This thesis describes the development of an integrated hydrogeologic/hydrologic site assessment and groundwater/surface water quality monitoring program at the University of Tennessee – Little River Dairy Farm, located near Townsend, TN. Hydrologic/hydrogeologic investigations of streams and groundwater at the site have been underway for more than 5 years, and these are expected to provide background data for assessing impacts of dairy wastes. The lower half of the ~180 ha site consists of low-relief fields used for row crops, which are underlain by 4 – 9 m of alluvial deposits on top of black shale or limestone that include sinkhole features. …

Mastitis is an economically distressing disease in the dairy industry. Bacterial pathogens enter the gland and encounter cell types that release immune mediators including interleukin (IL)-8. IL-8 has two membrane bound receptors: CXCR1 and CXCR2. CXCR1 and CXCR2 are expressed on neutrophils and other cell types in other species but their expression is unknown in the bovine mammary gland. To test this, mammary tissue samples from six Holstein dairy cows were subject to dual immunofluorescence with bovine specific CXCR1 and CXCR2 antibodies and cell type markers. CXCR1 was expressed on alveolar epithelial cells, fibroblasts, and leukocytes. CXCR2 expression was identified …

This project was funded by the United States of Department of Agriculture (USDA), which funds research aimed at improving production and animal health. The aim of this study was to advance knowledge of maternal immune tolerance to the fetus and mechanisms bovine non-classical MHC class I proteins employ to interact with immune cells and render them inert towards the fetus.

Milk fat is encapsulated in a milk fat globule membrane (MFGM) that contains bioactive glycoproteins and glycolipids. The MFGM inhibits infectivity of rotavirus (RV), activity that has been attributed to its glycoprotein and carbohydrate components. However, previous studies of proteins and oligosaccharides in the MGFM have not accounted for all the bioactivity associated with the complete MFGM. The lipid fraction of the MFGM accounts for half of its composition by weight, and we postulate that this fraction should be tested by itself to determine if it plays a role in antiviral activity. Alexandre Family EcoDairy Farms (AFEF) is a family dairy farm, producing organic milk in northern California. In the last decade, AFEF has been expanding in value added natural food enterprises, including pastured free-range eggs, pastured pork and grass-fed beef.

This study focuses on the analysis of an organic grass-fed ground beef from dairy-beef cross steers coming from AFEF. The grass fed beef enterprise was analyzed by adopting a UC Davis cost and returns study, modifying it to AFEF production limitations and conditions. The typical Western dietary pattern is characterized by the consumption of energy-dense, nutrient-poor foods and has been linked to increased risk of colorectal cancer (CRC). Our research group previously developed the total Western diet (TWD) that emulates typical human dietary intakes of macro- (carbohydrates, proteins, and fats) and micronutrients (vitamins and minerals) on an energy density basis for rodents. Single nucleotide polymorphisms (SNPs) have great power in resolving questions of unknown parentage on ranches or dairies where errors could have been made in pedigree record keeping or when such records were simply not kept. Currently a panel of 88 SNPs based on a panel originally created by USDA-MARC is commercially available from the company Fluidigm®. Mastitis is an inflammation of the mammary gland that costs the United States dairy industry between $1.7 and $2 billion annually [9]. Approximately 97% of mastitis infections are caused by bacteria, which can be treated with antibiotics [2]. However, mastitis infections can be caused by the alga, Prototheca. The technological properties of milk have significant importance for the dairy industry. Citrate, a normal constituent of milk, forms one of the main buffer systems that regulate the equilibrium between Ca2+ and H+ ions. Higher-than-normal citrate content is associated with poor coagulation properties of milk. To identify the genes responsible for the variation of citrate content in milk in dairy cattle, the metabolic steps involved in citrate and fatty acid synthesis pathways in ruminant mammary tissue using RNA sequencing were studied. Milk quality criteria and limits for somatic cell count (SCC) established in many countries make it difficult to maintain SCC of bulk tank goat milk below the threshold, due to non-infection factors linked to goat physiology. The aim of this study was to objectively verify the effects of SCC on fresh caprine milk cheese. Somatic cells were recovered from pooled healthy goat milk, and added to low SCC raw or pasteurised goat milk. Miniature cheeses were made and evaluated after 1 and 7 d. The objective of this study was to utilize electronically recorded data from on-farm milk recording systems and herd management software to determine if a sire has a genetic influence on his daughter’s milking duration. Measurements of milking duration from three dairy facilities were taken between January 27, 2013 and January 31, 2013. These measurements supplied 25,070 observations, corresponding to 9,418 cows and 895 sires. These data were evaluated with a SAS alongside a herd-mate comparison. The objective of this study was to determine any trends in odor evaluations and gas emissions of manure samples in heifers fed high/low concentrate and high/low forage diets. Heifers (n = 8) were enrolled randomly in a split plot design with diet type (high concentrate vs. low concentrate) as the whole plot and forage quality (4 different levels of corn stover within each diet type) as sub-plots in a 4-period (21d) 4X4 Latin square design. The objective of this experiment is to determine if holstein steers can be raised for a high quality, profitable beef product. The data collected came from Dyt Dairy and their steer program. Records came from the Souza Family, who does the calf raising for Dyt Dairy, and were compiled into a spread sheet. Daily costs incurred in raising a steer for the first five months were incorporated and analyzed relative to different rates of gain. The objective of this study was to determine if fresh frozen, pasteurized frozen, or fresh refrigerated colostrum impacted morbidity, growth rates, and the overall serum immunoglobulins (IgGs) in newborn calves. The second objective was to compare the effects that pasteurization and freezing had on colostrum IgG concentration and total bacteria counts compared to raw refrigerated colostrum. A controlled trial was completed on a 6,000-cow dairy in California where colostrum was harvested twice daily, pooled, and then divided into three different treatment groups and processed accordingly. The COA, required by law, prevents customers from questioning whether a product is safe and ensures manufacturers confirm every product tested passes the Food and Drug Administration’s required specifications.
