Background: Prolonged exposure to UVB radiation causes DNA damage in skin cells by raising the levels of reactive oxygen species, resulting in the production of inflammatory factors and skin issues. Plant extracts are frequently used to counteract photodamage due to their antioxidant properties. One example is the floral extract of the butterfly pea plant, which contains flavonoid antioxidants. However, the effect of the extract on inflammatory factors is unknown. This study investigated how tumor necrosis factor-alpha (TNF-α) and caspase-3 expression changed when a butterfly pea flower extract gel was applied topically to UVB-exposed animals.

Invasive plant species’ success may be a result of allelopathy, or the release of secondary metabolites that are harmful for surrounding plant species. Allelopathy can be mediated through the abiotic environment by chemical sorption or transformation, so the substrate on which interactions occur can lead to differential outcomes in allelopathic potential. One aggressive invader, Pyrus calleryana, has become dominant in many ecosystems throughout Eastern US, and has reduced the abundance of native species where it invades. Characterizing phages is becoming increasingly important in biochemistry and biotechnology due to their potential to treat bacterial infections without need for antibiotics. Last year, students attempted to isolate proteins from Microbacterium foliorum infecting phages Babydotz and Rosadiaz. Proteins were successfully extracted; however, after further analysis it was shown that the obtained proteins belonged to the host bacteria and not the phages. Carbonic anhydrases (CAs) are zinc-metalloenzymes that interconvert two inorganic carbon (Ci) species, CO2 and HCO3-. In Arabidopsis thaliana, there are eight αCA genes, six βCA genes, three γCA genes, and two γCA-like genes. The majority of CA research in plants has focused on finding a link between CA activity and photosynthesis rates. Since the CA genes are expressed in different plant tissues and multiple CA isoforms are distributed among various organelles of the plant cell, I hypothesize that CAs facilitate CO2 diffusion among cell compartments and maintain Ci pools for carbon-requiring reactions by interconverting …

The addition of cytokinin to the multiplication of white turmeric (Curcuma zedoaria) seedlings in vitro proved effective in shoot multiplication, but the addition of high cytokinin concentrations could cause stunted shoots and stunted growth. The addition of the hormone gibberellins (GA3) is often used in tissue culture for shoot elongation, so increasing the viability of plantlets. The experiment aims to determine the effect of GA3 and obtain the best concentrations of GA3 consisting of 0.50, 1, and 1.5 ppm GA3 on optimizing the growth of zedoary plantlets. The potential of whey from Italian cheese productions to support skin health and anti-ageing mechanism was studied. The effect of whey on dermal and epidermal cells was evaluated. Whey inhibited the activity of elastase and tyrosinase enzymes by 60% and 32%, respectively. Whey cytotoxicity against tested skin cell lines, human fibroblasts (HDFa) and keratinocytes (HaCaT) was not observed. Moreover, the antioxidant activity of the samples was noted, after treatment with whey intracellular ROS level was decreased by 87% in comparison with the hydrogen peroxide-treated cells. Brain glucose metabolism is highly heterogeneous among brain regions and continues postmortem. In particular, we demonstrate exhaustion of glycogen and glucose and an increase in lactate production during conventional rapid brain resection and preservation by liquid nitrogen. In contrast, we show that these post- mortem changes are not observed with simultaneous animal sacrifice and in situ fixation with focused, high- power microwave. We further employ microwave fixation to define brain glucose metabolism in the mouse model of streptozotocin-induced type 1 diabetes. Many plant essential oil, extracts and phytochemicals are known to possess toxic activity against Drosophila melanogaster. In the current work, the effect of aqueous leaves, flowers and seeds extracts of Peganum harmala on the fruit fly mortality and larval development were determined. The seeds extract was the most effective; 100 % of mortality was recorded for the highest concentrations (200 and 300 μg/ml) with lethal time values (LT50 and LT50) reached 3.72 to 4.47 and 6.61 to 8.51 days, respectively. Colonization with multidrug-resistant organisms (MDROs) is a risk factor for subsequent infection. Surveillance for MDROs, including methicillin-resistant Staphylococcus aureus, vancomycin-resistant enterococci, extended-spectrum beta-lactamase-producing Enterobacterales, and carbapenemase-producing organisms, is commonly conducted in hospitals to prevent spread of MDROs, in part to reduce the potential for additional infections. Although colonization is a risk factor for infection, data on colonization with various MDROs are often not considered when selecting anti-infective therapy. Purdue University has been a member of the SEA-PHAGES (Science Education Alliance-Phage Hunters Advancing Genomics and Evolutionary Science) program since 2011. This program, created in conjunction with the Howard Hughes Medical Institute (HHMI), facilitates undergraduate curriculum for the authentic discovery of novel bacteriophages within the classroom. Since joining the program, undergraduates at Purdue have harnessed wet lab and bioinformatics principles to contribute over 200 previously uncharacterized bacteriophages and 25 novel genomes to the wider scientific literature. The SEA-PHAGES classes at Purdue have resulted in tangible professional deliverables for students through conference presentations and publications. Leukocytes are in situ regulators critical for ovarian function. However, little is known about leukocyte subpopulations and their interaction with follicular cells in ovulatory follicles, especially in humans. Single-cell RNA sequencing (scRNA- seq) was performed using follicular aspirates obtained from four IVF patients and identified 13 cell groups: one granulosa cell group, one thecal cell group, 10 subsets of leukocytes, and one group of RBC/platelet. RNA velocity analyses on five granulosa cell populations predicted developmental dynamics denoting two projections of differentiation states. Armyworms are polyphagous insect pests of agricultural crops. Their population is usually managed below threshold level using insecticides. However, a single control measure is insufficient to attain a sustainable pest management.
