The development of reliable hydrogel-based scaffolds for extrusion bioprinting remains limited by the poor structural fidelity of low-viscosity bioinks and the lack of robust, high-throughput quality evaluation methods. This dissertation can resolve such challenges by developing the combination of optimized hydrogel formulations, cryogenic-assisted bioprinting, and artificial intelligence (AI)-based scaffold evaluation to the use of tissue engineering and preclinical cancer modelling applications. To assess the rheological behavior, printability, mechanical properties, and biocompatibility of the alginate – gelatin (Alg–Gel) system, a novel system of hydrogel was developed and characterized using Alg–Gel hydrogel system. Quantifying human arm muscle activity is essential for understanding human movements and helping individuals who suffer from motor or neurological impairments such as stroke or traumatic brain injuries. Today, these individuals, as well as clinicians, researchers, and athletes, face many challenges when using the traditional surface electromyography (sEMG) systems to monitor muscle activities, including complex setup, restricted mobility, discomfort, inconsistent placement of electrodes, and lack of accuracy. After suffering a stroke, many survivors experience physical disability that affects their ability to complete daily tasks using their hands. Physical therapy is often pre scribed to people post-stroke, but is not fully effective at restoring hand function. The goal of this project was to design and fabricate an assistive device for people post-stroke to help with grasp and release. Deep-sea fishing presents significant challenges for individuals with upper-limb differences, particularly when using prosthetic devices that are not designed for recreational fishing activities. This project developed a custom prosthetic fishing rod attachment for a user who had lost her forearm and desired a safe, comfortable, and independent method of fishing. Through interviews with the sponsor and an iterative design process, a lightweight aluminum attachment was created to interface directly with the user's existing prosthetic quick-connect system.
The design process included concept generation, CAD modeling, rapid prototyping with 3D-printed models, manufacturing, and user-centered refinement. The final design consists of a welded …

Endovascular procedures require devices with widely varying mechanical properties: flexibility for navigating tortuous vessels and rigidity for stable therapeutic delivery. Suction-actuated variable stiffness sheaths address this challenge by incorporating axial wire string arrays that mechanically couple under suction to increase flexural rigidity. However, prototype devices achieve stiffening ratios below theoretical predictions, suggesting that interlayer mechanics and string positioning play a critical role. This study investigates whether interweaving polytetrafluoroethylene (PTFE) tape within the string array enhances flexural rigidity modulation and curvature stability. Prototype devices with varying wrap configurations were fabricated and evaluated using flexural testing, curvature stability testing, and mathematical modeling. …

Hybrid structures formed between single-stranded DNA (ssDNA) and single-walled carbon nanotubes (SWCNT) have emerged as promising components in future direct DNA sequencing technologies due to their unique morphological and electronic properties. The nanoscale configuration of these hybrids, including the DNA wrapping angle, pitch, and contour behavior, directly influences their potential for stable electronic readout. This thesis presents an experimental study of DNA-SWCNT hybrid morphology using a fixed, reproducible wrapping protocol designed to minimize environmental variability and isolate the influence of DNA nucleotide sequence on hybrid structure. As described in prior work, this protocol produces well-dispersed hybrids suitable for high-resolution imaging. …

Hamstring strain injuries are among the most common and recurring injuries in sports, occurring most often during the terminal swing phase of gait. Despite progress in rehabilitation, the incidence of injury has only increased, suggesting a need for a novel intervention. We designed a passive, assistive, wearable device with two specific criteria in mind: to offload the hamstrings during running, and do so without hindering normal kinematic patterns. The device consists of an adjustable elastic band anchored proximally at the waist and distally just above the ankle joint center. Persistent gait asymmetry following anterior cruciate ligament reconstruction (ACLR) is a recognized contributor to post-traumatic knee osteoarthritis, yet the independent role of pain in driving altered movement patterns remains poorly understood due to confounding factors inherent to clinical populations. This study used exercise-induced muscle damage (EIMD) as a controlled experimental model to isolate the effects of musculoskeletal pain on gait, independent of kinesiophobia, structural damage, and proprioceptive disruption. Eighteen healthy adults underwent unilateral eccentric calf-raise protocols to induce delayed onset muscle soreness (DOMS), with gait assessed at baseline, 48 hours, and 168 hours post-exercise using bilateral inertial measurement units. Contemporary research suggests that shear force is a major contributor to discomfort and pressure injury. However, the variation of shear forces during human-exoskeleton interaction dynamics is a highly unexplored field. The high cost of commercial triaxial force sensors may be a major factor in the notable lack of research in this field. Therefore, in this paper, we present a low-cost, 3D-printed triaxial force sensor designed specifically to measure the triaxial interaction forces between an exoskeleton and its user. Anthropometric test devices (ATDs) are human surrogate models used to represent the human body response in the context of compliance and safety testing. These devices are used in multiple industries, including automotive and aerospace, and have recently been incorporated into impact testing for the evaluation of helmet performance in American football. Arteriovenous (AV) grafts are commonly used to provide vascular access for hemodialysis in patients with end-stage renal disease. Despite their widespread use, AV grafts are prone to complications such as stenosis and thrombosis. Early detection of these conditions remains challenging with current monitoring methods too costly or insufficient. This work presents the design, fabrication, and validation of an LC pressure sensor embedded within a model AV graft to enable real-time monitoring. Mechanical loading is known to regulate bone remodeling by driving interstitial fluid flow which stimulates cells and drives nutrient transport within the trabecular network. In microgravity, the absence of mechanical stimulation or loading suppresses convective flow processes, causing diffusion-driven nutrient mixing and renewal, and accelerated bone loss. This thesis investigates how oscillation frequency and trabecular bone density jointly control nutrient mixing and wall shear stress within trabecular cavities.

American football players are repeatedly subject to high-velocity impacts; collisions can exceed 100 g in the NFL [1]. Rotational accelerations of the head caused by high-velocity collisions have been proven to be extremely damaging to the highly organized brain fibers at the brain-skull interface [2]. The sensor system (SS) measures the nonplanar acceleration at four locations on a chin strap using ADXL314 accelerometers and utilizes rigid-body kinematics to estimate the head rotational state from the measured acceleration of the SS [3]. Effective carbon dioxide (CO₂) storage is essential for mitigating climate change amid increasing global greenhouse gas emissions. This study investigates the influence of geomechanics on CO₂ storage performance within carbon capture and storage (CCS), focusing on structural, residual, and solubility trapping mechanisms using a fully coupled modeling framework. Two numerical models, with and without geomechanical effects, are developed to evaluate impacts on reservoir behavior, CO₂ migration, and trapping efficiency. Each mechanism is analyzed separately and within an integrated framework to assess their combined contributions. Background: Microgravity exposure alters cardiovascular loading, yet its impact on left atrial flow dynamics and thrombotic risk remains poorly understood. This study investigates how spaceflight-relevant microgravity-induced changes in cardiac outflow affect left atrial hemodynamics in healthy individuals and patients with atrial fibrillation.
Methods: Patient-specific left atrial models were generated for three healthy individuals and three AF patients. Anterior knee pain affects approximately 25% of adults and is associated with altered joint mechanics. Patellofemoral joint (PFJ) models vary widely in complexity, with no established gold standard. Musculoskeletal models efficiently estimate joint loading but do not capture tissue-level mechanics, whereas finite element (FE) models provide tissue-level predictions at greater computational cost. This thesis developed a workflow to evaluate relationships between musculoskeletal model-predicted PFJ loads and FE-predicted cartilage mechanics, examined the impact of altered quadriceps activation, and introduced improved computational models.

Split-thickness skin grafts are widely used to treat chronic wounds. Procedure design requires surgeons to predict how much a patch of the patient's own skin expands when it is meshed with rows of slits and stretched over a larger wound area. Accurate prediction of graft expansion remains a challenge, with current models overestimating the actual expansion, leading to suboptimal outcomes. Calcium phosphate cements (CPCs) and biomaterials, such as mesoporous bioactive glass (MBG), are critical for bone tissue engineering. This study aimed to 3D-print CPC scaffolds modified with MBG to enhance their osteogenic potential and regenerative ability. MBG powder was synthesized and characterized using transmission electron microscopy (TEM), X-ray diffraction (XRD), and nitrogen adsorption-desorption techniques. A commercial CPC ink (hydroxyapatite/alpha-tricalcium phosphate) was mixed with 5% MBG (w/w; CPC/MBG), and, after rheological assessment, the mixture was used to obtain scaffolds via 3D printing. Injured ligaments are often repaired with tendon grafts which may over or under constrain the joint; there is a need for an artificial graft option with the mechanical properties of a native ligament. Crimped fibers within ligaments allow for recruitment which means the viscoelastic properties of stress relaxation and creep are not inversely related. A feasibility study using ACLs and large-scale crimp models to study viscoelasticity, led to an in-depth analysis of the scapholunate interosseous ligament.
