Off-Target At-Scale Scale Down Model Verification Of A Marketed Biopharmaceutical, Markus Emmler, Stefanie Menrad, Yosuke Watanabe

Cell Culture Engineering XVI

According to authority guidelines, process understanding is considered the key factor for a successful control strategy in submissions for biopharmaceutical production processes. This knowledge is usually gained in scale down model (SDM) experiments and therefore is subject to the ability of the SDM to reliably predict behavior of cell culture processes in manufacturing scale. In order to prove predictability, SDM runs at target conditions are typically compared to manufacturing scale target runs only. At the same time, predictability is also assumed within the whole design space described or even beyond as evaluated in process design studies. The proof at off-target …

Development Of A Platform Expression System Using Targeted Integration In Chinese Hamster Ovary Cells, Scott Bahr, Trissa Borgschulte, Michael Johns, Jeff Galligan

Cell Culture Engineering XVI

In recent years the biomanufacturing industry has seen significant improvements in recombinant protein production titers due to advancements in protein expression technologies as well as media, feed and manufacturing process development. However, the standard methods of recombinant cell line development have remained relatively unchanged. The majority of the biopharma industry introduces transgenes into Chinese Hamster Ovary (CHO) cells using mechanical or chemical transfection processes followed by metabolic or antibiotic selection of stable recombinant pools. Through this process, the transgene(s) are randomly integrated into the genome, often times resulting in significant heterogeneity within the stable pools. Individual recombinant CHO cells within …

The Relevance Of Cell Size In A Cho Fed Batch Process: Metabolic And Transcriptomic Characterization, Dirk Martens, Xiao Pan, Rene Wijffels, Ciska Dalm

Cell Culture Engineering XVI

Normally the growth profile of a CHO cell fed-batch process can be divided into a growth phase followed by a stationary (non-growth) phase and a cell death phase. In this study, using a commercially available media system in a 10 liter fed-batch reactor, the growth phase is followed by a phase where cell division stops but cell growth continues in the form of an increase in cell size (Figure 1). During the cell size increase (SI) phase, the average volume and dry weight per cell increase linearly with time up to nearly threefold. Cell cycle and transcriptome analysis show that …

Cell Culture Engineering XVI

Over the last several years demonstration of cell line clonality has been a topic of many industry and regulatory presentations and papers. Guidance has been provided by the regulatory authorities, especially the FDA, on a path forward for providing evidence of clonality with high probability. It has been recommended that two-rounds of limiting dilution cloning (LDC) at sufficiently low seeding densities (≤0.5 cells/well) provides sufficient evidence that a cell line is clonal. Furthermore, one-round of LDC may also suffice if supplemental data from a characterized FACS or plate-imaging workflow are also included in the package.

Cell lines generated by methods …

Probability Distributions In The Glass Failure Prediction Model, Samir Blanchet, H. Scott Norville, Stephen Morse May 2018

Probability Distributions In The Glass Failure Prediction Model, Samir Blanchet, H. Scott Norville, Stephen Morse

Michigan Tech Publications, Part 1

Glass, a brittle material, fractures under tensile stress acting over a time duration. Lateral loads, such as wind, acting on a simply supported rectangular glass lite, put one surface of the lite primarily into tension. ASTM E 1300 defines load resistance of glass as the uniform lateral loading acting over a duration of 3 seconds that is associated with a probability of breakage of 8 lites per 1000 at the first occurrence of the loading. To determine load resistance, the underlying window glass failure prediction model facilitates determination of a probability distribution of 3 second equivalent failure loads, P3. The …

Sustainable Agriculture: Integration Of Aquaponics At Punta Leona Hotel And Club In Costa Rica, Cailin Sierra Dyer, Paris Riley Smith

Senior Honors Projects, 2010-2019

Sustainable agriculture is becoming an increasingly important method of food production. As human populations continue to grow, attendant food demand has been increasingly met via agribusiness, including monoculture crop production and factory farming. As is well documented, the rise of agribusiness has led to resource degradation and declining stocks on which “sustainable agriculture” relies. This paper describes a local attempt to re-establish “sustainable agriculture” through the development of an aquaculture system that mimics a naturally occurring cycle that integrates fish and plants. The system was constructed over a three-week period in Punta Leona, Costa Rica. First, the ground was cleared …

Tanzania Journal of Science

Phenetics is one of approaches used to delimit species in plant classification. Conclusion in phenetics is based on overall similarity often of morphological data. The approach uses coded data that are analyzed using coefficients to create similarity matrices that are analyzed using clustering analysis to create the classification. Exists different similarity coefficients and coding methods though in practice are used intuitively sometimes giving results that have been challenged. Though similarity coefficients and coding methods have some times been blamed, studies to analyze their influences are limited. The trend however is to avoid morphological data in favor of DNA markers. The …

Tolerance Sensitivity Analysis And Robust Optimal Design Method Of A Surface-Mounted Permanent Magnet Motor By Using A Hybrid Response Surface Method Considering Manufacturing Tolerances, Cha-Seung Jun, Byung-Il Kwon, Ohbong Kwon

Publications and Research

This paper presents a robust optimal design method using a hybrid response surface method (H-RSM) which directly finds an optimal point satisfying a target Z-value or a probability of failure. Through three steps, this paper achieves the goal that is to increase the open-circuit airgap flux (OCAF) in a surface-mounted permanent magnet motor and decrease its variation caused by variations of the airgap lengths including an additional one between permanent magnets and rotor back yoke. First, the OCAF equation is derived from the magnetic equivalent circuit (MEC) considering the additional airgap. Then, the equation is validated by comparing its results …

Determinants Of Multi-Scale Patterning In Growth Plate Cartilage, Alek Erickson May 2018

Determinants Of Multi-Scale Patterning In Growth Plate Cartilage, Alek Erickson

Theses & Dissertations

ABSTRACT

Functional architectures of complex adaptive systems emerge by dynamic control over properties of individual components. During skeletal development, growth plate cartilage matches bone geometries to body plan requisites by spatiotemporally regulating chondrocyte actions. Bone growth potential is managed by the proximodistal patterning of chondrocyte populations into differentiation zones, while growth vectors are specified by the unique columnar arrangement of clonal groups. Chondrocyte organization at both tissue and cell levels is influenced by a cartilage-wide communication network that relies on zone-specific release and interpretation of paracrine signals. Despite genetic characterization of signaling interactions necessary for cartilage maturation, the regulatory mechanisms …

A Study Of Acoustically Activated Nanodroplets, Songita Choudhury

Theses & Dissertations

Current treatment of acute myocardial infarction (AMI), which is the main pathophysiological event leading to death in the United States, has advanced considerably with the introduction of emergent percutaneous interventions, but there remains an urgent need for novel techniques to rapidly and accurately detect infarcted or ischemic tissue that results from AMI. Ultrasound contrast agents, also known as microbubbles (MB), have become commonplace in echocardiography. However, MBs are purely intravascular tracers and unable to cross endothelial barriers due to size. The limitations of MBs, namely size and short circulation times within the human body, led to the development of phase-change …

Analysis Of The Contribution Of Transportation And Land Use To Citizen Perceptions Of Livability, Rebecca Lewis

PSU Transportation Seminars

What is livability? How does the built environment influence resident perceptions of livability? Although livability is a broadly used term and a key goal in land use and transportation plans at the state level, it is unclear whether residents think their neighborhoods are livable and what contributes to their perception of livability. The purpose of the project was to understand how Oregonians, in neighborhoods of varying densities and within Metropolitan Planning Organizations (MPOs), perceive livability at the nexus of transportation and land use. We sought to understand how residents define and perceive livability in three different MPOs in the state: …

Can Your P2o5 Be Commercially Exploited?, Ewan Wingate, Michael E. Kelahan, M. Robert Kelahan

Beneficiation of Phosphates VIII

World reserves of high-grade, high-quality phosphate rock are steadily decreasing. As a result, lower quality rock is being increasingly evaluated to determine its potential for commercial exploitation. It is well known that the chemical and mineralogical composition of phosphate rock varies widely from deposit to deposit, and therefore, the conditions for processing different phosphate rock types also varies.

Unfortunately, most early stage phosphate feasibility studies tend to focus on the P2O5 content and neglect the other chemical constituents or impurities in the rock. It is the remainder of the chemical constituents that often define the quality of …

Beneficiation of Phosphates VIII

Phosphorus has its main application in the production of phosphoric acid, being the apatite series its main ore. The Brazilian main phosphorus reserves are igneous, as is the case of Catalão alkaline dome, were Copebras/CMOC International has operations. The content of P2O5 in the ROM has been reduced over the years and, as a consequence, the content of the contaminants in the concentrate, produced by flotation, which enhances the difficulty in the phosphoric acid production. Only traces of arsenic, iron, and heavy metals can be present in high-purity phosphoric acid. The circulating load of the Copebras’ phosphate …
