RNA-sequencing (RNA-seq) technology allows for the identification of differentially expressed genes, which are genes whose mean transcript abundance levels vary across conditions. In practice, RNA-seq datasets often include covariates that are of primary interest in addition to a set of covariates that are subject to selection. Some of these covariates may be relevant to gene expression levels, while others may be irrelevant. Ignoring relevant covariates or attempting to adjust for the effect of irrelevant covariates can compromise the identification of differentially expressed genes. Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants widely detected in drinking water and food sources, resulting in chronic human exposure. Among these compounds, hexafluoropropylene oxide dimer acid (HFPO-DA), commonly known as GenX, has been introduced as a short-chain replacement for legacy PFAS such as perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). Although GenX is believed to be a safer alternative, emerging research suggests it can still affect human health.

Membrane trafficking is frequently disrupted during cancer progression, and the underlying mechanisms remain largely unknown. Currently, no effective drugs target dysregulated membrane trafficking for cancer treatment. Recent evidence has demonstrated that epithelial-to-mesenchymal transition (EMT) in lung adenocarcinoma (LUAD) employs a membrane trafficking program to coordinate cancer cell invasion and immunosuppression. To further dissect the pro-tumorigenic membrane trafficking program, we initiated an in vivo CRISPRi screen to assess more than 2,000 membrane trafficking-related genes in a syngeneic mouse LUAD model. Membrane trafficking plays a critical role in cellular function and is frequently dysregulated in cancer to promote metastasis. In lung adenocarcinoma (LUAD), the epithelial-to-mesenchymal transition (EMT) activating transcription factor, ZEB1, drives a pro-metastatic membrane trafficking program; however, the underlying molecular mechanisms remain poorly understood. Using a CRISPR interference (CRISPRi) in vivo screen of 2,099 membrane trafficking regulators in a syngeneic mouse model of EMT-driven LUAD, we identified REEP2 — an endoplasmic reticulum (ER) shaping protein — as a critical regulator of tumor progression. REEP2 mRNA expression correlates with poor prognosis, EMT signatures, and an immunosuppressive tumor microenvironment in LUAD patients. …

Base excision repair (BER) mechanisms fix single base lesions in DNA, such as T:G mismatches. During the base excision repair mechanism, an abasic site (AP site) is formed as an intermediate. AP sites are unstable and highly mutagenic; they can stop DNA replication. This research investigates how the conformational properties of abasic sites in DNA affect the binding recognition of enzymes involved in DNA repair mechanisms, including BER. Three different abasic sequences are being analyzed for this research project. A second project looks at the effects of a naturally occurring purine derivative, hypoxanthine, on the DNA backbone. After over 100 years of Zn and Pb mining in the Tri-State mining district (TSMD), former mines continue to be sources of metals. Metal contaminated soils can be remediated using plant-microbe interactions. Plants manipulate their microbiome and recruit microbes to increase metal tolerance. Remediating bacteria are site-specific and identifying native microbes can accelerate remediation efforts. Studies relating microbes and metal concentrations in remediated areas in the TSMD are scarce. Sustainable agriculture is a forefront of innovation compared to conventional agriculture. Living mulches, non-cash crops grown alongside cash crops, have demonstrated weed suppression and impact on the promotion of sustainable agriculture. Property taxes are the primary revenue instrument of local governments in the United States, yet the burdens they impose are distributed unevenly. Farmland, because it cannot be relocated, often absorbs a disproportionate share. This paper examines two related questions: what fiscal factors determine how heavily agricultural land is taxed, and do those tax rates capitalize into farmland asset values? Using an annual panel of Iowa, Illinois, Indiana, Kansas, Minnesota, Nebraska, and Wisconsin from 2009 to 2022, we first document how each state allocates property tax revenue across four major spending categories (education, public welfare, health, and utilities) finding that all …

Many neurodevelopmental disorders occur due to perturbations in prenatal brain development. More genetic variants associated with neurodevelopmental disorders are continuously being identified. However, there is a lack of understanding of the pathogenic mechanisms. Here, we describe a novel application of the technique Breasi-CRISPR in modeling specific patient variants in the embryonic mouse brain. Breasi-CRISPR is a highly efficient system combining in utero electroporation with CRISPR-CAS9 genome editing. We hypothesized that Breasi-CRISPR would be sufficient at inserting patient analogous variants into endogenous loci to be used as a new paradigm for modeling neurodevelopmental disorders. Toxic metal contamination in aquatic environments remains a persistent threat to human health and ecosystems. Yet, the high cost, infrastructure demands, and centralized nature of conventional analytical methods constrain routine monitoring. Ion-imprinted polymers (IIPs), a subclass of molecularly imprinted polymers, have emerged as promising synthetic recognition materials for metal-ion sensing because they generate coordination-defined binding sites with high selectivity, chemical stability, low cost, and reusability. Social stress is important for evaluating threats and resource opportunities, however, chronic exposure initiates the onset of psychological disorders, such as anxiety, depression, and post-traumatic stress disorder (PTSD). Stress responsiveness varies between individuals and sexes, resulting in disproportionate diagnoses and ineffective treatment outcomes. Enhancing therapeutic efficacy requires understanding of stress responsivity, phenotype development, and neurocircuitry modifications. Key contributors to stress responsiveness, such as threat of aggression and motivational initiation, influence the development of resilient or vulnerable phenotypes through behavioral and molecular adaptations to stress-related neurocircuitry. Background: Placenta accreta spectrum (PAS) involves abnormal placental invasion into uterine wall and is associated with life-threatening maternal complications. Doppler ultrasound is the primary diagnostic tool, though its ability to determine invasion depth and predict outcomes remains uncertain. Multidisciplinary management improves safety, and treatment often requires planned cesarean hysterectomy, or conservative management. Objective: to assess the correlation between blood levels of vascular endothelial growth factor (VEGF) and microvascular changes identified by nail fold videocapillaroscopy (NVC) in individuals with systemic sclerosis. Methods: A videocapillaroscope was used to evaluate the microvascular alterations in nail folds in SSc patients. The ELIZA technique was used to measure serum vascular endothelial growth factor (VEGF). Results: Nailfold videocapillaroscopy reveals significant microvascular alterations in patients with systemic sclerosis when compared to controls. Patients' capillary breadth, length, and diameter have increased but their capillary numbers have dramatically decreased. 75% of patients have hemorrhages, although controls do not. Alzheimer’s disease (AD) is a prominent neurodegenerative disease characterized by a decline in cognitive functioning. While current FDA-approved therapeutics exist, they fall short in creating significant and long-lasting improvements to cognition. A newer approach using neurotrophic factors has shown promising effects on improving cognition in clinical trials. Neurotrophic factors are secreted proteins that have multimodal effects in the brain, including supporting cell survival and functioning. Riparian ecosystems provide habitat for birds throughout their annual cycle, yet flow regulation along major rivers like the Missouri River has reduced early successional riparian forests. Following historic flooding in 2011, set-aside areas within the Missouri National Recreational River (MNRR) were established to allow natural riparian vegetation succession on newly created sandbars. I evaluated the value of these early successional forests for birds over three breeding seasons (2018–2020) and four migration periods (2020–2021). During migration, standardized point counts documented 7,462 individual birds representing 114 species across 828 surveys. Objectives: To assess the surgical outcomes among laryngotracheal stenosis patients who underwent tracheal/crico-tracheal resection anastomosis for with application of BloodSTOP iX on the anastomotic line as well as the operative field.
Methods: Fourteen patients participated in this prospective study. The stenotic airway segment was resected circumferentially using primary end-to-end anastomosis. The authors of this article used the same standardized surgical techniques described in the literature and used by them previously. All patients had suprahyoid release, and trachea-hyoid de-tensioning stitches were inserted to reduce the anastomotic tension. Background: Anorectal malformations (ARMs) encompass a broad range of congenital defects and are frequently associated with genitourinary (GU) anomalies, which may contribute to long-term morbidity if undetected. This study aimed to determine the prevalence and spectrum of GU anomalies in children with ARMs, describe the urological and genital findings, and assess associations with sex and ARM type.
Patients and Methods: A prospective survey enrolled children diagnosed with ARMs at a tertiary pediatric surgery center. Background: Traumatic brachial plexus injuries (TBPIs) cause severe upper limb disability. Owing to the complex anatomic architecture of the brachial plexus (BP), TBPIs are difficult to diagnose. Magnetic resonance neurography of the BP using three-dimensional isotropic imaging provides excellent background fat suppression and adequate soft-tissue contrast, thereby allowing better delineation of the BP along its pathway. It is well known that publications with collaborators from external institutions increase citations. This effect scales with spatial distance. There are also many barriers to long-distance collaborations, including linguistic differences, funding constraints, and the incremental costs of remote collaboration.
