Neurological disorders affect millions of people worldwide. In the case of Alzheimer’s disease (AD), the rising number of cases significantly increases global healthcare costs. Therefore, early detection of biomarkers at the onset of the disease is essential for early diagnosis. This study presents an electrochemical biosensor designed for the detection of Brain-Derived Neurotrophic Factor (BDNF) in human plasma, incorporating reduced graphene oxide (rGO) to enhance signal sensitivity. Dielectrophoresis (DEP) was employed to deposit rGO flakes on the electrode surface, improving conductivity and sensor performance. Capillary zone electrophoresis (CZE) is a powerful analytical technique widely used for biomolecular separation due to its high resolution, efficiency, and sensitivity. This report presents the development and validation of a novel CZE system designed to improve modularity, reproducibility, and operational efficiency. The system integrates a laser-induced fluorescence (LIF) detection method with a 488 nm laser, enhanced fluid control through a negative pressure system, and a user-friendly GUI for automated operation and data acquisition. Performance verification included pressure stability tests, fluorescence accuracy assessments, and electrophoretic reproducibility studies. The Yang procedure is a new aortic root enlargement technique used to enlarge the aortic annulus by multiple valve sizes. The procedure prevents patient prosthesis mismatch and establishes a viable platform for future valve-in-valve implantation. This study used the Yang procedure to investigate the hemodynamics in the aortic root and bioprosthetic valve regions after aortic root enlargement. Results indicate the velocity magnitude at the sinus regions of a patient who underwent the Yang procedure was slower, indicating risks of flow stasis and thrombosis. Simulation results denote computational models can be created for optimization of surgical procedures.

Aortic stenosis impacts approximately 7% of the global population. In the past decade, the role of computational modeling has been becoming considerably important in the design of BHVs. To obtain reliable solutions in computational modeling, it is essential to consider accurate properties of bioprosthetic heart valves (BHVs), such as density and mechanical properties. This study proposes a novel computational method to quantify guide stability for Patient Specific Instrumentation (PSI) guides. A finite element contact model was used to analyze the final position of PSI guides on a femur across a range of loading parameters representing forces applied by a surgeon during operative use. Separate segmentation methods were used for the guide and bone geometry to represent differences between segmentation and actual patient geometry. Transcatheter mitral valve replacement (TMVR) is being developed to become a substitute therapy for surgery in prohibitive or high surgical risk patients to treat severe mitral regurgitation. A limited number of TMVR systems are under clinical evaluation. However, transcatheter mitral valve (TMV) long-term durability and hemodynamic performance is not known. TMV durability and hemodynamics must match with that of surgical bioprostheses for potential commercialization of TMVR. Mathematical models can be very useful for understanding complicated systems and for testing algorithms through simulation that would be difficult or expensive to implement. This dissertation presents a model that attempts to simulate the sound localization performance of persons using bilateral cochlear implants. The expectation is that this model could prove to be a useful tool in developing new signal processing algorithms for neural encoding strategies.

In the last thirty years, cochlear implants have become an invaluable instrument in the treatment of severe-to-profound hearing impairment. An important aspect of research in the continued development of cochlear implants is the in vivo assessment of signal processing algorithms intended to improve perception of speech and other auditory signals.
