The Ush2a Gene: An Analysis Of Ultrasonic Vocalizations In A Mouse Model Of Usher Syndrome Type 2, Kiana R. Akhundzadeh
Honors Scholar Theses
Usher syndrome type 2 is a complex autosomal recessive genetic disorder that is characterized by moderate to severe congenital sensorineural hearing loss, the onset of retinitis pigmentosa in the second decade of life, and in some cases, vestibular dysfunction. Mutations in the USH2A gene account for 85% of cases of type 2. The USH2A gene is responsible for encoding the protein usherin, which has an important role in the development and function of inner ear hair cells and retinal photoreceptors. Until recently, it has been believed that carriers of the USH2A mutation were phenotype free. However, recent data has suggested …

Dissertations and Theses (Open Access)
Cancer-associated mortality has been declining for two decades but remains a significant public health problem, especially when patients initially present with advanced disease. Early detection methods have improved survival rates but remain unavailable for a majority of cancers due to a lack of sensitive biomarkers or numerous limitations associated with current diagnosis strategies. Approaches to develop “liquid biopsies” by detecting tumor cells or DNA in the blood have led to several breakthroughs and create the potential for non-invasive, routine assessment of diseases status. However, these biomarkers are rare and currently difficult to isolate, especially in the early stages of disease. …

Undergraduate Theses
Chalcones, a precursor to flavonoids, are chemical compounds found naturally in plants. The chalcones’ structure consists of a ketone bridge attached to two aromatic rings. Varying substituents on the aromatic rings allow for different affects, including anti-cancer properties. As a Michael acceptor, chalcones interact with pathways that cause inhibition of the initiation, promotion, and progression of cancer tumors. We have screened 32 compounds for growth inhibition in lung cells that vary the flexibility and confirmation of the 3 carbon bridge between the two aromatic rings as well as the effects of electronic modifications to the aromatic ring. We have found …

Taxanes are spindle poisons that bind to and stabilize microtubules resulting in mitotic arrest. Herpes simplex Typ-1 (HSV-1) virions utilize the microtubular network for intracellular transport during both virus entry and virus egress from infected cells. It has been reported previously that taxanes may synergize with oncolytic herpes simplex viruses in the treatment of experimental prostate and breast tumors in mice. Other reports have indicated that taxanes may inhibit viral replication in infected cells. In this study the previously characterized Oncolytic Herpes Simplex Virus type 1 (OSVP), which was constructed in Kousoulas lab was used in conjugation with paclitaxel (taxol) …

Contaminated water sources can cause problems for scientific research and result in costly delays and failures of experiments. At Bowling Green State University, the reverse osmosis supply circulating in the Life Sciences Building has been measurably contaminated for nearly three years, corresponding to a change in servicing of the system. While servicing has been accelerated, the contamination in the system remains. The focus of this research was to identify the species of bacteria and fungi growing inside of the water system so that it might alert those servicing the system, and to begin to eliminate the contamination. Reverse osmosis water …

Background: Bacterial lung infections are leading causes of death worldwide. Unfortunately, increasing resistance to antibiotics and the inflammation often accompanying these infections are leading to poor outcomes despite antibiotic intervention. Complicating treatment further, the tree-like branching structure of the lung makes drug delivery to distal sites of infection difficult. Our research aims to address these challenges by developing new therapeutics and new tools to improve and assess drug delivery, bacterial killing and inflammation. Our therapy combines host defense peptides, which have been shown to kill antibiotic-resistant bacteria and down regulate inflammation, with a pulmonary vehicle, exogenous surfactant, that can improve …

The Mississippi River watershed is the third largest in the world and drains from 31 states in the United States. Due to its route through the U.S., it passes through hundreds of EPA labeled Superfund sites as well as agricultural and suburban watersheds. These sites are known to have historical chemical contamination, and as a result chemicals can be transported by runoff down the Mississippi River into the Gulf of Mexico where their ultimate fate is unknown. Organochlorine chemicals are known to be persistent and are found in agricultural land as well as US EPA Superfund sites, especially DDT, PCBs, …

Smoothened receptor (SMO) is a protein that in humans, is encoded by the SMO gene. A systemic mutation in its binding pocket helps predict the sensitivity of mutant proteins to different drugs. Known as a GPCR-like receptor, it is a component of the hedgehog signaling pathway; a pathway involved in body patterning and the regulation of adult stem cells. An uncontrolled or inappropriate activation of the Hedgehog pathway drives tumor progression in cancers and a number of birth defects. To achieve these goals, the molecular modeling software MOE was used to build small molecules and drug molecules like Vismodegib and …

Elucidating the biology of yeast in its full complexity has major implications for science, medicine and industry. One of the most critical processes determining yeast life and physiology is cellular demise. However, the investigation of yeast cell death is a relatively young field, and a widely accepted set of concepts and terms is still missing. Here, we propose unified criteria for the definition of accidental, regulated, and programmed forms of cell death in yeast based on a series of morphological and biochemical criteria. Specifically, we provide consensus guidelines on the differential definition of terms including apoptosis, regulated necrosis, and autophagic …

Growth Rate And Hatch Date Of Age-0 Fishes On Leech Lake, Minnesota Before Zebra Mussel Establishment, Kayla Morey, Emily Larson
Journal of Earth and Life Science
The introduction of an invasive species to an ecosystem is considered a very severe environmental problem. Zebra Mussel veligers Dreissena polymorpha have recently been found in Leech Lake, Minnesota, and are likely going to have a large influence on this ecosystem. Zebra Mussels are filter-feeders that rely on phytoplankton and zooplankton as their main source of energy. A shift in populations of plankton due to Zebra Mussels may affect the growth rates of larval fish. Median hatch dates and growth rates were estimated from four age-0 fish species (Yellow Perch Perca flavescens, Walleye Sander vitreus, Smallmouth Bass Micropterus …

Age-0 Walleye Growth Rates Following Spiny Water Flea Bythotrephes Longimanus Invasion, Joseph Amundson
Journal of Earth and Life Science
Within Voyageurs National Park, spiny water flea Bythotrephes longimanus has reduced Lake Kabetogama and Rainy Lake native zooplankton biomass during summer peak by 40-60%. Subsequently, planktivorous age-0 Yellow Perch Perca flavescens growth has decreased. A possible trophic cascade may influence predatory fish growth rates like that of age-0 Walleye Sander vitreus. Using seine net and electrofishing data, age-0 Walleye growth rates were modeled as a linear function of growing degree days (GDD) and the slope was compared between pre- and post-spiny water flea establishment. Nearby Lake Vermilion, assumed to have been unaffected by spiny water flea until 2015 and …

Burbot Lota lota is the only freshwater member of the family Gadidae and has a circumpolar distribution in cold-water regions. Information related to Burbot biology is limited in comparison to most fishes. Snow pack depth and ice conditions influence light penetration and therefore primary production levels. Primary production can have cascading effects on higher trophic levels. Therefore, the objective of this study was to observe if light levels penetrating through cleared and covered ice has an effect on Burbot activity in Lake Bemidji, MN. Angling for Burbot occurred for a minimum of 30 four hour sample periods at both cleared …

Lorcaserin As A Potential Opioid-Sparing Adjunct, Kumiko M. Lippold
Theses and Dissertations
Opioids, such as oxycodone, morphine, and fentanyl, are commonly used medications in the treatment of moderate to severe pain. In spite of their efficacious analgesic properties, their increased prescribing rates by physicians and inherent abuse-related effects have led to the ongoing opioid epidemic. Their clinical utility is limited by the risk of adverse dose-dependent side effects, such as constipation and respiratory depression, and the development of tolerance and dependence. Opioid-sparing adjunctive therapies are sought to address these issues by reducing the dose of opioid needed to achieve analgesia through alternative non-opioidergic mechanisms and as a result, reduce the incidence of …

Effectiveness Of A Physical Activity Enhancing Program For Use During Indoor Recess, Cassandra M. Blase
Theses and Dissertations--Kinesiology and Health Promotion
Purpose: The purpose of this study was to determine if a novel physical activity intervention game (Bingocize®) designed for use in confined spaces and modified to include age appropriate activities would increase the time spent engaged in physical activity (PA) during indoor recess. Methods: Fifty-two third grade children wore triaxial accelerometers during three different recess conditions. The recess conditions included: “typical” indoor recess (TIR), indoor recess with children engaged in Bingocize® (IRB), and “typical” outdoor recess (TOR). Results: There were significant (p < 0.05) differences among the recess conditions for the time spent in sedentary, light, moderate-to-vigorous physical activity (MVPA) intensity categories, as well as PA counts, and steps. During IRB, TIR, and TOR the subject were sedentary 42.4% (±0.10), 71.9%(±0.10), and 17.5% (±0.10) of the recess time, respectively. During IRB, TIR and TOR the subjects spent 43.4%(±0.10), 18.5%(±0.10), and 74.2%(±0.12) engaged in MVPA, respectively. Conclusions: These results indicate that Bingocize® promotes increased PA during times when inclement weather necessitates indoor recess and larger space requirements for activity are not available.

Copepod Distribution In Relation To Environmental Parameters On Diel And Tidal Time Scales In Mission Bay, San Diego, California, Joy Renee Shapiro
Theses
The vertical distribution of copepods in estuaries is known to vary in relation to environmental factors. However, the relationships between environmental conditions (e.g., tides, hydrography) and copepod distributions are not well understood. This project examined connections between environmental parameters and copepod distribution in Mission Bay, San Diego, California. Copepods (adults, juveniles, and nauplii) were collected every two hours over a diel cycle at three sites across the bay. A plankton pump was used to draw ~2 m3 of water from each of two depths - just below the surface and just above the bottom. Copepods were retained in a …

The Wisconsin State Laboratory of Hygiene challenged Wisconsin laboratories to examine their biosafety practices and improve their culture of biosafety. One hundred three clinical and public health laboratories completed a questionnaire-based, microbiology-focused biosafety risk assessment. Greater than 96% of the respondents performed activities related to specimen processing, direct microscopic examination, and rapid nonmolecular testing, while approximately 60% performed culture interpretation. Although they are important to the assessment of risk, data specific to patient occupation, symptoms, and travel history were often unavailable to the laboratory and, therefore, less contributory to a microbiology-focused biosafety risk assessment than information on the specimen source …
