The global human population is rapidly increasing, escalating interactions of people, animals and the environment. This has led to more convoluted health challenges, for which African smallholder farmers bear the brunt. The One Health (OH) approach recognises the interconnectedness of these health challenges and thus follows a transdisciplinary approach involving diverse stakeholders to address them. Although there have recently been global concerted efforts and studies on OH, an information gap exists in Africa, particularly regarding smallholder farmers and their needs. The replacement of soybean meal (SBM) with insect meal, e.g. black soldier fly larvae meal (BSFLM), as an alternative protein source in livestock feed is gaining traction globally. To date, no in vivo studies have been conducted to test the impact of BSFLM on the milk production of dairy cows. This study quantified the effects of replacing SBM with defatted black soldier fly larvae meal (DBSFLM) in diets of lactating dairy cows on feed intake, milk production and composition, apparent total-tract digestibility, feed- and nitrogen use efficiency. Intranasal instillation of SE36, a malaria vaccine candidate antigen, in lactating BALB/c strain (derived from the Bagg and albino laboratory inbred mice) female mice resulted in the appearance of the antigen in breast milk as demonstrated by sandwich ELISA and Western blot. Pups born of immunologically naive mice and breastfed on lactating foster mothers exposed intranasally to SE36 developed IgG anti-SE36 antibodies. These data demonstrate that maternal immunization in mice by this route in lactating mothers can result in active immunization of offspring via ingestion of breast milk containing antigen. Background: Tsetse flies (Glossina sp.) are the vectors of human and animal trypanosomiasis throughout sub-Saharan Africa. Tsetse flies are distinguished from other Diptera by unique adaptations, including lactation and the birthing of live young (obligate viviparity), a vertebrate blood-specific diet by both sexes, and obligate bacterial symbiosis. This work describes the comparative analysis of six Glossina genomes representing three sub-genera: Morsitans (G. morsitans morsitans, G. pallidipes, G. austeni), Palpalis (G. palpalis, G. fuscipes), and Fusca (G. brevipalpis) which represent different habitats, host preferences, and vectorial capacity. Tsetse flies undergo drastic fluctuations in their water content throughout their adult life history due to events such as blood feeding, dehydration and lactation, an essential feature of the viviparous reproductive biology of tsetse. Aquaporins (AQPs) are transmembrane proteins that allow water and other solutes to permeate through cellular membranes. Here we identify tsetse aquaporin (AQP) genes, examine their expression patterns under different physiological conditions (blood feeding, lactation and stress response) and perform functional analysis of three specific genes utilizing RNA interference (RNAi) gene silencing. Milk glands, dissected out and collected from Glossina morsitans centralis (Machado) females, artificially inoculated at the third-instar larval stage with a virus suspension obtained from hypertrophied salivary glands of wild-caught virus-infected Glossina pallidipes (Austen), were processed for routine electron microscopy and examined for pathological changes. They were compared to milk glands dissected out from normal female G.m. centralis at the same stage of pregnancy cycle. Upon dissection there were notable physical differences between control and virus-infected milk glands. 1. 1. The major protein in the milk gland secretions of the tsetse fly, Glossina morsitans morsitans, was isolated by a combination of gel permeation chromatography and crystallization. 2. 2. It has a native Mr ∼47,000 and is composed of two identical polypeptide chains (Mr ∼21,000) as determined by chemical cross-linking studies. The protein has no covalently-bound carbohydrates or lipids. Amino acid analysis of the protein revealed relatively high amounts of the aromatic amino acids, tyrosine (9.1 mol.%) and phenylalanine (8.5 mol.%).
