Alzheimer Disease And Anesthesia, Gözde İnan, Zerri̇n Özköse Şatirlar Jan 2015

Alzheimer Disease And Anesthesia, Gözde İnan, Zerri̇n Özköse Şatirlar

Turkish Journal of Medical Sciences

Alzheimer disease (AD) is one of the most common neurodegenerative diseases and the most prevalent form of dementia. Some factors in the development of AD, age being the best-known one, have been suggested; however, no causes have been found yet. The pathophysiology of the disease is highly complex, current therapies are palliative, and a cure is still lacking. Adverse effects of anesthetics in the elderly have been reported since the 1950s; however, awareness of this old problem has recently gained importance again. Whether exposure to surgery and general anesthesia (GA) is associated with the development of AD has been questioned. …

Turkish Journal of Medical Sciences

Background/aim: The uptake of the ferric-acinetobactin complex into the periplasmic space relies on the baumannii acinetobactin utilization (BauA) protein. BauA is composed of cork and the β-barrel domains. We constructed a recombinant protein from conserved antigenic domains of cork and the β-barrel of BauA to evaluate their immunogenic role in an animal model. Materials and methods: The selected bauA domains were amplified from a purified genome of Acinetobacter baumannii ATCC 19606. The domains were then cloned into pET28a and the proteins expressed in E. coli BL21 (DE3) were purified using nickel nitrilotriacetic acid chromatography. Mice and rabbits were immunized with …

Ebola Viral Disease: What Should Be Done To Combat The Epidemic In 2014?, Ali̇ye Baştuğ, Hürrem Bodur

Turkish Journal of Medical Sciences

Ebola virus disease (EVD) was defined first in 1976. Since then, more than 24 epidemics have been reported from Africa, predominantly with the Zaire species. On 21 March 2014, the current West Africa outbreak was reported by the World Health Organization, the largest one ever recorded. The Congo epidemic was reported in July 2014. It was considered that the two epidemics had unrelated origins with 96.8% identical genomic sequence of the virus. EVD outbreaks occurred in areas with limited resources but it has a potentially global effect due to the possibility of imported infection and the potential misuse of the …

Utility Of M30, An Apoptotic Serum Marker, In Liver Diseases, Aki̇f Altinbaş, Şahi̇n Çoban, Ömer Başar, Osman Yüksel

Turkish Journal of Medical Sciences

The aim of this paper is to evaluate the role of apoptosis in some common liver diseases, and the utility of M30, an apoptotic serum marker, in the diagnosis of the severity of underlying hepatic injury. As is widely known, apoptosis is programmed cell death, and its deregulation results in an uncontrolled inflammatory process leading to upregulation of liver fibrogenesis. Both extrinsic and intrinsic pathways are crucial in apoptosis, and caspase cleavage of cytokeratin proteins occurs in both. Therefore, the measurement of caspase-cleaved cytokeratin fragments could be a novel method to assess the intensity of apoptotic cell numbers in epithelial …

Salivary Glucose And Antioxidant Defense Markers In Type Ii Diabetes Mellitus, Sayeeda Mussavira, Mala Dharmalingam, Bindhu Omana Sukumaran

Turkish Journal of Medical Sciences

To evaluate salivary antioxidant defense markers, their correlation with salivary glucose, and glycemic status in type II diabetes mellitus (DM). Materials and methods: The study included 53 diabetic patients and 40 healthy subjects. Salivary glucose, blood glucose, and uric acid (UA) were determined by specific enzymatic methods. Total antioxidant activity (AOA), glutathione (GSH), catalase (CAT), superoxide dismutase (SOD), and total protein were determined spectrophotometrically. Results: Salivary UA (3.12 vs. 1.89 mg/dL), GSH (47 vs. 11.92 µg/mL), and total protein (375.12 vs. 202.23 mg/dL) were significantly higher (P < 0.001; r = 0.455, 0.735, 0.498 respectively) and AOA (653.1 vs. 897.3 µmol/L) was significantly lower in the DM group (P < 0.001, r = -0.431) compared to healthy controls. Among the antioxidant enzymes, CAT was significantly lower (1214 vs. 9468.9 kat) in the DM group (P < 0.001, r = -0.886). Spearman correlation analyses within the diabetic group showed a strong positive association between salivary glucose and blood glucose (P < 0.001, r = 0.9), salivary glucose and GSH, and salivary glucose and UA. Salivary glucose showed a negative correlation with AOA and CAT (P = 0.008, r = -0.447) in the diabetic group. Conclusion: Findings of this study, showing a strong correlation between salivary glucose and blood glucose as well as changes in antioxidant components in the DM group, suggest that saliva can be used for the diagnosis and management of DM.

Turkish Journal of Medical Sciences

Background/aim: To determine whether macrophage migration inhibitory factor (MIF) and monocyte chemoattractant protein-1 (MCP-1) levels in patients with hepatitis B (HB) are different than in normal individuals and whether the HB surface antigen (HBs Ag) level and viral load are correlated with each other and with the two aforementioned parameters. Materials and methods: Sera were obtained from 52 chronic active HB (CAHB) patients and 33 healthy controls, and their MIF and MCP-1 levels were measured. Statistical analyses were performed. A value of P < 0.05 was considered statistically significant. Results: The MIF and MCP-1 values of the control group were increased compared to those of the CAHB group. The MIF and MCP-1 levels were negatively correlated with HBs Ag levels and viral loads. The MIF and MCP-1 levels were positively correlated. The HBs Ag levels and the log10 of the viral loads were positively correlated. Conclusion: We conclude that the negative correlation of MIF and MCP-1 with viral load and HBs Ag levels may be due to T-cell deficiency, antinuclear antibody seropositivity, and/or inhibition of chemokine ligand 2 receptors by viral antigens. More studies with a greater number of subjects are needed to evaluate the potential role of MIF and MCP in CAHB.

Background/aim: The severity of disease outcome in dyspepsia has been attributed to Helicobacter pylori virulence genes. The aim of this study was to determine the distribution of H. pylori virulence genes (cagA, babA2, and dupA) and to determine whether or not there arises a significant correlation with clinical dyspepsia outcomes. Materials and methods: H. pylori genotypes cagA, babA2, and dupA were identified by polymerase chain reactions from gastric biopsy samples in 105 H. pylori-positive patients. Results: The positive rates for cagA, babA2, and dupA genes in H. pylori dyspeptic patients were 69.5%, 41.0%, and 22.9%, respectively. cagA was more prevalent …

Background/aim: Asymmetric dimethylarginine (ADMA) plays role in the pathogenesis of coronary artery disease and related mortality and morbidity through a number of mechanisms. We hypothesized that plasma ADMA levels would be increased in the presence of reversible ischemia as measured by GATED single photon emission computed tomography (SPECT) myocardial perfusion scintigraphy (MPS). Materials and methods: Fasting i.v. blood samples were drawn before testing. All patients underwent 99mTc-sestamibi GATED SPECT MPS with a one-day stress-rest protocol; the images were visually analyzed. Post-stress GATED parameters, including ejection fraction, end systolic and end diastolic volumes, and automatic stress defect scores, were recorded. Results: …
