Fta™ Card Use For Detection Of Mycoplasma Gallisepticum In Wild Birds, Linda Purvis, Dawn E.W. Drumtra
Georgia Journal of Science
Flinders Technology Associates (FTA) cards are increasingly accepted as an accurate method for nucleic acid detection, including testing for the presence of various avian diseases. However, the accuracy of FTA cards for detection of Mycoplasma gallisepticum (MG) in wild birds has not been published. Traditional techniques for testing MG in birds include serum plate agglutanation (SPA), hemagglutination inhibition (HI), polymerase chain reaction (PCR), and enzyme linked immunosorbent assay (ELISA) all of which are time consuming and require significant financial investment into specialized equipment. We tested the efficacy of using FTA cards to detect MG in wild bird carcasses collected in …

Characterization Of The Slc7a5/Slc3a2 Amino Acid Transporter In Human Pleural Mesothelial Cells, Ann Francis
Biotechnology Theses
The SLC7A5/SLC3A2 amino acid transporter moves essential amino acids, such as leucine, phenylalanine, and tryptophan, across the cellular plasma membrane. SLC7A5 overexpression was identified in diverse cancers, suggesting a role in tumorigenesis and cancer cell proliferation. However, the role of SLC7A5/SLC3A2 in other aggressive cellular phenotypes, including mesothelial mesenchymal transition (MesoMT), remains unexplored. RNA sequencing analyses identified SLC7A5 as one of three highly upregulated SLCs in mesothelial cell derived myofibroblasts. This sodium independent transporter exhibits a high affinity for branched chain amino acids such as leucine; however, what role, if any, this channel plays in MesoMT is unclear. SLC7A5 knockdown …

Explorations Of Dna-Single-Walled Carbon Nanotube Interactions To Develop Multiplexed Molecularly Specific Biosensors For Inflammation, Amelia K. Ryan
Dissertations and Theses
Inflammatory cytokines such as interleukin-6 (IL-6) and interleukin-12 (IL-12) are central regulators of immune signaling and key biomarkers of disease, yet existing assays for their detection remain slow, invasive, and lack multiplexing ability. This dissertation advances the development of single-walled carbon nanotube (SWCNT) optical nanosensors capable of real-time, multiplexed, and molecularly specific cytokine detection through innovative use of single-stranded DNA (ssDNA) interfaces.
First, an IL-6-specific DNA aptamer was employed as both a dispersing agent and recognition probe for SWCNT fluorescence sensing. Sequence modifications, including anchor domains, truncations, and thermally induced refolding, were systematically tested to optimize sensitivity and selectivity. The …

Genetic Property Rhetoric And The Public Domain, Dave Fagundes
Texas A&M Journal of Property Law
Among the many themes that illuminate Jorge Contreras’s The Genome Defense, this Essay focuses on property. It considers the rhetorical role played by property in convincing both courts and the public that patenting genetic data was legally and ethically objectionable. This Essay begins by locating property—and public property—as a major theme in the dialogue surrounding the Myriad litigation. It then situates this rhetorical move in the context of property-talk generally, highlighting the distinctiveness of leveraging public rather than private ownership as a means of accessing moral intuitions. Finally, this Essay reflects on what the use of public property rhetoric …

Optimization Of Transformation And Regeneration In The Sunflower (Helianthus Annuus), Sara M. Kilar
Masters Theses
In addition to their value as prized ornamentals, sunflowers are important crop species due to the nutritional value and high oil content of the seeds. Sunflower oil is a significant contributor to global seed oil production, which can be used as a food additive and cooking agent as well as a raw material for biodiesel production. While traditional breeding efforts have led to the development of many unique cultivars, genetically modified variations have proven more difficult due to the recalcitrance of sunflower plants to genetic transformation. As a result, facile and efficient transformation methods for sunflowers have the potential to …

Functional Analysis Of Ligand Binding Domain 2 Of Outer Surface Protein C Of Borreliella Burgdorferi, Gavin Z. Chambers
Theses and Dissertations
Lyme disease, caused by the spirochete Borreliella burgdorferi, is the most common vector-borne infection in North America. A critical determinant of early mammalian infection is outer surface protein C (OspC), a highly variable lipoprotein that is indispensable for spirochete survival but whose precise function remains unresolved. OspC has been proposed to mediate diverse roles, including complement evasion, adhesion, tissue dissemination, and antiphagocytic activity, supporting the view that it is a multifunctional virulence factor. Structural studies have identified two putative ligand-binding domains (LBD1 and LBD2), with LBD2 forming a solvent-exposed crown at the membrane-distal end of the OspC dimer.

Urban water sewage is a potential health concern due to its possibility to spread contagious RNA viruses such as Coxsackievirus B3. However, detection of viral particles remains challenging because of low viral concentrations in wastewater and high mutation rates of the RNA virus. To address this, this study proposes a novel viral detection method using synthetic riboswitches that bind to the target virus and trigger a reporter gene, amplifying the detection signals. To support the design of effective riboswitches, we present a machine learning model for classifying riboswitch performance, integrating RNA sequence data with secondary structural features. This model used …

Prostasin, also known as PRSS8 or CAP1, is a glycosylphosphatidylinositol (GPI)-anchored extracellular serine protease involved in various physiological and pathophysiological processes. Prostasin expression is observed during key phases of trophoblast invasion and placental development, critical steps in embryo implantation in Rhesus monkeys. This suggests a role for prostasin in extracellular matrix remodeling and tissue invasion, both essential for a healthy pregnancy. Moreover, prostasin appears vital for development as studies in mice show that a germline knockout of the gene leads to significant placental defects and eventual embryonic death, highlighting its importance in vascular and structural development within the placenta.

Intradermal injections of silicone-based fillers for the face and body may cause chronic inflammation and fibrotic reactions in some individuals. Chickens from the UCD 200/206 White Leghorn (WL) line spontaneously develop a fibrotic autoimmune disease known as scleroderma/systemic sclerosis (SSc). Using the SSc-prone UCD and healthy WL chickens, a longitudinal study was conducted to examine the cellular inflammatory responses to intradermal (i.d.) injections of sterile SILIKON-1000. Specifically, the pulp (a skin derivative) of growing feathers (GFs) was i.d. injected with sterile SILIKON-1000 or phosphate-buffered saline (PBS) as injection control (8 and 3 birds/line, respectively; 10 µL/GF; 20 GFs/bird). Two GFs …

Theses and Dissertations
The rise in drug-resistant bacteria, together with the decline in antibiotic development, requires new strategies for infectious disease control. Gram-negative pathogens are particularly challenging to combat due to their outer membrane. This study highlights the effectiveness of the silver-containing antimicrobial AGXX® against the Gram-negative bacterium Escherichia coli. AGXX® is a promising silver coating that presumably kills bacteria through the generation of reactive oxygen species (ROS). However, neither AGXX®’s mode of action is fully understood, nor have its effects on Gram-negative bacteria or bacterial response and defense mechanisms towards AGXX® been studied in detail. Here, I report that the bactericidal …
