The objective of this experiment was to investigate the effect of restricting pasture access time on milk production and composition, body weight and body condition score change, dry matter intake, and grazing behavior of autumn calving dairy cows in midlactation. Fifty-two (19 primiparous and 33 multiparous) Holstein-Friesian dairy cows (mean calving date, August 17 ± 91.2 d) were randomly assigned to a 4-treatment (n = 13) randomized block design grazing study. Destruction of the architectural and subsequently the functional integrity of the lung following pulmonary viral infections is attributable to both the extent of pathogen replication and to the host-generated inflammation associated with the recruitment of immune responses. The presence of antigenically disparate pulmonary viruses and the emergence of novel viruses assures the recurrence of lung damage with infection and resolution of each primary viral infection. Thus, there is a need to develop safe broad spectrum immunoprophylactic strategies capable of enhancing protective immune responses in the lung but which limits immune-mediated lung damage. Background Successful reprogramming of a somatic genome to produce a healthy clone by somatic cells nuclear transfer (SCNT) is a rare event and the mechanisms involved in this process are poorly defined. When serial or successive rounds of cloning are performed, blastocyst and full term development rates decline even further with the increasing rounds of cloning. Identifying the "cumulative errors" could reveal the epigenetic reprogramming blocks in animal cloning. Results Bovine clones from up to four generations of successive cloning were produced by chromatin transfer (CT). Severe acute respiratory syndrome (SARS) is a highly lethal emerging disease caused by coronavirus SARS-CoV. New lethal animal models for SARS were needed to facilitate antiviral research. We adapted and characterized a new strain of SARS-CoV (strain v2163) that was highly lethal in 5–6 week old BALB/c mice. It had nine mutations affecting 10 amino acid residues. Strain v2163 increased IL-1α, IL-6, MIP-1α, MCP-1, and RANTES in mice, and high IL-6 expression correlated with mortality. The infection largely mimicked human disease, but lung pathology lacked hyaline membrane formation. In vitro efficacy against v2163 was shown with known inhihibitors of SARSCoV …

Influenza A viruses causes a variety of illnesses in humans. The most common infection, seasonal influenza, is usually a mild, self-limited febrile syndrome, but it can be more severe in infants, the elderly, and immunodeficient persons, in whom it can progress to severe viral pneumonitis or be complicated by bacterial superinfection, leading to pneumonia and sepsis. Seasonal influenza also occasionally results in neurologic complications. Rarely, viruses that have spread from wild birds to domestic poultry can infect humans; such “avian influenza” can range in severity from mild conjunctivitis through the rapidly lethal disease seen in persons infected with the H5N1 …

The ability to rescue an infectious, recombinant, negative-stranded, RNA virus from a cDNA clone, has led to new opportunities for measuring viral replication from a viral expressed reporter gene. In this study, the enhanced green fluorescent protein (EGFP) gene was inserted into the human parainfluenza virus type 3 (HPIV-3) antigenome and a recombinant, infectious virus was rescued. Maximum EGFP expression levels, measured by fluorescence, were seen at day 3. Comparison of a three-day, viral expressed EGFP fluorescence assay to a seven-day, neutral red assay, based on complete cell destruction in virus infected MA-104 cells, yielded Z′-factor values of 0.83 and …

For the first part of my thesis, the effects of filtration temperature and heat treatment on the compositional properties of whole milk Ultrafiltration retentate (UF) were studied. Ultrafiltration is primarily run at temperatures in the range of 50-55°C but more and more plants are starting to filter at refrigeration temperatures. This article analyzes ongoing controversy over how to best label rBST-free milk. Recombinant bovine somatotropin is a genetically engineered drug administered by some farmers to their dairy herds to increase milk production. FDA first approved its use in 1994, despite great controversy. The FDA also issued labeling guidelines that allowed voluntary disclosure of rBST-free milk, so long as it carried the disclaimer that no difference could be detected between milk produced with rBST and rBST-free. The controversy continues today as consumers express a preference for rBST-free milk and many rBST-free producers label their milk this way. "Conventional" milk (with rBST) …

Conventional agriculture acidifies soil. As your soil becomes more acid, productive pasture species disappear and are replaced by species of low agricultural value. It is a waste of money to apply expensive fertiliser to pastures which are inherently poorly productive because they are growing in an acid soil.

A heifer feeding trial evaluated the impact of high/low forage quality and high/low concentrate level nutrient-balanced diets on simultaneous odor and gas emissions from the manure. Gas concentration was determined using an infrared photoacoustic analyzer over a 24-hour period using a steady-state flux chamber setup containing urine:feces as-excreted from eight individual heifers. Odorous air samples were collected from chamber headspace and evaluated by six human assessors for pleasantness, intensity and detection threshold using a forced-choice dynamic olfactometer. Ammonia emission ranged from 0.64 to 3.94 mg NH3 cm-2 d-1 across diets. Average ammonia emission from the low concentrate (80% forage) diets …

The objective was to conduct a broad survey of dairy management practices that have an effect on animal well-being. Dairies were visited during the fall and winter of 2005 and 2006 in Wisconsin, Minnesota, Indiana, Iowa, and New York. Data were collected on 113 dairies on colostrum feeding, dehorning, tail-docking, euthanasia methods, producer statements about welfare, use of specialized calf-raising farms (custom), level of satisfaction with calf-raising by producers, and cow behavior. Calves were raised by the owner on 50.4% of dairies; 30.1% were raised on custom farms during the milk-feeding period, 18.6% were custom raised after weaning, and 1% …

This study was conducted in nine villages near Mount Meru, Tanzania between 14 April 2008 and 1 May 2008. The study question and methods were developed in conjunction with Heifer Tanzania. Heifer is a non-profit organization that provides dairy-cattle and other livestock to farmers in need. Heifer’s agents also provide training in sustainable farming techniques, such as zero-grazing livestock husbandry, and building contours. Some farmers receive this training in their village from a project supervisor, and other farmers are sent to a government facility, Liti Tengeru Animal Husbandry Training Center. Phenothiazine and derivatives were tested for inhibition of SARS-CoV replication. Phenothiazine slightly inhibited SARS-CoV replication in a neutral red (NR) uptake assay. Adding a propylamino group to give promazine reduced virus yields (VYR assay) with an EC90 = 8.3 ± 2.8 μM, but without selectivity. Various substitutions in the basic phenothiazine structure did not promote efficacy. Phenazine ethosulfate was the most potent compound by VYR assay (EC90 = 6.1 ± 4.3 μM). All compounds were toxic (IC50 = 6.6–74.5 μM) except for phenoxathiin (IC50 = 858 ± 208 μM) and 10-(alpha-diethylamino-propionyl) phenothiazine·HCl (IC50 = 195 ± 71.2 μM). Consequently, none …

Forty-eight calves (heifers n=20, bulls n=28) were randomly assigned one of four dietary treatments which included no additive (C); the yeast culture Saccharomyces cerevisiae (YC); the probiotics Bacillus lichenformis and Bacillus subtilis (P); and both yeast culture and probiotics (YCP). Calves were administered treatments from day 2 to 56 in starter and from day 57 to 84 in grower diets. Body weights were measured at birth and weekly thereafter until day 112 of age. The primary targets for SARS-CoV infection are the epithelial cells in the respiratory and intestinal tract. The angiotensin-converting enzyme 2 (ACE-2) has been identified as a functional receptor for SARS-CoV. ACE-2 has been shown to be expressed at the apical domain of polarized Calu-3 cells. In this report, interferon alfacon 1 was examined for inhibitory activities against SARS-CoV on human lung carcinoma epithelial Calu-3 cell line and the other three African green monkey kidney epithelial cell lines. Interferon alfacon 1 demonstrated significant antiviral activity in neutral red uptake assay and virus yield reduction assay. The data might provide an important …
