Alzheimer’s disease (AD) is a progressive brain disorder that gradually destroys an individual’s mental functioning and social capabilities, including the ability to carry out everyday activities. Although memory deficits affect AD patients’ ability to perform these activities, research suggests that visual perception impairments also contribute. One impaired visual perception ability, contrast sensitivity, enables one to distinguish an object from its immediate surroundings. The present project measured contrast sensitivity in a real-world task by having AD patients find a pill of various shades of gray on a tiled background. We report here that an epitope (aa, 83-95) derived from Acanthamoeba castellanii (ACA) induces clinical signs of experimental autoimmune encephalomyelitis (EAE) in SJL/J mice reminiscent of the disease induced with myelin proteolipid protein (PLP) 139-151. By using IAs/tetramers, we demonstrate that both ACA 83-95 and PLP 139-151 generate antigen-specific cross-reactive CD4 T cells and the T cells secrete identical patterns of cytokines and induce EAE with a similar severity. These results may provide insights into the pathogenesis of multiple sclerosis and ACA-induced granulomatous encephalitis.

Cholesterol is an essential component of both the peripheral and central nervous systems of mammals. Over the last decade, evidence has accumulated that disturbances in cholesterol metabolism are associated with the development of various neurological conditions. In addition to genetically defined defects in cholesterol synthesis, which will be covered in another review in this Thematic Series, defects in cholesterol metabolism (cerebrotendinous xanthomatosis) and intracellular transport (Niemann Pick Syndrome) lead to neurological disease. A subform of hereditary spastic paresis (type SPG5) and Huntington's disease are neurological diseases with mutations in genes that are of importance for cholesterol metabolism. The most difficult people to disclose this information to are most often those who are also at-risk, notably the children of individuals who are themselves at-risk for developing the disease or know that they will develop the disease.
