Integrated regional water management (IRWM) is an approach to water resource management in California that is being strongly promoted by the State as a way to increase regional self-sufficiency. IRWM offers an approach for managing the uncertainties that lie ahead, particularly in light of climate change. The IRWM planning process brings together water and natural resource managers, along with other community stakeholders, to collaboratively plan for and ensure the region’s continued water supply reliability, improved water quality, flood management, and healthy functioning ecosystems—allowing for creative new solutions and greater efficiencies. Climate change is a global concern as it may affect many aspects of life, including water supply. A tool used to model climate change’s impacts is called a General Circulation Model (GCM). GCMs project future scenarios including temperature and precipitation, but these are designed at a coarse resolution and require downscaling for employment for regional hydrologic modeling. There is a vast amount of research on downscaling and bias-correcting GCMs data, but it is unknown whether these techniques alter precipitation signals embedded in these models or reproduce climate states that are viable for water resource planning and management. Centralized communication-based control is one of the main methods that can be implemented to achieve autonomous advanced energy management capabilities in DC microgrids. However, its major limitation is the fact that communication bandwidth and computation resources are limited in practical applications. This can be often improved by avoiding redundant communications and complex computations. In this paper, an autonomous communication-based hybrid state/event driven control scheme is proposed. Electrodes implanted in the brain or spinal cord trigger the activation of resident astrocytes. In their reactive state, astrocytes surrounding the electrode form a glial scar, compromising the ability of the electrode to interface with the surrounding neural tissue. One approach to reduce the inhibiting scar tissue is to incorporate nanoarchitecture on the surface of the implanted materials to modify the astrocytic response. The incorporated nanoarchitecture changes both the surface characteristics and the material properties of the implant interface. Targeted photopolymerization is the basis for multiple diagnostic and cell encapsulation technologies. While eosin is used in conjunction with tertiary amines as a water-soluble photoinitiation system, eosin is not widely sold as a conjugate with antibodies and other targeting biomolecules. Here we evaluate the utility of fluorescein-labeled bioconjugates to photopolymerize targeted coatings on live cells. We show that although fluorescein conjugates absorb approximately 50% less light energy than eosin in matched photopolymerization experiments using a 530 nm LED lamp, appreciable polymer thicknesses can still be formed in cell compatible environments with fluorescein photosensitization. Conventional petroleum-derived plastics are recalcitrant to biodegradation and can be problematic as they accumulate in the environment. In contrast, it may be possible to add novel, biodegradable bioplastics to anaerobic digesters at municipal water resource recovery facilities along with primary sludge to produce more biomethane. In this study, thermal and chemical bioplastic pretreatments were first investigated to increase the rate and extent of anaerobic digestion. Subsequently, replicate, bench-scale anaerobic co-digesters fed synthetic primary sludge with and without PHB bioplastic were maintained for over 170 days. Two polyhydroxybutyrate (PHB), one poly(3-hydroxybutyrate-co-4-hydroxybutyrate) and one polylactic acid (PLA) bioplastic were investigated. Biochemical methane …

The ink-jet technique was developed to print the wedge polymer microdisk lasers. The characterization of these lasers was implemented using a free-space optics measurement setup. It was found that disks of larger edge inclination angles have a larger free spectral range (FSR) and a lower resonance wavelength difference between the fundamental transverse electric (TE) and transverse magnetic (TM) whispering-gallery modes (WGMs). This behavior was also confirmed with simulations based on the modified Oxborrow’s model with perfectly matched layers (PMLs), which was adopted to accurately calculate the eigenfrequencies, electric field distributions, and quality parameters of modes in the axisymmetric microdisk resonators. …

Low-fidelity aerostructural optimization routines have often focused on determining the optimal spanloads for a given wing configuration. Several analytical approaches have been developed that can predict optimal lift distributions on rectangular wings with a specific payload distribution. However, when applied to wings of arbitrary geometry and payload distribution, these approaches fail. Increasing the utility and accuracy of these analytical methods can result in important benefits during later design phases. The broad goal of the research described in this thesis was to better understand the potential for biological toluene production to occur in groundwater at a Superfund Site located in southeast Louisiana. Previous literature reported that bacterial isolates under laboratory test conditions and undefined microbial communities in anaerobic sludge digesters and seasonally stratified lakes can produce toluene biogenically. Most of the research reported on this subject previously, however, has been on systems in Europe and little research has been reported previously on such phenomena in North America.

The recent Mirai botnet attack demonstrated the danger of using default passwords and showed it is still a major problem. In this study we investigated several common applications and their password policies. Specifically, we analyzed if these applications: (1) have default passwords or (2) allow the user to set a weak password (i.e., they do not properly enforce a password policy). Our study shows that default passwords are still a significant problem: 61% of applications inspected initially used a default or blank password. When changing the password, 58% allowed a blank password, 35% allowed a weak password of 1 character.

The objective of this study was to examine three well designs: drilled wells (20–30 m deep), closed dug wells (>5 m deep), and hand-dug open wells (<5 m deep), to determine the water quality for improving access to safe and clean water in rural communities. Heterotrophic plate count (HPC), total coliforms (TC), Escherichia coli (E. coli) and turbidity, were used to assess the water quality of 97 wells. Additionally, the study looked at the microflora diversity of the water, focusing on potential pathogens using outgrowth, PCR, and genome sequencing for 10 wells. Concentrations of TC for the open dug wells (4 × 104 CFU/100 mL) were higher than the drilled (2 × 103 CFU/100 mL) and closed dug wells (3 × 103 CFU/100 mL). E. coli concentration for drilled and closed dug wells was <22 MPN (most probable number)/100 mL, but higher for open wells (>154 MPN/100 mL). The drilled well turbidity (11 NTU) was within the standard deviation of the closed well (28 NTU) compared to open dug wells (49 NTU). Drilled and closed wells had similar microbial diversity. The isotropic average conduction band minimum electron effective mass in Sn-doped monoclinic single crystal B-Ga2O3 is experimentally determined by the mid-infrared optical Hall effect to be (0.28460.013)m0 combining investigations on (010) and (_201) surface cuts. This result falls within the broad range of values predicted by theoretical calculations for undoped B-Ga2O3. The result is also comparable to recent density functional calculations using the Gaussian-attenuation- Perdew-Burke-Ernzerhof hybrid density functional, which predict an average effective mass of 0.267m0. This research investigated the behavior of a Manduca sexta inspired biomimetic wing as a function of Reynolds number by measuring the aerodynamic forces produced by varying the characteristic wing length and testing at air densities from atmospheric to near vacuum. A six degree of freedom balance was used to measure forces and moments, while high speed cameras were used to measure wing stroke angle. The complexity of biological soft matter at the sub-micrometer level is fundamentally correlated to the functionalities at the larger scale. Reflecting the level of heterogeneities in the properties of systems remains challenging when probing small scales, due to the mismatch between the area surveyed with the tools offering nanoscale resolution, such as atomic force microscopy (AFM), and the scale of natural variations inherent to biology. Hence, to understand the physiological and mechanical alterations that occur within a single cell relative to a cell population, a multiscale approach is necessary. Polyelectrolyte based hydrogel fibers can mimic extracellular matrix and have applications such as drug delivery and tissue scaffolding. Metal ions play a critical role in hydrogel fiber stability via electrostatic interactions, but knowledge of how they modulate mechanical properties of individual polyelectrolyte polymers is lacking. In this study, electrospun polyacrylic acid with chitosan is used as a model system to evaluate ferric ion effect on nanofiber mechanics. Using dark field microscopy imaging and persistence length analysis, we demonstrate that ferric ions modulate the bending stiffness of nanofibers. Young's modulus of individual nanofibers is estimated at values of a few kilopascals, …

WetLab-2 performs quantitative polymerase chain reaction (qPCR) aboard the micro-gravity environment of the International Space Station in order to research how the stress of micro-gravity affects an organism’s RNA production and gene expression. This on-station qPCR allows researchers to gather information earlier and thereby allow them to modify experiments in a matter of hours rather than wait until the next flight opportunity. After a successful validation flight on SpaceX-8 and follow-up investigation on SpaceX-9, system components must now be adjusted for a new type of biological sample. Astrobee is a free-flying, 1'x1'x1' cuboid robot that is planned to be onboard the International Space Station (ISS) by February 2019. Astrobee will be the successor of the current ISS free-flyer, SPHERES. Free-flying robots possess a range of potential. They can facilitate work for astronauts aboard the ISS by performing rudimentary tasks and housekeeping (e.g., record-keeping, sensor analysis, etc.) efficiently. Free-flyers also act as innovative testbeds for guest scientists to perform zero-gravity control theory research. In recent years, SPHERES has hosted annual Zero Robotics (ZR) programming competitions in collaboration with MIT, challenging middle and high school students to write code …
