Morphological Features Of Dysplastic Progression In Epithelium: Quantification Of Cytological, Microendoscopic, And Second Harmonic Generation Images, Sandra Patricia Gordon

Graduate Theses and Dissertations

Advances in imaging technology have led to a variety of available clinical and investigational systems. In this collection of studies, we tested the relevance of morphological image feature quantification on several imaging systems and epithelial tissues. Quantification carries the benefit of creating numerical baselines and thresholds of healthy and abnormal tissues, to potentially aid clinicians in determining a diagnosis, as well as providing researchers with standardized, unbiased results for future dissemination and comparison.

Morphological image features in proflavine stained oral cells were compared qualitatively to traditional Giemsa stained cells, and then we quantified the nuclear to cytoplasm ratio. We determined …

This dissertation describes fabrication of devices and other tools for biomedical applications through the integration of acoustofluidic systems with bio separation assays, instrumentation components, and soft materials interfaces. For example, we engineer a new class of transparent acoustic flow chambers ideal for optical interrogation. We demonstrate efficacy of these devices by enhancing the signal for high throughput acoustic flow cytometry, capable of robust particle focusing across multiple parallel flowing streams. We also investigate an automated sampling system to determine the parameters of transient particle stream focusing in between sample boluses and air bubbles to model a high throughput, multi-sampling acoustic …

The Application Of Electric Cell-Substrate Impedance Sensing (Ecis) Biosensors, Xudong Zhang, Sunghoon Jang Nov 2018

The Application Of Electric Cell-Substrate Impedance Sensing (Ecis) Biosensors, Xudong Zhang, Sunghoon Jang

Publications and Research

Electric cell-substrate impedance sensing (ECIS) is a label-free and non-invasive technique for analyzing the activities and morphologies of cells. The ECIS sensors are able to measure the impedance spectroscopy of cells attaching on the sensor substrates. The cell activities and morphologies influence the measure impedance directly. Traditional toxicity analysis has complicated processes compared to biosensor-based analysis. In this study, the toxicity analysis was performed with biosensor based on ECIS technique. The experimental results show that the ECIS sensor is able to quickly distinguish the toxic and non-toxic substance.

An Engineer’S Take On The Bone-Ligament Interface: Utilizing Novel Technology To Improve Clinical Outcomes, Emma Garcia Nov 2018

An Engineer’S Take On The Bone-Ligament Interface: Utilizing Novel Technology To Improve Clinical Outcomes, Emma Garcia

Shared Knowledge Conference

Ligament repair is a common surgical practice with a significant lack of viable replacements. The current gold standard for repair is the use of tendon grafts from cadavers or from another place in the patient’s body; however, these often cause more problems than they solve including immune responses or a lack of mobility in another place in the body. Synthetic replacements are of growing interest, though the ability to mimic the complex structure of the ligament and how it connects to the bone remains an obstacle. Our lab built a 3D bioprinter combined with an electrospinner to address this complicated …

Dual-Display Laparoscopic Laser Speckle Contrast Imaging For Real-Time Surgical Assistance, Jaepyeong Cha, Corey Zheng, Lung Wai Lau Nov 2018

Dual-Display Laparoscopic Laser Speckle Contrast Imaging For Real-Time Surgical Assistance, Jaepyeong Cha, Corey Zheng, Lung Wai Lau

Pediatrics Faculty Publications

Laser speckle contrast imaging (LSCI) utilizes the speckle pattern of a laser to determine the blood flow in tissues. The current approaches for its use in a clinical setting require a camera system with a laser source on a separate optical axis making it unsuitable for minimally invasive surgery (MIS). With blood flow visualization, bowel viability, for example, can be determined. Thus, LSCI can be a valuable tool in gastrointestinal surgery. In this work, we develop the first-of-its-kind dual-display laparoscopic vision system integrating LSCI with a commercially available 10mm rigid laparoscope where the laser has the same optical axis as …

Impoved Hardware Design Of Iot Prosthetic Device, Yu Wang, Warren Hunter, Xiaolin Chen, Housney Ahmed, Haneefah Safo

Publications and Research

Our previous IoT based prosthetic arm prototype used servo motors to control finger movement through an Arduino Mega, which is connected to the muscle, pulse, and temperature sensors. The Arduino Mega was also connected to a Raspberry Pi 3 model B to transfer data from/to an online web application. One major limitation encountered during testing this prosthetic device was the space occupied by these components, which makes the device bulky. In addition, these servo motors cannot control the movement of the prosthetic device precisely. In this paper, we propose to improve on the existing prosthetic limb prototype by transitioning the …

Hyperspectral Imaging For Characterizing Single Plasmonic Nanostructure And Single-Cell Analysis, Nishir Sanatkumar Mehta

LSU Master's Theses

Orientation of plasmonic nanostructures is an important feature in many nanoscale applications such as photovoltaics, catalyst, biosensors DNA interactions, protein detections, hotspot of surface-enhanced Raman spectroscopy (SERS), and fluorescence resonant energy transfer (FRET) experiments. Silver nanocubes with significant spectral signatures between 400-700 nm are observed in this experimental research. Whereas study of single cells will enable the analysis of cell-to-cell variations within a heterogeneous population. These variations are important for further analysis and understanding of disease propagation, drug development, stem cell differentiation, embryos development, and how cells respond to each other and their environment. Adipose-derived mesenchymal stem cells possess the …

Microfluidic Electrical Impedance Spectroscopy, John J. Foley Sep 2018

Microfluidic Electrical Impedance Spectroscopy, John J. Foley

Master's Theses

The goal of this study is to design and manufacture a microfluidic device capable of measuring changes in impedance valuesof microfluidic cell cultures. Tocharacterize this, an interdigitated array of electrodes was patterned over glass, where it was then bonded to a series of fluidic networks created in PDMS via soft lithography. The device measured ethanol impedance initially to show that values remain consistent over time. Impedance values of water and 1% wt. saltwater were compared to show that the device is able to detect changes in impedance, with up to a 60% reduction in electrical impedance in saltwater. Cells were …

Design Modifications For A Small, Affordable Low Intensity Focused Ultrasound Device, Sui Shen, Kelsey C. Bayer, Pedro P. Irazoqui

The Summer Undergraduate Research Fellowship (SURF) Symposium

Depression is a prevalent and serious medical illness, and while there are antidepressant drugs to mitigate depressive symptoms, 10 - 30% of patients either do not respond or develop a tolerance to these medications. Literature supports that there is an interrelation between the inflammatory response and treatment-resistant depression. A promising method to tackle depressive symptoms is to block the inflammatory signaling pathway with vagus nerve stimulation (VNS), reducing pro-inflammatory cytokine levels. Although electrical VNS devices exist, they are invasive, expensive, and have side effects including voice alteration, dyspnea, and cough. Low intensity focused ultrasound (LIFU) is a promising method that …

The Summer Undergraduate Research Fellowship (SURF) Symposium

The common yeast infection, vulvovaginal candidiasis, affects three out of four women throughout their lifetime and can be spread to their child in the form of oral candidiasis (thrush). This disease is caused by the fungal pathogen Candida albicans, which is also a major cause of systemic candidiasis, a rarer but deadly disease with up to a 49% lethality rate. Current widely-used diagnostic methods include cell cultures, pH tests, and antibody detection, to assist effective treatment. Despite availability of various diagnostic methods, there is no inexpensive, rapid, and accurate way to detect C. albicans infection. This project aims to …

Magnetically Actuated Cell Stretching Platform To Induce Phenotypic Changes In Metastatic Cells, Yong Gyun Cho, Hyunsu Park, Hyowon Lee, Sarah Libring Aug 2018

Magnetically Actuated Cell Stretching Platform To Induce Phenotypic Changes In Metastatic Cells, Yong Gyun Cho, Hyunsu Park, Hyowon Lee, Sarah Libring

The Summer Undergraduate Research Fellowship (SURF) Symposium

Although metastasis is responsible for about 90% of cancer deaths, only few in vitro models can be used to evaluate dynamic behaviors of metastatic cancer cells. Many studies have shown that mechanical stimuli can trigger various cellular responses such as gene and protein expression, which could lead to changes in cellular phenotype. Similarly, metastasized breast cancer cells in the lung tissue are constantly stretched by cyclic mechanical stress due to breathing, which alters cellular morphology and proliferation state. Such transitions can make the secondary tumors resistant to the chemotherapy used to effectively treat the primary tumors. In this work, we …

Non-Invasive Diagnostic Measures Of Sensorineural Hearing Loss In Chinchillas, Hannah M. Ginsberg, Satyabrata Parida, Michael G. Heinz Aug 2018

Non-Invasive Diagnostic Measures Of Sensorineural Hearing Loss In Chinchillas, Hannah M. Ginsberg, Satyabrata Parida, Michael G. Heinz

The Summer Undergraduate Research Fellowship (SURF) Symposium

According to the World Health Organization, disabling hearing loss affects nearly 466 million people worldwide. Sensorineural hearing loss (SNHL), which is characterized as damage to the inner ear (e.g., cochlear hair cells) and/or to the neural pathways connecting the inner ear and brain, accounts for 90% of all disabling hearing loss. More concerning is that significant perceptual and physiological aspects of SNHL remain “hidden” from standard clinical diagnostics. Hidden hearing loss (HHL) manifests as the inability to understand speech in loud, noisy environments (e.g., listening in a noisy restaurant) despite a normal audiogram (i.e., normal detection of soft sounds). Recently, …

Bioelectrics Publications

Antimicrobial lock solutions are important for prevention of microbial colonization and infection of long-term central venous catheters. We investigated the efficacy and safety of a novel antibiotic-free lock solution formed from gas plasma-activated disinfectant (PAD). Using a luminal biofilm model, viable cells of methicillin-resistant Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Candida albicans in mature biofilms were reduced by 6 to 8 orders of magnitude with a PAD lock for 60 min. Subsequent 24-h incubation of PAD-treated samples resulted in no detectable regrowth of viable bacteria or fungi. As a comparison, the use of a minocycline-EDTA-ethanol lock solution for 60 …

Rotary Pump Speed Modulation To Produce Pulsatile Flow And Ventricular Volume Unloading., Connor Joseph Smith Aug 2018

Rotary Pump Speed Modulation To Produce Pulsatile Flow And Ventricular Volume Unloading., Connor Joseph Smith

Electronic Theses and Dissertations

Background: Continuous-flow (CF) left ventricular assist devices (LVADs) have gained widespread clinical acceptance as a treatment option for advanced heart failure (HF); however, they have also been associated with an increased risk of adverse events, including gastrointestinal bleeding, aortic insufficiency, and hemorrhagic stroke. It has been hypothesized that the increase in adverse event incidence may be due in part to diminished vascular pulsatility and high shear stress when CF-LVADs are operated at fixed speeds. Previous studies have shown that pump speed modulation generates greater levels of pulsatility in rotary pumps than when operated at fixed speeds. The objective of this …

Developing Droplet Based 3d Cell Culture Methods To Enable Investigations Of The Chemical Tumor Microenvironment, Jacqueline Alexandria De Lora

Biomedical Sciences ETDs

Adaptation of cancer cells to changes in the biochemical microenvironment in an expanding tumor mass is a crucial aspect of malignant progression, tumor metabolism, and drug efficacy. In vitro, it is challenging to mimic the evolution of biochemical gradients and the cellular heterogeneity that characterizes cancer tissues found in vivo. It is well accepted that more realistic and controllable in vitro 3D model systems are required to improve the overall cancer research paradigm and thus improve on the translation of results, but multidisciplinary approaches are needed for these advances. This work develops such approaches and demonstrates that new droplet-based cell-encapsulation …

Extended-Use Ecg Monitor, Daniel Aaron Soski Jun 2018

Extended-Use Ecg Monitor, Daniel Aaron Soski

Master's Theses

In this thesis, a prototype ECG monitor was developed that is integrated into an elastic shirt and takes a 3-lead ECG for over 5 days. The high-quality measurements can be used to identify markers indicative of various detrimental heart conditions. Measurements recorded by the device are encrypted and stored onto a micro-SD card. Current Holter monitors are expensive and have functional lives less than 48 hours; however, extended duration monitoring has been proven more useful in diagnosis. The device designed demonstrates that ECG measurements can be taken over longer durations without sacrificing quality, comfort, or device cost.
