Ecoimmunology examines the relationship between the physiology of an organism, its immune responses, and the external environment. As such, this field is critical to answering important questions in ecology, physiology, and evolution, and it can be vital in making policy and conservation decisions. As the only ectothermic amniotes, class Reptilia represents a group of unique study organisms. However, in comparison to other vertebrate taxa, this group has remained relatively understudied in the field of ecoimmunology. Phytophthora sojae is a soil-borne oomycete and the causal agent of Phytophthora stem and root rot (PSR) in soybean (Glycine max [L.] Merrill). Yield losses attributed to P. sojae are devastating in disease-conducive environments, with global estimates surpassing 1.1 million tonnes annually. Historically, management of PSR has entailed host genetic resistance (i.e., vertical and horizontal) complemented by disease-suppressive cultural practices (e.g., oomicide application). However, the vast expansion of complex and/or diverse P. sojae pathotypes complicated by climate instability necessitates the development of novel technologies for the attenuation of PSR in field environments. During pregnancy, the fetus relies on the dam for its nutrient supply. Nutritional stimuli during fetal organ development can program hepatic metabolism and function. Herein, we investigated the role of vitamin and mineral supplementation (VTM or NoVTM—at least 71 days pre-breeding to day 83 of gestation) and rate of weight gain (low (LG) or moderate (MG)—from breeding to day 83) on the fetal liver transcriptome and the underlying biological pathways. Over the past half century, birds have changed, including in their morphology. Biogeographic rules including Bergmann’s and Allen’s rules have been used to try to elucidate morphological changes; however, there has been dispute over the rules’ applicability. Thus, I aimed to identify 1) morphometric trends over time of 15 North American bird species, including eastern bluebirds (Sialia sialis); 2) if these morphometric trends follow Bergmann’s rule; and 3) if the observed morphometric trends are associated with life history and other traits, as well as climatic variables (e.g., average, maximum and minimum temperatures, and total precipitation). In the realm of immunology, the use of reptilian models remains low in comparison to their mammalian counterparts. Consequently, much of the inner workings of reptilian immunity remain to be elucidated. Specifically, chelonian species are vastly understudied. In the past decade, research focusing on reptilian immunology has increased. This thesis seeks to contribute to the growing body of work concerning immune trade-offs. I studied reptilian innate immune ability in both captive and free-living populations with consideration of both intrinsic and extrinsic factors. Ceriodaphnia dubia is an EPA-recommended organism used in regulatory toxicity testing of surface waters and commercial chemicals. They are often cultured for extended periods in the laboratory with little knowledge of how it impacts subsequent generations. This thesis investigated if C. dubia cultured for an extended period were representative of three recently-collected field populations by comparing their culturing characteristics and sensitivities to toxicants. Comparative chronic toxicity tests with sodium chloride, the neonicotinoid insecticide thiamethoxam, and mixtures of the two toxicants measured differences in sensitivities among the laboratory and field cultures, specifically in the variability of responses. Continued increases in nitrogen and phosphorus loads in waterways cause problems with water quality, impacting human health, the economy, and the environment. To reach nutrient load reduction targets, innovative conservation strategies should be evaluated. Application of the right time and amount of fertilizer N is challenging in rice fertilization because crop N supply is highly variable in the field. Research plots were established at four research plot fields and one commercial field in Arkansas. Independent variables included six nitrogen (N) application rates (0, 67, 101, 134.5, 168, and 202 kg N ha-1), multiple rice cultivars, and five locations. The response variable was 24 vegetative indices values using high spatio-temporal multispectral and thermal imagery obtained from an Unmanned Aerial Vehicle (UAV). Central metabolism produces amino and fatty acids for protein and lipids that establish seed value. Biosynthesis of storage reserves occurs in multiple organelles that exchange central intermediates including two essential metabolites, malate, and pyruvate that are linked by malic enzyme. Malic enzyme can be active in multiple subcellular compartments, partitioning carbon and reducing equivalents for anabolic and catabolic requirements. Prior studies based on isotopic labeling and steady-state metabolic flux analyses indicated malic enzyme provides carbon for fatty acid biosynthesis in plants, though genetic evidence confirming this role is lacking. The polymer, polyacrylamide (PAM), is a high molecular weight, chemical anionic polymer that is prescribed and used for irrigation improvement in row crops primarily in the United States. A field research study was conducted in 2014, 2015 and 2016 in Northeast Arkansas to evaluate how addition of PAM in furrow irrigation affected soil erosion, water infiltration, movement of nutrients and sediments, and crop yields in cotton (Gossypium hirsutum). Overall results from the study were mixed. Sediment loss was similar among treatments. Irrigation advancement rates were not significantly different. No differences in crop yield were observed associated with PAM. Differences in …

Microbial symbionts range from mutualistic to commensal to antagonistic. While these roles are distinct in their outcome, they are also fluid in a changing environment. Here, we used the Lotus japonicus–Mesorhizobium japonicum symbiosis to investigate short-term and long-term shifts in population abundance using an effective, fast, and low-cost tracking methodology for M. japonicum. We use quantitative polymerase chain reaction (qPCR) to track previously generated signature-tagged M. japonicum mutants targeting the Tn5 transposon insertion and the flanking gene. Regulation of RNA processing contributes profoundly to tissue development and physiology. Here, we report that serine-arginine-rich splicing factor 1 (SRSF1) is essential for hepatocyte function and survival. Although SRSF1 is mainly known for its many roles in mRNA metabolism, it is also crucial for maintaining genome stability. We show that acute liver damage in the setting of targeted SRSF1 deletion in mice is associated with the excessive formation of deleterious RNA-DNA hybrids (R-loops), which induce DNA damage. Combining hepatocyte-specific transcriptome, proteome, and RNA binding analyses, we demonstrate that widespread genotoxic stress following SRSF1 depletion results in global inhibition of mRNA …

The discovery of monokinase-selective inhibitors for patients is challenging because the 500+ kinases encoded by the human genome share highly conserved catalytic domains. Until now, no selective inhibitors unique for a single transforming growth factor β (TGFβ) family transmembrane receptor kinase, including bone morphogenetic protein receptor type 2 (BMPR2), have been reported. This dearth of receptor-specific kinase inhibitors hinders therapeutic options for skeletal defects and cancer as a result of an overactivated BMP signaling pathway. In the field of rare diseases, progress in molecular diagnostics led to the recognition that variants linked to autosomal-dominant neurodegenerative diseases of later onset can, in the context of biallelic inheritance, cause devastating neurodevelopmental disorders and infantile or childhood-onset neurodegeneration. TOR1A-associated arthrogryposis multiplex congenita 5 (AMC5) is a rare neurodevelopmental disorder arising from biallelic variants in TOR1A, a gene that in the heterozygous state is associated to torsion dystonia-1 (DYT1 or DYT-TOR1A), an early-onset dystonia with reduced penetrance. BACKGROUND: Intravenous fluids and vasopressor agents are commonly used in early resuscitation of patients with sepsis, but comparative data for prioritizing their delivery are limited.
METHODS: In an unblinded superiority trial conducted at 60 U.S. centers, we randomly assigned patients to either a restrictive fluid strategy (prioritizing vasopressors and lower intravenous fluid volumes) or a liberal fluid strategy (prioritizing higher volumes of intravenous fluids before vasopressor use) for a 24-hour period. Randomization occurred within 4 hours after a patient met the criteria for sepsis-induced hypotension refractory to initial treatment with 1 to 3 liters of intravenous fluid. We hypothesized that …

Within this family of neoplasms, outcomes for retroperitoneal sarcomas (RPS) are currently limited given a lack of effective therapies. In this review, we focus on immunotherapy and its relationship with the common RPS histologic subtypes. Although initial outcomes for RPS patients with immune checkpoint inhibition alone have been somewhat disappointing, subsequent analyses on histologies, the tumor microenvironment, sarcoma immune class, tumor infiltrating lymphocytes and genetic analysis for tumor mutational burden have yielded insight into the interplay between sarcomas and immunotherapy. Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare hematologic malignancy with historically poor outcomes and no worldwide consensus treatment approach. Unique among most hematologic malignancies for its frequent cutaneous involvement, BPDCN can also invade other extramedullary compartments, including the central nervous system. Generally affecting older adults, many patients are unfit to receive intensive chemotherapy, and although hematopoietic stem cell transplantation is preferred for younger, fit individuals, not all are eligible. One recent therapeutic breakthrough is that all BPDCNs express CD123 (IL3Rα) and that this accessible surface marker can be pharmacologically targeted. Animals ranging from mosquitoes to humans often vary their feeding behavior when infected or merely exposed to pathogens. These so-called “sickness behaviors” are part of the innate immune response with many consequences, including avoiding orally transmitted pathogens. Fully understanding the role of this ubiquitous behavior in host defense and pathogen evolution requires a quantitative account of its impact on host and pathogen fitness across environmentally relevant contexts. This study investigated the effects of continuous head cooling on 5-km running time-trial (TT) performance in hot conditions. Six male and four female triathletes completed two experimental sessions consisting of two 10-minute runs at 50% and 70% O2max followed by a 5-km TT in the heat (32.0±0.3 °C, 50.1±1.2% RH). Novel phenotypes can come about through a variety of mechanisms including standing genetic variation from a founding population. Cave animals are an excellent system in which to study the evolution of novel phenotypes such as loss of pigmentation and eyes. Asellus aquaticus is a freshwater isopod crustacean found in Europe and has both a surface and a cave ecomorph which vary in multiple phenotypic traits. An orange eye phenotype was previously revealed by F2 crosses and backcrosses to the cave parent within two examined Slovenian cave populations. There are two methods that I specifically focused on and learned about which are "Super Kelp" and "Green Gravel." Supervised practice is a prerequisite to becoming a registered dietitian. Research suggests that environmental and social factors may affect dietary choices. This focus group research aimed to gather opinions from dietetic interns to understand what factors related to supervised practice, if any, affected their dietary habits. Qualitative data were collected via seven recorded virtual focus groups in which trained moderators facilitated a discussion using a series of controlled questions. Human-mediated environmental challenges are severely impacting local and global levels of biodiversity. Phenotypic plasticity can play a major role in aiding population persistence because plasticity can buffer populations from novel challenges. Given this role, it is possible for the degree of plasticity to evolve rapidly as it may place phenotypes near, but not exactly on, an adaptive peak for the new environment. Here, we asked if behavioral plasticity evolves following the introduction of a novel predator.
