient information on biofilm in the drinking water lines within the poultry housing facilities. The buildup of microbes in the drinking water system could potentially result in biofilm formation which eventually deteriorates water quality. Hence, in this preliminary study, we sought to evaluate biofilm formation in the drinking water lines of selected broiler houses and characterize their microbial composition. Three random houses in each of 5 …

Bacterial chondronecrosis with osteomyelitis (BCO) lameness is a bone disease characterized by the translocation of bacteria from the gastrointestinal tract, which colonize microfractures in broiler leg bones caused by rapid animal growth rate and weight gain, resulting in lameness. As such, BCO lameness represents a significant challenge for the poultry industry. This study aims to evaluate the effect of spraying broiler chicks on d0 at hatch with an Enterococcus faecium probiotic on the incidence of BCO-induced lameness, utilizing a Staphylococcus aureus challenge model. There were four treatments: (1) negative control (no probiotic + no challenge, NC); (2) positive control (no …

Bacterial chondronecrosis with osteomyelitis (BCO) lameness is the most critical animal health and welfare issue facing the broiler industry worldwide. It is estimated that 1 to 2% of bird condemnation at marketing age is caused by BCO lameness, resulting in tens of millions of dollars in annual losses. Fast-growing broilers are prone to mechanical stress that triggers bacterial translocation across epithelial barriers into the bloodstream, followed by bacterial colonization in the growth plate of long bones, and eventually, bone necrosis and lameness. Mycotoxins (MTX) are secondary metabolites produced naturally by microfungi, of which deoxynivalenol (DON …

Lameness disease attributed to bacterial chondronecrosis with osteomyelitis in broilers affects production, animal welfare, and food safety in the poultry industry. The disease is characterized by necrotic degeneration of the rapidly growing femora and tibiae due to bacterial translocation from the respiratory or gastrointestinal tracts into the blood circulation, eventually colonizing the growth plate of the long bones. To investigate the etiology, pathogenesis, and intervention measures for BCO, developing an experimental model that reliably induces BCO lameness is of the utmost importance. In the past, we have employed a wire -flooring model and a litter -flooring model administered with a …

Salmonella remains a top bacterial pathogen implicated in several food-borne outbreaks, despite the use of antimicrobials and sanitizers during production and processing. While these chemicals have been effective, Salmonella has shown the ability to survive and persist in poultry processing environments. This can be credited to its microbial ability to adapt and develop/acquire tolerance and/or resistance to different antimicrobial agents including oxidizers, acids (organic and inorganic), phenols, and surfactants. Moreover, there are several factors in processing environments that can limit the efficacy of these antimicrobials, thus allowing survival and persistence. This mini-review examines the antimicrobial activity of common disinfectants/sanitizers used …

The aims of this study were to find suitable environmental enrichment (EE) and evaluate the combined effect of two EEs, variable light intensity (VL) lighting program and EH, on mental health and hepatic metabolic regulation in commercial broilers. To find the advantageous EEs for broilers, three different EEs (board, hut, and ramp) were tested in trial 1. EEs were placed and the engagement of birds to EEs, dustbathing behavior, and daily physical activity were observed. Birds treated with huts showed higher engagement than the board- or ramp-treated birds (p < 0.05). The results of dustbathing behavior and daily physical activity indicated that the environmental hut (EH) is the most favorable enrichment for broilers. In the second trial, to test the effect of EHs on mental health and hepatic metabolic conditions, the brain and liver were sampled from the four treatment birds (20 lx_Con, 20 lx_Hut, VL_Con and VL_Hut) on day 42. The lower expression of TPH2 (tryptophan hydroxylase 2) of VL_Hut birds than those of VL_Con and 20 lx_Hut treated birds suggests the combining effect of EHs with the VL lighting program on the central serotonergic homeostasis of broilers. Reduced expressions of TH (tyrosine hydroxylase), GR (glucocorticoid receptor), BDNF (brain-derived neurotrophic factor) of VL_Hut treated birds compared to those of VL_Con and 20 lx_Hut birds suggest lower stress, stress susceptibility, and chronic social stress in VL_Hut treated birds. The expression of CPT1A (carnitine palmitoyl transferase 1) increased over three-fold in the liver of VL_Con birds compared to 20 lx_Con birds (p < 0.05). EHs treatment in VL birds (VL_Hut) significantly decreased CPT1A but not in 20 lx birds (20 lx_Hut). The expression of ACCα (acetyl-CoA carboxylase alpha) was significantly decreased in VL_Con birds compared to 20 lx_Con birds. There was no significant difference in the hepatic FBPase (fructose-1,6-bisphosphatase), GR, and 11β-HSD1 (11 β-hydroxysteroid dehydrogenease-1) expression between 20 lx_Con and VL_Con birds, but EHs significantly stimulated GR in 20 lx_Hut birds, and stimulated FBPase and 11β-HSD1 expression in the VL_Hut birds compared to 20 lx_Con birds, suggesting that the VL lighting program reduced fatty acid synthesis and increased fatty acid β-oxidation in the broilers’ liver and VL_Hut improved the hepatic de novo glucose production. Taken together, the results suggest that the stimulated voluntary activity by EHs in the light-enriched broiler house improved mental health and hepatic metabolic function of broilers and may indicate that the improved hepatic metabolic function contributes to efficient nutritional support for broilers.

Water scarcity is a global reality and with the anticipated population growth, freshwater resources will be further strained to meet both human and agriculture applications. To ensure a water sustainable and food secure future, all aspects of agriculture must become more efficient. Two strategies were explored. The first strategy was to develop a more efficient and accurate method for measuring water consumption/inputs in agriculture. To date, water measuring technology has lacked the necessary sophistication to assure accuracy and repeatability of low flow water usage. The second strategy utilized the improved water measuring technology, to develop water-efficient genetics in agricultural populations …
