Bovine respiratory disease (BRD) costs the cattle industry millions of dollars in costs in treatment and loss every year in the United States. A significant pathogen often contributes to BRD is Bovine Herpesvirus 1 (BoHV-1), a double stranded DNA virus with the ability to establish latency in the trigeminal ganglia and neurons. Primary infection with BoHV-1 results in immunosuppression that increases the risk of secondary bacterial infection and pneumonia. Because herpesviruses infect their hosts for life and can be reactivated in times of stress, BoHV-1 can present a recurring risk of BRD. The following research aims to expand the knowledge …

Reproductive function is crucial to the success of a beef or dairy herd. Both the female and male play significant roles in the outcome of bovine reproduction. The bull contributes to reproductive success in many ways, such as genetics. The initial role that the bull has in reproductive success is fertilization, whether natural or by artificial insemination. One method of assessing bull performance are central bull test stations, which are traditionally administered by a university, extension center, or are privately owned. Embryos resulting from assisted reproductive technologies, such as in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI), develop with lower efficiencies than embryos resulting from their in vivo counterparts. The reasons behind the developmental discrepancies remain largely unknown. Because the egg is the primary determinant of embryo developmental success, it is reasonable to consider inherent egg quality as a possible cause. The hypothesis for this project is that there are distinct mRNA transcript patterns, or molecular “fingerprints,” that distinguish high- versus low-quality eggs developing within the same environment. Bovine mastitis, caused by a wide array of pathogens, results in a substantial economic loss for the dairy cattle livestock industry. β-defensins are a part of the bovine’s innate immune system and act as the first line of defense against mastitis. Only foundational research has been done on β-defensins’ ability to treat and prevent mastitis. Alternatives to routine antibiotic treatment of dairy cattle during the dry period before their next calving are of interest. This was a preliminary study of whether intramammary infusion of casein hydrolysate, administered alone or combined with standard dry treatment, accelerated the rate of mammary involution early in the dry period. Four treatments were studied in a split udder design. One udder half was assigned a treatment, and the contralateral half was administered dry cow treatment + internal teat sealant as a control. Dairy calf immune development leaves calves susceptible to illness during the pre-weaned stage of life. Previous studies have shown that the use of probiotics during this time may result in increased growth rates and decreased incidence of illness, particularly diarrhea and scours. We hypothesized that by providing probiotics during the time from birth until weaning, calves on study would experience similar growth and health benefits. Neonatal calves (N=32) were provided milk replacer three times daily, with half of the calves receiving Probios® supplements mixed in with their first morning feeding. In vitro produced bovine embryos are critical to the cattle industry. However, these embryos have altered morphology, epigenetics, and metabolism when compared to their in vivo counterparts. The aim of this thesis was to alter maturation and culture media to improve the developmental competence of in vitro bovine embryos. Prevalence of Bovine viral diarrhea virus (BVDV) (“detected” test results) among all bovine samples tested at the Utah Veterinary Diagnostic Laboratory from 2008 - 2013 was calculated, and results were compared by age, sex, or breed of the cattle and BVDV diagnostic test methods. Necropsies were tested for BVDV when lesions suggestive of infection were identified. Adults, juveniles and most calves were tested by antigen (Ag) capture ELISA, while fetuses and some calves were tested by real-time reverse transcriptase PCR. This project was funded by the United States of Department of Agriculture (USDA), which funds research aimed at improving production and animal health. The aim of this study was to advance knowledge of maternal immune tolerance to the fetus and mechanisms bovine non-classical MHC class I proteins employ to interact with immune cells and render them inert towards the fetus.

Single nucleotide polymorphisms (SNPs) have great power in resolving questions of unknown parentage on ranches or dairies where errors could have been made in pedigree record keeping or when such records were simply not kept. Currently a panel of 88 SNPs based on a panel originally created by USDA-MARC is commercially available from the company Fluidigm®. Advances in in vitro fertilization and in vitro culture techniques have allowed considerable progress in identifying physiological requirements of mammalian embryos. Parrish et al. (11) reported a major breakthrough on in vitro fertilization when his group identified heparin as an important factor for the capacitation of spermatozoa.
