A Novel Cyanopyridine Derived Fluorescent Sensor For Selective Determination Of Uranyl Ions In Different Water Samples, Ekram Hany Mohamed, Amany R Salem, Abeer M. El-Naggar,, Mohammed A. Amin,, Mohamed S. S. Attia

Pharmacy

A newly developed spectrofluorometric method is based on the fabrication of an optical cyanopyridine derived sensor, 4-(6-diaryl)-2-oxo -1,2-dihydropyridine-3-carbonitrile using microwave irradiation. The fluorescence intensity of the proposed chemo sensor was obviously quenched in DMSO owing to the presence of UO22+ ions at λex/λem = 320/360 nm. The quenching effect of the cyano-derivative based sensor was found to be proportional to the concentration of UO22+ ion ranging between 9.60 × 10–9–5.28 × 10–5 mol L−1 with a detection and quantification limits of 1.71 × 10–11 and 5.2 × 10–11 mol L−1, respectively. © 2021, Akadémiai Kiadó, Budapest, Hungary.

Stressors are threats perceived within the environment that activate a response within the …

Regulation Of Skeletal Muscle Plasticity By The Gut Microbiome, Taylor R. Valentino 2022 University of Kentucky

Regulation Of Skeletal Muscle Plasticity By The Gut Microbiome, Taylor R. Valentino

Theses and Dissertations--Physiology

Recent evidence suggests that the gut microbiome could play a role in skeletal muscle plasticity, providing novel treatments for muscle wasting diseases and/or performance enhancements. I first sought to determine if the gut microbiome is necessary for skeletal muscle adaptation to exercise. Forty-two, four-month old, female C57Bl/6J underwent nine weeks of weighted wheel running or remained in cage with a locked wheel, without or without the administration of antibiotics (treated). In response to wheel running, I found that antibiotic depletion of the microbiome led to a blunted hypertrophic response in the soleus muscle as measured by normalized muscle wet weight …

Tetramerization Of Stat5 Has Opposing Roles In Governing Autoimmune Pathogenesis, Kelly Lynn Monaghan 2022 West Virginia University

Tetramerization Of Stat5 Has Opposing Roles In Governing Autoimmune Pathogenesis, Kelly Lynn Monaghan

Graduate Theses, Dissertations, and Problem Reports

Signal transducers and activators of transcription 5 (STAT5A and STAT5B) are members of the STAT protein family. STAT5 plays a critical role in mediating cellular responses following cytokine stimulation. Critical STAT5 signaling is achieved following functional dimerization, which occurs via SH2 domain interactions. The binding of two STAT5 dimers to tandemly linked TTCN3GAA γ-interferon-activated sequence (GAS) motifs and N-terminal domain interactions, facilitates functional tetramerization. The biological role of STAT5 tetramers has been investigated using the Stat5a-Stat5b N-domain double knock-in (DKI) mouse strain, in which STAT5 tetramers cannot be formed due to substitutions in critical amino acid …

The Rotator Cuff Tendon-To-Bone Interface: Maturation, Aging, And 3d Bioprinting For Regeneration, Xiping Jiang

Theses & Dissertations

Rotator cuff tendon injuries often occur at the tendon-to-bone interface (i.e., enthesis) area with a high prevalence for the elderly population. In addition, regeneration of the gradient structure of the enthesis is still a significant clinical challenge. Our studies aim to identify the histological, molecular, and biomechanical alterations of the rotator cuff enthesis with maturation and aging, and develop a novel therapeutic method using three-dimensional (3D) bioprinting technique to regenerate a functional enthesis. Striking variations of the entheses were observed both histologically and biomechanically during the maturation process. The histological features did not show many differences at the insertion site …

4d Ex Vivo Crispr/Cas9 Whole-Genome Screen To Identify Genes Regulating Lung Cancer Metastasis, Alexandria Plumer

Dissertations & Theses (Open Access)

Metastatic lung cancer has a 5-year survival rate of 5%. Lung cancers tend to be asymptomatic until late stages, and almost 90% are not diagnosed until they are advanced. Metastases are very rare events, often initiated by a single cell from a primary tumor into a new niche at a distant location. Investigation of the early metastatic process is of urgent need for the development of early diagnostics and targeted therapeutics. We performed a proof-of-concept CRISPR/Cas9 whole genome knockout screen in the A549 lung adenocarcinoma cell line and utilized a novel ex vivo 4D lung metastasis model to find gene …

Differentiating The Mechanistic Role And Chemotherapeutic Potential Of Src And Podoplanin In Oncogenic Transformation, Edward P. Retzbach

Graduate School of Biomedical Sciences Theses and Dissertations

There were an estimated 20 million new cancer cases worldwide in 2020, resulting in nearly 1000 deaths per hour [1]. Oral cancer exemplifies the difficulties of treating cancer patients. The first line for oral cancer treatment is surgery and radiation that can lead to patient disfigurement and decreased quality of life in cancer survivors [2-4]. Though there have been many developments in chemotherapy in the last 30 years, the 50% mortality rate associated with oral cancer has not changed [4, 5]. Longitudinal studies that track survival rates in oral cancer patients demonstrate a 3-fold reduction in patient deaths when patients …

The Role Of The Erbb Signaling Pathway In Cardiovascular Progenitor Cell-Based Repair, Christopher Ramos 2021 California State University, San Bernardino

The Role Of The Erbb Signaling Pathway In Cardiovascular Progenitor Cell-Based Repair, Christopher Ramos

Electronic Theses, Projects, and Dissertations

Adult mammalian hearts lack self-renewal and proliferative capabilities necessary for cardiovascular regeneration. Current treatments using cardiac progenitor cells (CPCs) for cell-based repair do not restore cardiac function in patients who experience a myocardial infarction (MI). Our laboratory has been studying Islet-1+ neonatal CPCs as a promising candidate for cell-based repair due to their ability to significantly improve cardiac function after MI in sheep. The current study addresses the hypothesis that the ERBB pathway is linked to the Hippo-pathway to activate YAP1 by the involvement of an autocrine loop that upregulates neuregulin (NRG). In our sheep model of MI and cardiovascular …

Drivers Of Extracellular Polysaccharide Production By A Mat-Forming Diatom, Kaitlin A. Stansbury 2021 Florida International University

Drivers Of Extracellular Polysaccharide Production By A Mat-Forming Diatom, Kaitlin A. Stansbury

FIU Electronic Theses and Dissertations

Microbial biofilms are held together by extracellular polymeric substances (EPS), which can be secreted by many organisms. EPS production can facilitate intercellular communication and inter-guild microbial mutualisms, intraspecific gamete exchange, nutrient sequestration, and desiccation resistance. Benthic microbial mats (periphyton) of the Florida Everglades and other karstic wetlands contain abundant EPS generated by mat-producing filamentous blue-green algae and many other species of mat-dwelling algae, cyanobacteria, and bacteria, including the most abundant Everglades diatom, Mastagloia calcarea. The benthic diatom genus Mastogloia is characterized by several morphological and physiological features that foster production of a ‘halo’ of EPS around the frustule, but the …

Motor Unit Firing Rate Control Of Agonist Skeletal Muscle During Voluntary Isometric And Shortening Contractions With Limb Movement, Eric A. Kirk 2021 The University of Western Ontario

Motor Unit Firing Rate Control Of Agonist Skeletal Muscle During Voluntary Isometric And Shortening Contractions With Limb Movement, Eric A. Kirk

Electronic Thesis and Dissertation Repository

Voluntary movements throughout mammalian lifespan require skeletal muscle contractions that are controlled by time- and recruitment-dependent firing rate patterns. Single motor unit (MU) activity reflects the final efferent neural drive to the muscle, yet the underlying neural control of movement at the MU level is not well understood. Using intramuscular electromyography single MU recordings, relationships between voluntary contraction kinematics and MU firing rates were evaluated in vivo, in groups of young and older adult participants.

The purpose of study one was to characterize how MU firing rates are differently scaled among muscles relative to voluntary contraction intensity. Across 12 …

Exogenous Surfactant As A Delivery Vehicle For Intrapulmonary Therapeutics, Brandon J. Baer 2021 The University of Western Ontario

Exogenous Surfactant As A Delivery Vehicle For Intrapulmonary Therapeutics, Brandon J. Baer

Electronic Thesis and Dissertation Repository

As an organ system, the lung has unique advantages and disadvantages for direct drug delivery. Its contact with the external environment allows for the airways to be easily accessible to intrapulmonary delivery. However, its complex structure, which divides into more narrow airways with each branch, can make direct delivery to the remote alveoli challenging. The objective of this thesis was to overcome this issue by using exogenous surfactant, a lipoprotein complex used to treat neonatal respiratory distress syndrome, as a carrier for pulmonary therapeutics. It was hypothesized that therapeutics administered with a surfactant vehicle would display enhanced delivery to the …
