Purpose: Advancements in sequencing technologies have significantly improved clinical genetic testing; yet, the diagnostic yield remains around 30% to 40%. Emerging technologies are now being deployed to address the remaining diagnostic gap.

Methods: We tested whether short-read genome sequencing could increase the diagnostic yield in individuals enrolled into the UCI-GREGoR research study, who had suspected Mendelian conditions and prior inconclusive testing. Two other collaborative research cohorts, focused on aortopathy and dilated cardiomyopathy, consisted of individuals who were undiagnosed but had not undergone harmonized prior testing.

Radiotherapy is one of the most important adjuvant treatment methods for gastric cancer (GC). However, radioresistance remains a major clinical obstacle. In this study, APOL2 is identified as a key player in promoting non-homologous end joining (NHEJ)-mediated double-strand break (DSB) repair and enhancing radioresistance in GC. Bioinformatics and clinical data revealed that high APOL2 expression is correlated with poor prognosis in GC patients. Functional experiments showed that APOL2 overexpression enhances genomic stability by accelerating DSB repair via the NHEJ pathway, while APOL2 knockout impairs repair capacity. Background: Targeted radionuclide therapy (TRT) has emerged as a unique and effective treatment modality for cancer. Monte Carlo simulations have greatly advanced investigations into radiation-caused DNA damage, including the complexity of this damage.

Purpose: This study aimed to evaluate DNA damage induced by high-linear energy transfer therapeutic radionuclides used in TRT, specifically 225Ac, 177Lu, and 212Pb, using Geant4-DNA Monte Carlo simulations.

Rising soil salinity and water scarcity demand sustainable solutions to maintain crop productivity. This study evaluated four agro-industrial organic amendments—fish emulsion, neem cake, coffee grounds, and seaweed extract on alfalfa (Medicago sativa) under combined salinity and drought stress, assessing their effects on soil respiration, plant physiology, and yield. Soil CO₂ efflux varied significantly among treatments: coffee grounds induced the highest microbial activity (6.2 μmol m⁻² s⁻¹), suggesting rapid decomposition, while neem cake exhibited stable emissions (3.8 μmol m⁻² s⁻¹), indicating slower organic matter turnover. Multilevel selection (MLS) is a foundational framework that expands the concept of natural selection beyond the traditional focus on individuals or genes to larger units, like groups. Despite strong empirical support and applications in a broad range of biological fields, MLS, often referred to as group selection, has a controversial history, which has impeded its wide use and led to incorrect inferences on the sources and strength of selection. Highly pathogenic avian influenza (HPAI) is an extremely transmissible virus that poses a significant threat to the U.S. turkey industry through contagion from migratory birds and results in whole-farm depopulation. To combat the disastrous economic impacts of HPAI, this study examined how biosecurity investments may have economic benefits to producers and the industry. Using data collected from a farm-focused survey of commercial turkey producers, the differences in biosecurity practices between farms affected by HPAI and unaffected farms in 2022 were analyzed. Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by complex interactions between genetic risk factors and structural brain changes. Traditional diagnostic approaches that rely on single-modality data, such as imaging or genomics alone, often fall short in both predictive accuracy and biological interpretability. To address these limitations, AlzCLIP, a novel contrastive learning framework that integrates single nucleotide polymorphism (SNP) profiles and MRI-derived imaging features into a unified embedding space is introduced. RNA modification, which is evolutionarily conserved, is crucial for modulating various biological functions and disease pathogenesis. High resolution transcriptome-wide mapping of RNA modifications has facilitated both data resources and computational prediction of RNA modification. While these prediction algorithms are promising, they are limited in interpretability or generalizability, or the capacity for discovering novel post-transcriptional regulations. Here, we present NetRNApan, a deep learning framework for RNA modification site prediction, motif discovery and trans-regulatory factor identification. Two experiments were conducted to determine relative Ca bioavailability based on bone ash and apparent ileal digestibility (AID) of Ca in reagent-grade calcium carbonate (RCaCO3) and RCaCO3 + phytic acid in commercial broiler chickens. In Experiment 1, relative Ca bioavailability of RCaCO3 and RCaCO3 + phytic acid was determined in ad libitum-fed broiler chickens using the slope-ratio method. Dietary treatments were corn-soybean meal-based diets. This bioinspired study explores the fascinating realm of clean and green chemistry by unveiling the potential of FeO NPs fabricated by taking advantage of botanical extract obtained from the leaves of Justicia adhatoda as a reducing agent because it reduced Fe (II) to Fe (0). Background: Salinity represents a major global constraint on crop productivity. Promoting the cultivation of tall fescue in saline environments offers not only nutritional advantages for livestock but also enhances its potential for ornamental use. In this mesocosm study, we examined the effects of paclobutrazol (PBZ) on tall fescue performance under salt stress, focusing on key physiological traits to evaluate salt tolerance.

Results: Under high salt stress, paclobutrazol application increased the total number of lateral roots by 85%, from 18.39 to 34.04, and widened their growth angle by 24%, from 24.10° to 29.88°, fundamentally enhancing topsoil exploration. This reconfigured root system …

Structural brain differences in the thalamus and the cortex have been widely reported in schizophrenia (SCZ) relative to neurotypical control individuals (NCs). Most previous studies examined the thalamusas a whole as a single region of interest. In addition, findings in individuals at familial high risk for SCZ (FHRs) remain inconclusive. Objective: To examine the effects of incorporating (1) a neurocognitive reactive component and (2) a neurocognitive multitask component on performance degradation of a single-limb hop functional performance test.

Design: Randomized within-subject design of 32 healthy young adults.

Methods: Participants performed 3 randomly assigned variations of the single-limb T-Drill Hop Test (TDHT). The time to complete each test was recorded. The reactive TDHT (R-TDHT) consisted of the TDHT with a flashing light, indicating the "T" intersection hop direction. Embryonic Transcription Factors (TFs) are often reactivated in cancer, driving developmental gene programs that support phenotypic plasticity. Metabolic adaptation fuels this plasticity by supplying energy and molecular building blocks for growth. RUNX2, the master regulator of bone morphogenesis, is ectopically expressed in epithelial cancer, promoting metastasis through trans-differentiation processes like Epithelial-to-Mesenchymal Transition (EMT) and osteomimicry. By combining omics data with functional validation, we demonstrated that RUNX2 drives cancer cell metabolic rewiring by repressing mitochondrial respiration while promoting anabolic processes. Abdominal obesity is a condition characterized by excessive fat accumulation in the abdominal region, which can increases the risk of type 2 diabetes mellitus, dyslipidemia, hypertension, and cardiovascular diseases. In 2023, Riau Islands Province recorded the highest prevalence of abdominal obesity in the island of Sumatra, at 42.5%. This study aimed to analyze the determinants of abdominal obesity among adults aged 19–44 years in Riau Islands Province based on gender, using secondary data from the 2023 Indonesia Health Survey (SKI). The prevalence of chronic energy deficiency (CED) among pregnant women in Indonesia remains relatively high, ranging from 11.6% to 57.6%. This study aimed to examine the differences in the proportion of CED based on macronutrient intake and related factors. The study used a quantitative cross-sectional design with secondary data involving 124 pregnant women in selected areas served by community health centers in Bogor City and Regency. In sweetpotato, global production exceeds 90 million tons annually, yet yields are threatened by numerous pathogens, including the potyvirus sweet potato feathery mottle virus (SPFMV). Virus infections can reduce yields by 25–40%, but surveillance is hampered by asymptomatic infections and the labor-intensive nature of detection. Hyperspectral imaging (HSI), combined with machine learning (ML), offers a promising pathway to scalable, non-destructive detection; however, studies focused on virus infection in sweetpotato remain limited.

Ungrading is a pedagogical approach that, in some iterations, emphasizes formative feedback and student self-assessment over traditional point-based grading. While increasingly popular in the humanities and social sciences, ungrading remains uncommon in STEM courses, particularly those that are content-intensive. This article presents a structured example of ungrading implemented in an upper-division ecosystem ecology course through a 7-week independent research project. Students used public or previously collected ecological data to investigate a scientific question, culminating in a short-format, manuscript-style research paper and a 10 min oral presentation. BACKGROUND: Recent randomized controlled trials have established chemoimmunotherapy as the standard of care for patients with advanced biliary cancers with a median overall survival (OS) of about thirteen months. No data exist to demonstrate the safety and efficacy of liver-directed radiotherapy (RT) for extrahepatic and intrahepatic cholangiocarcinoma (CCA) in the era of chemoimmunotherapy. The purpose of this study is to report our early experience treating patients with CCA using RT and chemoimmunotherapy.

Although dietary counseling is essential in managing obesity, maintaining a calorie deficit and preventing weight regain remain difficult. Functional foods like high-protein, high-fiber meal replacements may enhance adherence and support weight loss. This randomized controlled trial evaluated the subjective outcomes of a weight loss program incorporating a protein- and fiber-enriched meal replacement among 73 overweight women aged 20–30 years. The objective of this study was to examine the socio-demographic determinants of tobacco use among Bangladeshi men, focusing on regional variation, education, media exposure, and family influence. This study investigates post-fire vegetation transitions and spectral responses in the Snowstorm Fire (2017) and South Sugarloaf Fire (2018) in Nevada using Landsat 8 Operational Land Imager (OLI) surface reflectance imagery and unsupervised ISODATA classification. By comparing pre-fire and post-fire conditions, we have assessed changes in vegetation composition, spectral signatures, and the emergence of novel land cover types. The results revealed widespread conversion of shrubland and conifer-dominated systems to herbaceous cover with significant reductions in near-infrared reflectance and elevated shortwave infrared responses, indicative of vegetation loss and surface alteration. This paper explores the results of introducing affixes to develop vocabulary and increase motivation for beginner to intermediate-level English language learners (ELLs) pursuing academic disciplines in law, science, and technology at a New York City community college.
