Since the discovery of ammonia-oxidizing archaea (AOA), new questions have arisen about population and community dynamics and potential interactions between AOA and ammonia-oxidizing bacteria (AOB). We investigated the effects of long-term fertilization on AOA and AOB in the Great Sippewissett Marsh, Falmouth, MA, USA to address some of these questions. Sediment samples were collected from low and high marsh habitats in July 2009 from replicate plots that received low (LF), high (HF), and extra high (XF) levels of a mixed NPK fertilizer biweekly during the growing season since 1974. Additional untreated plots were included as controls (C). Terminal restriction fragment …

Background
The NF-κB activating kinases, IKKα and IKKβ, are key regulators of inflammation and immunity in response to infection by a variety of pathogens. Both IKKα and IKKβ have been reported to modulate either pro- or anti- inflammatory programs, which may be specific to the infectious organism or the target tissue. Here, we analyzed the requirements for the IKKs in myeloid cells in vivo in response to Francisella tularensis Live Vaccine Strain (Ft. LVS) infection.
Methods and Principal Findings
In contrast to prior reports in which conditional deletion of IKKβ in the myeloid lineage promoted survival and conferred resistance …

Development Of Plant Lectin Rtb For Delivery Of Therapeutic Proteins, Walter Acosta Gamboa
Student Theses and Dissertations
One of the main challenges for successful drug development is the creation of molecules that are able to cross the cell membrane to deliver therapeutics into specific subcellular compartments where they can serve their purpose. Among the therapeutic applications burdened with cell uptake limitations are enzyme replacement therapeutics (ERTs) for both lysosomal storage disorders (LSD) and endoplasmic reticulum storage diseases (ERSD). LSD and ERSD are rare disorders caused by defects in specific proteins within these cellular compartments, and current treatments rely on the regular provision of active enzyme to replace the missing/defective protein in patients. Plant lectin RTB, naturally present …

Metallization Of Dna And Dna Origami Using A Pd Seeding Method, Yanli Geng
Theses and Dissertations
In this dissertation, I developed a Pd seeding method in association with electroless plating, to successfully metallize both lambda DNA and DNA origami templates on different surfaces. On mica surfaces, this method offered a fast, simple process, and the ability to obtain a relatively high yield of metallized DNA nanostructures. When using lambda DNA as the templates, I studied the effect of Pd(II) activation time on the seed height and density, and an optimal activation time between 10 and 30 min was obtained. Based on the Pd seeds formed on DNA, as well as a Pd electroless plating solution, continuous …

Globoid cell leukodystrophy (GLD) is a neurological disease caused by deficiency of the lysosomal enzyme galactosylceramidase (GALC). In the absence of GALC, the cytotoxic glycosphingolipid, psychosine (psy), accumulates in the nervous system. Psychosine accumulation preferentially affects oligodendrocytes, leading to progressive demyelination and infiltration of activated monocytes/macrophages into the CNS. GLD is characterized by motor defects, cognitive deficits, seizures, and death by 2–5 years of age. It has been hypothesized that psychosine accumulation, primarily within lipid rafts, results in the pathogenic cascade in GLD. However, the mechanism of psychosine toxicity has yet to be elucidated. Therefore, we synthesized the enantiomer of …

Student Theses and Dissertations
The cell wall, one of the most complex and interesting organelle in a plant cell, still holds a mystery to be solved by the research community. In maize (Zea mays L.), the primary cell wall contains several structural proteins, the most abundant and well-studied being hydroxyproline-rich glycoproteins (HRGPs). These structural proteins give strength, flexibility during cell expansion and participate in the process of cell wall formation and assembly. Extensin is the dominant member of the HRGP family is accumulated in silk and pericarp of maize. We hypothesized that extensin comprises a small gene family in maize and that a specific …

The Type II Secretion System (T2SS) is a molecular machine that drives the secretion of fully-folded protein substrates across the bacterial outer membrane. A key element in the machinery is the secretin: an integral, multimeric outer membrane protein that forms the secretion pore. We show that three distinct forms of T2SSs can be distinguished based on the sequence characteristics of their secretin pores. Detailed comparative analysis of two of these, the Klebsiella-type and Vibrio-type, showed them to be further distinguished by the pilotin that mediates their transport and assembly into the outer membrane. We have determined the crystal structure of …

Triacylglyceride Profiles Of Integumentary Lipids Isolated From Three Bat Species Determined By Matrix-Assisted Laser Desorption/Ionization Time-Of-Flight Mass Spectrometry, Evan Lacy Pannkuk
Student Theses and Dissertations
Lipids secreted from sebaceous glands in bat integument may play a role in determining host pathogenicity by the fungus Geomyces destructans in White-Nose Syndrome (WNS). To investigate this, I have developed analytical methods to determine the triacylglycerol (TAG) profiles for three bat species: Eastern red bats (Lasiurus borealis, Müller, 1776), evening bats (Nycticeius humeralis, Rafinesque, 1818), and big brown bats (Eptesicus fuscus, Beauvois, 1796). Neutral lipids extracted from the hair and wing tissue were fractionated by preparative thin-layer chromatography (TLC) into four major lipid bands corresponding to cholesterol, free fatty acids (FFAs), TAGs, and sterol/wax esters. Densitometry showed higher proportions …
