In vivo wear of ultra-high molecular weight polyethylene (UHMWPE) orthopedic bearings represents a major problem in shoulder and knee arthroplasty, with significant amounts of devices failing due to the complication itself, and even more due to associated reasons for retrieval, including aseptic loosening. However, quantification of UHMWPE wear has largely been limited to linear and in vitro volumetric analyses, obscuring the understanding of the amount of clinically relevant material loss. Mechanically assisted corrosion and non-mechanically (chemically) driven corrosion in modular junctions of orthopedic implants using cobalt-chromium-molybdenum (CoCrMo) alloys remain clinical concerns and may contribute to implant failure. The underlying corrosion mechanisms are not yet fully understood, leaving a critical research gap. We hypothesized that corrosion in modular junctions may be driven by aggressive local environments, including fretting, metal ion release (cobalt ion, Co2+), low pH and reactive oxygen species (ROS). The specificity and efficiency with which enzymes catalyze selective chemical reactions far exceeds the performance of traditional heterogeneous catalysts that are predominant in industrial applications such as conversion of commodity chemicals to value-added products, fuel cells, and petroleum refinement. Moreover, biocatalysts exhibit exceptionally high product turnover at ambient conditions with little health and environmental burden. Naturally sourced, renewable biomaterials possess outstanding advantages for a multitude of biomedical applications owing to their biodegradability, biocompatibility, and excellent mechanical properties. Of interest in this dissertation are silk (protein) and chitin (polysaccharide) biopolymers for the fabrication of functional biodevices. One of the major challenges restricting these materials beyond their traditional usage as passive substrate materials is the ability to combine them with high-resolution fabrication techniques. The synthesis and in vitro degradation analysis of thin films of poly[(glycineethylglycinato)75(phenylphenoxy)25phosphazene] (PNGEG75PhPh25) and poly[(ethylphenylalanato)25(glycine- ethylglycinato)75phosphazene] (PNEPA25GEG75) blended with poly(lactic-co-glycolic acid) (PLGA) was conducted to determine the blends’ potential for use as scaffolding materials for tissue regeneration applications. Altering the foreign body reaction by targeting macrophages has been of interest in the biomaterials field to improve the integration of longevity of implanted biomedical devices. The objective of this dissertation was to study the effect of locally delivered bioactive modulators on macrophage activation at the implantation site of different biomaterials in rats. Iloprost, a prostacyclin analog, was tested for its ability to direct macrophages to their pro-wound healing phenotype after the implantation of microdialysis probe in the subcutaneous space of male Sprague Dawley rats. Ultra high molecular weight polyethylene (UHMWPE) is a material used in artificial implants for articular joint replacements. However, these implants have a limited lifespan in which the patient will be pain-free due to the wear of the UHMWPE components. Recently crosslinking, or exposing the material to radiation, has been used to extend the wear resistance of UHMWPE. Crosslinking introduces another set of drawbacks; mainly the reduction of the fracture toughness of UHMWPE and the generation of free radicals, which leave the polymer vulnerable to damage from oxidation.
