Preterm birth is the leading cause of neonatal morbidity and mortality worldwide, yet current clinical tools for predicting its risk lack specificity and reliability. Cervical mucus plays a crucial role in maintaining pregnancy by forming a barrier that protects the uterus against infection. This study investigates the use of laser speckle rheology as a noninvasive technique to assess cervical mucus biomechanics. Synthetic cervical mucus was fabricated at varying hydration levels and tested using a conventional rheometer that confirmed that the mechanical properties of the synthetic mucus samples were representative of physiological cervical mucus. Abdominal gunshot wounds are amongst the deadliest injuries encountered in combat and remote emergency settings. For military medics and first responders, the challenge lies in controlling internal bleeding and preventing infection before patients can reach surgical care-often within a narrow window of time known as the “golden hour”. Currently available field treatments are either too slow, not designed for internal wounds, or fail to provide antimicrobial protection. Heart murmurs are critical indicators of potential cardiovascular conditions yet their detection can often go unnoticed due limitations in equipment and is dependent on clinician experience. Cardiovascular disease remains a leading cause of death worldwide, therefore, early detection of heart murmurs play a crucial role in improving patient outcomes. Accurate prediction of breast cancer recurrence plays a critical role in guiding treatment decisions, particularly regarding the use of systemic therapy for patients. While genomic assays such as the Oncotype DX Recurrence Score offer valuable prognostic information, they are limited in accessibility and application. The work presented in this thesis explores an imaging-based approach for the prediction of breast cancer recurrence that combines multiphoton microscopy (MPM) with deep learning to classify individual breast cancer biopsy samples by risk of recurrence. Cancer is a result of uncontrolled growth and reproduction of cells. Damaged or abnormal cells grow and multiply when they should not and in turn the cells continue to proliferate and metastasize. Malignant cells rely on functional mitochondria for proliferation, metastasis, and survival. These mitochondria maintain a functional mitochondrial network through a quality control mechanism called mitophagy. Mitophagy is a conserved intracellular process to maintain quality control and remove damaged mitochondria.

Post-operative orthopedic surgical site infections (SSIs), specifically those caused by methicillin resistant Staphylococcus aureus (MRSA), present a significant clinical challenge due to delayed detection, increased morbidity, and current rising healthcare costs. Early detection of infection remains limited due to the current methods of diagnosis, which consist of observation of the surgical site and laboratory infrastructure for conformation of infection. Various microbial communities in marine biofilms cause biofouling on submerged surfaces, posing challenges to marine industries. Despite their ecological and economic importance, biofilms' biomechanical and biochemical responses to hydrodynamic shear stress are insufficiently understood, particularly in dynamic flow conditions. To fill this gap, our study uses an innovative fiber-optic hyperspectral imaging (HSI) system and a supercontinuum laser source to examine marine biofilms' structure, composition, and detachment behavior at different shear stress levels.

In this experiment, the goal was to design, build, and test an Electroencephalogram (EEG) circuit that would then be paired with a touchscreen display as a portable unit for use in Schizophrenic research at East Tennessee State University at the Quillen College of Medicine. Cardiovascular diseases remain the leading cause of death worldwide and coronary microvascular dysfunction is one cardiovascular disease which is particularly challenging to diagnose and treat. Coronary thermodilution is a widely used, invasive procedure to evaluate microvascular function by generating physiological indices such as coronary flow reserve and the index of microvascular resistance. Although commonly cited as an indicator dilution method in the cardiovascular literature, current coronary thermodilution techniques lack rigorous application of the accompanying mathematical framework, thereby leaving potentially valuable diagnostic information unexploited. Cancer is a complicated disease and one in every three diagnoses are women with breast cancer. Recent research shows that the progression of cancer depends strongly on interactions with other non-cancerous cells and non-cellular components. Metastasis, the spread of cancer to other parts of the body, is a key step in cancer progression and makes treatment harder, leading to poor response to treatments culminating in death in ~90% of patients. This report details the design and development process of Cruising on the Beach (COB), a modular motorized wheelchair intended to improve beach accessibility for high school students with mobility impairments. The project aimed to create an affordable, portable, and sand-capable device using balloon wheels, an adjustable control panel, and a customizable base. Extensive background research, market analysis, design modeling, and test plans were completed. However, final fabrication was not achieved due to financial constraints and limited access to industrial machining tools needed for custom metal components. In the United States, due to the increasing prevalence of cardiovascular disease, over 900,000 cardiac surgeries are performed annually, with projections exceeding 1.3 million by 2029. Many post-surgical patients require continuous in-hospital cardiac monitoring, which is typically conducted using electrocardiograms (EKGs). Approximately 1.6 million people in the U.S. have lower limb amputations, which is expected to double by 2050 (Wong et al.). Socio-economic factors: income, education, rural residency & race, impact amputation risk (Okesina et al.), with Black Americans 70% are more likely to undergo major lower limb amputations than white Americans, even after adjusting for care access & comorbidities (Feinglass et al., Holman et al., Regenbogen et al.). Within 5 years, up to 50% of first-time amputees require another amputation (Dilingham et al., Glaser et al.) & those with comorbidities face a 77% five-year mortality rate (Fortington et al.).

Capillary zone electrophoresis (CZE) is a powerful analytical technique widely used for biomolecular separation due to its high resolution, efficiency, and sensitivity. This report presents the development and validation of a novel CZE system designed to improve modularity, reproducibility, and operational efficiency. The system integrates a laser-induced fluorescence (LIF) detection method with a 488 nm laser, enhanced fluid control through a negative pressure system, and a user-friendly GUI for automated operation and data acquisition. Performance verification included pressure stability tests, fluorescence accuracy assessments, and electrophoretic reproducibility studies. WSPEs are largely caused by circumvention of the timeout procedure (a planned pause before incision to stop and confirm crucial details of the procedure to be performed known as ‘The Universal Protocol’). Accurately diagnosing appendicitis remains a significant challenge in emergency medicine due to its varied clinical presentation and overlap with other abdominal conditions. Existing diagnostic models often rely on unimodal data, such as computed tomography (CT) images or clinical notes, limiting their ability to capture the full complexity of patient information. This document outlines the key steps taken in the specification, design, and functionality of the Tiny Temp Neonatal Intensive Care Unit Infant Temperature Monitor. The BICEP team identified temperature monitors for infants in incubators in the NICU as an area of need after surveying hospitals in the San Luis Obispo area. Current thermometers are oversimplified in design, consisting of only the temperature probe and a piece of adhesive holding the probe directly onto the infant's skin. Our research team developed a PVNS for Epilepsy device which is intended to act as a preventative measure against seizures for patients with epilepsy. The device is a less invasive alternative to the VNS device on the market using electroacupuncture at the Shenmen and Xin points along the posterior pinna to stimulate the vagus nerve.

Currently, track and field sprinters ranging from beginner level to world-class have no practical tool that provides quantitative feedback of value on quality of start and quality of the three distinct phases of their sprint race, such that they can quantify the value of their adjustments. Our team will create a device that integrates with the sprint starting block to provide quantifiable data on the quality of a sprinter’s start. This Statement of Work describes drawbacks of current solutions, existing patents, customer requirements, engineering specifications, and our project plan.

The increasing demand for affordable and accessible prosthetic solutions has driven innovation in the field of robotics. In this project, we present the design and development of a 3D-printed robotic prosthetic arm aimed at addressing the challenges faced by individuals with upper limb amputations. Our prosthetic arm utilizes five micro linear actuators to achieve precise and naturalistic movement. The design of the prosthetic arm prioritizes affordability and accessibility, with a focus on leveraging 3D printing technology to reduce manufacturing costs and enable customization. Mechanical and material properties of bone are essential for understanding both normal skeletal function and the progression of bone-related diseases. Microindentation has become a widely used method for evaluating tissue mechanical properties of bone, as it samples the contributions of microarchitecture and composition. Microindentation using a standard pointed tip inherently introduces both elastic and plastic deformation into the material. In contrast, indentation with a spherical tip can be operated entirely in the elastic range, providing for a non-damaging test that has significant advantages over sharp indentation techniques. Evaluating the effectiveness of transcranial direct current stimulation (tDCS) is essential for guiding its integration into therapeutic and performance-enhancement applications. In our laboratory, we investigate the efficacy of tDCS across multiple experimental models, including both animal and human studies. I have contributed significantly to the execution and analysis of these experiments, which include studies in rats and healthy human participants aimed at evaluating whether electrical stimulation of the motor cortex can enhance motor learning. These studies assess improvements in fine motor performance resulting from tDCS. In stroke patients, I contribute to our investigation of tDCS as a rehabilitative intervention, particularly …

Over two decades, there has been a significant rise in awareness regarding Type 1 Diabetes (T1D), leading to a deeper comprehension of numerous variables, including hereditary factors, patterns of occurrence, and health challenges. According to the Type 1 Diabetic Index, around 8.7 million people worldwide are affected by this chronic autoimmune illness marked by a lack of insulin and hyperglycemia. Hand function is often impaired following neurological trauma or in conditions such as stroke, limiting independence and the ability to perform activities of daily living (ADLs). Current clinical assessments rely heavily on subjective rating scales that lack the resolution to detect subtle changes in motor performance, which can limit access to rehabilitation. Recent advances in wearable technologies highlight the potential for hand-worn devices to provide objective, real-time measures of hand function.
