In vitro produced bovine embryos are critical to the cattle industry. However, these embryos have altered morphology, epigenetics, and metabolism when compared to their in vivo counterparts. The aim of this thesis was to alter maturation and culture media to improve the developmental competence of in vitro bovine embryos. This thesis is comprised of three experiments and one proof of concept study. The purpose of this project was to create training modules for the Cal Poly Dairy. The author’s goal was to incorporate the safety aspect of dairy work into the modules as well. To properly obtain all the necessary information for the modules, the author first had to compile lists from the Cal Poly Dairy’s written standard operating procedures. From here, the majority of time was used to gather video and picture content using a surface tablet that would later be used in the online modules. The objective of this study was to measure the effects of both a nutritional (water nipple) and social (partner calf) enrichment on calf body weight, grain intake, water intake, behavior and learning. The enrichments included 1) water provided from a nipple vs. a bucket (nutritional) and 2) visual and tactile access to a partner vs. isolated rearing with no visual or tactile access to a partner calf (social). A total of 72 Holstein and Jersey dairy calves were pseudo-randomly distributed into 4 treatments at birth [Individual/Bucket (IB), Paired/Bucket (PB), Individual/Nipple (IN), Paired/Nipple (PN)]. Poor rehydration properties of skim milk powder (SMP) can impact processing efficiency and functionality in finished product applications. Rehydration can be split into four stages: wettability, sinkability, dispersibility, and solubility. Previous work has suggested that chelator addition during SMP manufacture leads to higher solubility compared to SMP without chelators. This study focuses on the addition of ethylene diamine tetraacetic acid (EDTA) or sodium citrate dihydrate (SCD) at concentrations of 5, 10, and 15 mM to skim milk prior to evaporating and spray drying. Caseinate powders have been well accepted because of their foaming properties. In this study, 10% solution of reconstituted skim milk powder (SMP) chelated with sodium hexametaphosphate (SHMP) and trisodium citrate (TSC) at 1 mEq, 50 mEq, and 100 mEq were prepared to conduct a comparison with sodium caseinate, potassium caseinate, and calcium caseinate solutions. Foamability, foam stability as well as the preferential locations of αs-casein, β-casein and !-casein in their foams were analyzed. The proteins found in milk are largely important in the functionality of many dairy products and dairy processes. The casein micelle system in milk is a complex and highly studied system. The micelle is thought to be a sponge like structure containing four caseins, αs1, αs2, β, and κ casein, and bound together with colloidal calcium phosphate. A previous study in our laboratory showed that addition of 150 mM NaCl or KCl into diafiltration water improved the solubility of freshly made milk protein concentrate 80 (MPC80). In the present study, the objectives were (1) to evaluate the solubility of NaCl- or KCl-treated MPC80 samples kept at varying temperatures and then stored for extensive periods at room temperature (21°C ± 1°C); and (2) to determine if MPC80 samples stored at different temperatures and protein conformation can be grouped or categorized together. Milk protein concentrate powders (MPC) with improved rehydration properties are often manufactured using processing steps, such as acidification and high-pressure processing, and with addition of other ingredients, such as sodium chloride, during their production. These steps are known to increase the amount of serum caseins or modify the mineral equilibrium, hence improving solubility of the retentates. The processing functionality of the micelles may be affected. Packed-bed digesters are an alternative to covered lagoon digesters for methane production and anaerobic treatment of dilute wastewaters such as dairy barn flush water. The physical media of packed-beds retain biofilms, often allowing increased treatment rates. Previous studies have evaluated several types of media for digestion of dilute wastewaters, but cost and media fouling have setback commercial development. A major operational cost has been effluent recirculation pumping.

This study’s objectives were to determine if the biopsy of expanded blastocysts prior to cryopreservation affected the survival rates and determine if there is a difference between slow-freezing in 10% glycerol, slow freezing in ethylene glycol, and vitrification in terms of survival rate of the embryos. Blastocoeles were reduced prior to cryopreservation by means of biopsy. Embryos for slow freezing were exposed to ethylene glycol (SynGRO Ethylene Glycol with Sucrose, BIONICHE, Pullman, WA, USA) or 10% glycerol (SynGRO 10% Glycerol, BIONICHE, Pullman, WA, USA) as cryoprotectants. This project involved an experiment conducted using a 550-ton capacity drive-over pile of whole-plant corn silage. The main effects compared were: packing the final forage surface with a loader or crawler, delay or immediate sealing, and covering with standard plastic or an oxygen barrier film.

Numerous studies have shown that the absence of oxygen in silage stored in a bunker silo or pile is crucial for proper fermentation and to ensure the highest quality silage at feed out. The objective of this study was to determine if there was a significant difference in weight gain and grain intake between Holstein calves fed milk replacer with an alternative protein source and an all-milk replacer. The study took place on a large commercial calf operation in the Central Valley of California. All calves on trial were Holstein bull calves and started the trial within three to ten days of birth. Due to the growing concern of raw milk, there is a growing need for other methods of preparing rancid milk for milk judging. The origin of rancidity in milk is due to lipolysis caused the lipase enzyme. However, with pasteurization, the lipase enzyme is inactivated. Heat stress has a major effect on management decisions for a dairy operation because of the decrease in the cows productivity. Dairy cattle loose efficiency of economic production due to lower milk yields and decreased pregnancy rates. Milk loss due to heat stressed cows in California averages a loss of 293 kg of milk/cow/year(St.-Pierre et al., 2003), and with milk prices reaching record highs, dairies are losing out on increased revenue opportunity.
