Choline and vitamin B12 are essential nutrients for growth and performances of production animals. However, both nutrients are extensively degraded during digestion in the rumen. This thesis comprised three experiments. First, four cows equipped with a rumen cannula and catheters in the portal vein and a mesenteric artery received a post-ruminal bolus of: 1) cyanocobalamin (CN-CBL) alone (0.1 g) [gram], 2) CN-CBL (0.1 g) + casein (10 g) or 3) CN-CBL (0.1 g) + whey proteins (10 g). Packed-bed digesters are an alternative to covered lagoon digesters for methane production and anaerobic treatment of dilute wastewaters such as dairy barn flush water. The physical media of packed-beds retain biofilms, often allowing increased treatment rates. Previous studies have evaluated several types of media for digestion of dilute wastewaters, but cost and media fouling have setback commercial development. A major operational cost has been effluent recirculation pumping.

Metabolizable protein (MP) is protein that reaches the small intestine and is available for absorption and utilization by the cow. Dairy rations may be limited in the supply of MP essential to meeting the demands of milk synthesis, however as much as half of the MP flowing to the small intestine may be attributed to microbial origins and is referred to as microbial CP (MCP). The golden Syrian hamster is the model of choice or the only rodent model for studying many human diseases. However, the lack of gene targeting tools in hamsters severely limits their use in biomedical research. Here, we report the first successful application of the CRISPR/Cas9 system to efficiently conduct gene targeting in hamsters. We designed five synthetic single-guide RNAs (sgRNAs)—three for targeting the coding sequences for different functional domains of the hamster STAT2 protein, one for KCNQ1, and one for PPP1R12C—and demonstrated that the CRISPR/Cas9 system is highly efficient in introducing site-specific mutations in hamster somatic cells. Mycobacterium avium subsp. paratuberculosis (Mpt) is the etiologic agent of Johne’s disease, a chronic intestinal disease in cattle that threatens the economic viability of dairy farming. Diagnostic culture is typically unrewarding until several years after infection when clinical signs can be observed. This leads to widespread infection within the herd. Difficulty in culturing Mpt may be a result of oxidative damage due to the increased metabolic rate when dormant organisms are recovered in a nutrient rich medium. The internal structure of casein micelles is not fully understood. In the present work, we explore some of the basic questions about casein micelles in bovine milk including its size distribution, native binding properties and effect of ultra-high pressure homogenization (ultra-HPH) on technological properties of casein micelles. The size distribution of casein micelles was studied by cryo-TEM using skim milk from four cows. The degree of variation in casein micelle sizes (polydispersity) ranged between 0.39 and 0.47 for a size distribution ranging from 10 to 693 nm. These results indicated high polydispersity of casein micelles in milk from single cows. …

Infant formula derived from bovine milk contains a number of metabolic forms of choline, all contribute to the growth and development of the newborn. Heat-induced decreases in bovine blastocyst development have been related to reductions in latent matrix metallopeptidase-9 (proMMP9) production from maturing cumulus-oocyte complexes. Elevated intrafollicular proMMP9 levels at the time of oocyte retrieval have been positively related to pregnancy following human IVF. Thus, we hypothesized that heat-induced reductions in proMMP9 levels during oocyte maturation may be responsible for decreased blastocyst development. As a first step towards testing this hypothesis, bovine cumulus-oocyte complexes were matured at 38.5°C for 24 h with 0 or 300 ng/mL recombinant human proMMP9 (rhMMP9) added at 0 h of in vitro maturation (hIVM). Eight Holstein and 8 Jersey multiparous, lactating cows were used to complete 56 energy balances to determine the energy content of reduced-fat distillers grains and solubles (RFDDGS). A repeated switchback design was used to compare treatments with and without RFDDGS. Diets consisted of 24.2 % corn silage, 18.4 % alfalfa hay, 6.94 % brome hay with either 22.9 % rolled corn and 14.8 % soybean meal (Control), or 8.95 % rolled corn, 28.8 % RFDDGS, and 0 % soybean meal (Co-P; DM basis). This study’s objectives were to determine if the biopsy of expanded blastocysts prior to cryopreservation affected the survival rates and determine if there is a difference between slow-freezing in 10% glycerol, slow freezing in ethylene glycol, and vitrification in terms of survival rate of the embryos. Blastocoeles were reduced prior to cryopreservation by means of biopsy. Embryos for slow freezing were exposed to ethylene glycol (SynGRO Ethylene Glycol with Sucrose, BIONICHE, Pullman, WA, USA) or 10% glycerol (SynGRO 10% Glycerol, BIONICHE, Pullman, WA, USA) as cryoprotectants. Dairy calves are born with a naive immune system, and must obtain passive immunity through ingestion of colostrum. Colostrum provides the newborn with antimicrobial proteins called immunoglobulins, such as immunoglobulin G (IgG), that protect the calf for the first few weeks of life. Calves that receive good quality colostrum (>50 g/L of IgG) are more likely to survive to adulthood. Niacin is a vasodilator that may increase blood flow to the mammary gland during production of colostrum in the mother. The first objective of this study was to determine the effect of bedding on the welfare of Holstein and Jersey calves housed using individual hutches bedded with gravel, rubber mats, or sand. A second objective was to determine the effects of stable and house fly populations on calf welfare. Bedding quality and fly management are aspects of improving animal welfare on dairy farms. Ghrelin is a hormone produced mainly by the cells lining the gastric mucosa. Ghrelin was first extracted from human and rat stomachs, and identified as an endogenous stimulator of growth hormone release. Ghrelin is synthesized and produced in several tissues, but the gastric mucosa remains the major source of circulating ghrelin. Sporeforming bacteria (such as Bacillus and Paenibacillus spp.) can survive pasteurization conditions (Collins, 1981) and grow in pasteurized fluid milk during refrigerated storage (Huck et al., 2008; Ivy et al., 2012), causing fluid milk spoilage and limiting the further extension of fluid milk’s shelf life (Fromm and Boor, 2004; Durak et al., 2006). Historically, the most significant losses from halogeton poisoning have been reported in sheep, with multiple catastrophic deaths documented. While recorded death losses in cattle from halogeton poisoning are less common than in sheep, recent cases, including 2 reported here, and anecdotal reports from other ranchers suggest that the impact of halogeton losses in cattle herds in the western United States is much more widespread than originally thought. Halogeton may accumulate up to 30% oxalates; a small amount of the plant (300 g) is enough to cause death in sheep. Commonly present in the muscle of animals are the tissue cysts of Toxoplasma and Sarcocystis. The aim of this study was to determine the presence and levels of Toxoplasma/Sarcocystis tissue cysts in retail pork, beef, and mutton throughout the Michiana area. A total of 36 samples, 12 from each species, were exposed to a digestive solution mimicking stomach conditions. The solution consisted of Pepsin 0.75% , NaCl 0.86%, and HCl adjusted to a pH of 1-2. The digested samples were then strained through several layers of cheesecloth, centrifuged, and examined for the presence of parasites.

Towards the goal of producing fully human polyclonal antibodies (hpAbs or hIgGs) in transchromosomic (Tc) cattle, we previously reported that Tc cattle carrying a human artificial chromosome (HAC) comprising the entire unrearranged human immunoglobulin (Ig) heavy-chain (hIGH), kappa-chain (hIGK), and lambda-chain (hIGL) germline loci produced physiological levels of hIgGs when both of the bovine immunoglobulin mu heavy-chains, bIGHM and bIGHML1, were homozygously inactivated (bIGHM-/-, bIGHML1-/-; double knockouts or DKO). This project involved an experiment conducted using a 550-ton capacity drive-over pile of whole-plant corn silage. The main effects compared were: packing the final forage surface with a loader or crawler, delay or immediate sealing, and covering with standard plastic or an oxygen barrier film.

Numerous studies have shown that the absence of oxygen in silage stored in a bunker silo or pile is crucial for proper fermentation and to ensure the highest quality silage at feed out. The objective of this study was to determine if there was a significant difference in weight gain and grain intake between Holstein calves fed milk replacer with an alternative protein source and an all-milk replacer. The study took place on a large commercial calf operation in the Central Valley of California. All calves on trial were Holstein bull calves and started the trial within three to ten days of birth. The origin of rancidity in milk is due to lipolysis caused the lipase enzyme. However, with pasteurization, the lipase enzyme is inactivated.
