Accurately completing DNA replication when two forks converge is essential to genomic stability. The RecBCD helicase–nuclease complex plays a central role in completion by promoting resection and joining of the excess DNA created when replisomes converge. chi sequences alter RecBCD activity and localize with crossover hotspots during sexual events in bacteria, yet their functional role during chromosome replication remains unknown. Here, we use two-dimensional agarose gel analysis to show that chi induces replication on substrates containing convergent forks. Restoring damaged and disturbed lands can be difficult and expensive with increasing drought and changing climatic conditions. Wicking boxes or other irrigation forms can improve restoration efforts' success. These technologies provide a consistent water supply, which often enhances seedling growth and survival in dry conditions. However, current water catchment technologies are limited and often difficult to use. Factors such as production costs, size, and labor needed to install these devices typically make current water catchment technologies inefficient for large-scale use.

SARS-CoV-2, a novel betacoronavirus strain, has caused a pandemic that has claimed the lives of nearly 6.7M people worldwide. Vaccines and medicines are being developed around the world to reduce the disease spread, fatality rates, and control the new variants. Understanding the protein-protein interaction mechanism of SARS-CoV-2 in humans, and their comparison with the previous SARS-CoV and MERS strains, is crucial for these efforts. These interactions might be used to assess vaccination effectiveness, diagnose exposure, and produce effective biotherapeutics. Background: Dual-specificity phosphatases (DUSPs) can dephosphorylate both tyrosine and serine/threonine residues of their substrates and regulate T cell-mediated immunity and autoimmunity. The aim of this study was to investigate the potential roles of DUSPs in ankylosing spondylitis (AS).
Methods: Sixty AS patients and 45 healthy controls were enrolled in this study. Associations of gene expression of 23 DUSPs in peripheral T cells with inflammatory cytokine gene expression and disease activity of AS were analyzed. Finally, we investigated whether the characteristics of AS are developed in DUSP-knockout mice.

Plants are attacked by multiple insect pest species and insect herbivory can alter plant defense mechanisms. The plant defense responses to a specific herbivore may also contribute to the herbivore growth/survival on plants. Feeding by one insect species can modulate the plant defenses, which can either facilitate or hamper the colonization of subsequent incoming insects. However, little is known about the effect of sequential herbivory on sorghum plants. In this study, we demonstrate that a specialist aphid, sugarcane aphid (SCA; Melanaphis sacchari) grows faster on sorghum than a generalist aphid species, greenbug (GB; Schizaphis graminum). Lower Mississippi River islands experience intense flood regimes that result in a supply of sediment and seeds from upstream. The islands are some of the last remaining floodplain habitats in the Delta region, and surveying their botanical diversity is important for learning about floodplain plant community structure. From June 2020 through December 2021, I conducted a flora of six Lower Mississippi River islands from Helena-West Helena, Arkansas, to Greenville, Mississippi. I collected 2,310 plant specimens for a total of 491 taxa, including five taxa not previously recorded in Arkansas and seven not previously recorded in Mississippi. From mid-July to late-August …

Row crop agriculture in Arkansas relies predominantly on groundwater resources for irrigation. This places great demand on the Mississippi River Valley Alluvial Aquifer (MRVAA), which is in a current state of decline. The Eastern Arkansas region produces commodity crops including soybeans, rice, corn, cotton, and peanuts, all of which utilize irrigation for optimal productivity. Conservation practices that reduce groundwater use include precision land leveling, multiple inlet rice irrigation, and computerized hole size selection for furrow irrigation. Automated irrigation systems and new management practices may help address aquifer depletion, labor shortages, and irrigation management challenges. This study evaluated the impact of various automated irrigation systems on commercial farms that produce maize, rice, and soybean. It also modeled how Early Cascade Rice Irrigation Shutoff (ECIS) performs under a range of field conditions. Growers observed various benefits of using automated systems other than irrigation savings or yield increase. Timers and remote shutoff are the most practical and beneficial options for growers willing to adopt some level of automation. Developers and users of artificial-intelligence-based tools for automatic contouring and treatment planning in radiotherapy are expected to assess clinical acceptability of these tools. However, what is 'clinical acceptability'? Quantitative and qualitative approaches have been used to assess this ill-defined concept, all of which have advantages and disadvantages or limitations. The approach chosen may depend on the goal of the study as well as on available resources. In this paper, we discuss various aspects of 'clinical acceptability' and how they can move us toward a standard for defining clinical acceptability of new autocontouring and planning tools.

The C4 grass pearl millet is one of the most drought tolerant cereals and is primarily grown in marginal areas where annual rainfall is low and intermittent. It was domesticated in sub-Saharan Africa, and several studies have found that it uses a combination of morphological and physiological traits to successfully resist drought. This review explores the short term and long-term responses of pearl millet that enables it to either tolerate, avoid, escape, or recover from drought stress. The work is a taxonomic revision of 28 species of the genus Chrysina Kirby (Scarabaeidae: Rutelinae: Rutelini) found in Panama (25), Colombia (2), and Ecuador (3). Chrysina tricolor (Ohaus), Chrysina chalcothea (Bates), and Chrysina cupreomarginata (F. Bates) are new records for Panama. A new country record from northern Colombia is confirmed for Chrysina mercedesae Barria. Chrysina gaitalica Curoe and Hawks and Chrysina galbina Hawks are discovered at new localities in Panama; females of both species are discovered and described. Chrysina aurora (Bates) known from other localities in the west of the country, is rediscovered in Veraguas province 147 years after …

Mouse mammary glands comprise ductal trees, which are lined by epithelial cells and have one opening at the tip of each nipple. The epithelial cells play a major role in mammary gland function and are the origin of most mammary tumors. Introducing genes of interest into mouse mammary epithelial cells is a critical step in evaluating gene function in epithelial cells and generating mouse mammary tumor models. This goal can be accomplished through the intraductal injection of a viral vector carrying the genes of interest into the mouse mammary ductal tree. Background: Cancer patients with financial toxicity experience psychological distress and often miss medical appointments and quit treatments early, which could be a barrier to the effective management of oral chemotherapy drugs at home. Purpose: Overexpression of c-Met protein and epidermal growth factor receptor (EGFR) mutations can co-occur in non-small-cell lung cancer (NSCLC), providing strong rationale for dual targeting. Telisotuzumab vedotin (Teliso-V), a first-in-class antibody-drug conjugate targeting c-Met, has shown a tolerable safety profile and antitumor activity as monotherapy. Herein, we report the results of a phase Ib study (ClinicalTrials.gov identifier: NCT02099058) evaluating Teliso-V plus erlotinib, an EGFR tyrosine kinase inhibitor (TKI), in patients with c-Met-positive (+) NSCLC.

Purpose: To evaluate the outcomes and cost-effectiveness of the Children's Oncology Group Guideline recommendation for breast cancer (BC) screening using mammography (MAM) and breast magnetic resonance imaging (MRI) in female chest-irradiated childhood Hodgkin lymphoma (HL) survivors. Digital breast tomosynthesis (DBT), increasingly replacing MAM in practice, was also examined.
Methods: Life years (LYs), quality-adjusted LYs (QALYs), BC mortality, health care costs, and false-positive screen frequencies of undergoing annual MAM, DBT, MRI, MAM + MRI, and DBT + MRI from age 25 to 74 years were estimated by microsimulation. Although tissue-resident memory T (TRM) cells specific for previously encountered pathogens have been characterized, the induction and recruitment of brain TRM cells following immune therapy has not been observed in the context of glioblastoma. Here, we show that T cells expressing fibrinogen-like 2 (FGL2)–specific single-chain variable fragments (T-αFGL2) can induce tumor-specific CD8+ TRM cells that prevent glioblastoma recurrence. These CD8+ TRM cells display a highly expanded T cell receptor repertoire distinct from that found in peripheral tissue. When adoptively transferred to the brains of either immunocompetent or T cell-deficient naïve mice, these CD8+ TRM cells reject glioma cells. Mechanistically, T-αFGL2 …

BACKGROUND: Food prescription programs are gaining interest from funders, policy makers, and healthcare payers as a way to provide value-based care. A small body of research suggests that such programs effectively impact health outcomes; however, the quality of existing studies is variable, and most studies use small samples. This study attempts to address these gaps by utilizing a quasi-experimental design with non-equivalent controls, to evaluate clinical outcomes among participants enrolled in a food prescription program implemented at scale.

Human T-cell leukemia virus type 1 (HTLV-1), a retrovirus, is the causative agent for adult T cell leukemia/lymphoma and many other human diseases. Accurate and high throughput detection of HTLV-1 virus integration sites (VISs) across the host genomes plays a crucial role in the prevention and treatment of HTLV-1-associated diseases. Here, we developed DeepHTLV, the first deep learning framework for VIS prediction de novo from genome sequence, motif discovery, and cis-regulatory factor identification. We demonstrated the high accuracy of DeepHTLV with more efficient and interpretive feature representations. Plants are attacked by multiple insect pest species and insect herbivory can alter plant defense mechanisms. The plant defense responses to a specific herbivore may also contribute to the herbivore growth/survival on plants. Feeding by one insect species can modulate the plant defenses, which can either facilitate or hamper the colonization of subsequent incoming insects. However, little is known about the effect of sequential herbivory on sorghum plants. In this study, we demonstrate that a specialist aphid, sugarcane aphid (SCA; Melanaphis sacchari) grows faster on sorghum than a generalist aphid species, greenbug (GB; Schizaphis graminum). Objective: This study examined the similarities in metabolic and physiological demands of a fire ground test (FGT) and a live fire training evolution. Methods: Twenty-seven firefighters completed either a FGT (n = 13) or a live fire training evolution (n = 14). Salivary samples were collected pre, immediately post, and 30-minutes post FGT and live fire training evolution, and analyzed for cortisol, uric acid, and interleukin-1β (IL-1β). We investigated whether acute ischemic preconditioning (IPC) would affect upper limb maximal strength performance in resistance-trained men. Using a counterbalanced randomized crossover design, fifteen men (29.9 ± 5.9 yrs.; 86.3 ± 9.6 kg; 8.0 ± 5.0 yrs. resistance training experience) performed one-repetition maximum (1-RM) bench press tests on three different occasions: control, 10 min post-IPC or 10 min post-placebo (SHAM). One-way analysis of variance showed that the post-IPC condition increased (P < 0.0001) 1-RM loads compared to both control and post-SHAM (control 113.3 ± 15.9 kg vs. SHAM 113.9 ± 15.8 kg vs. IPC 115.7 ± 15.6 kg), while control and SHAM did not differ (P > 0.05). A new species Scorpio granulomanus sp. n. is described and illustrated from Dibbeen Forest, Jerash Governorate, Jordan. The new species is compared with the previously recorded species of the genus Scorpio L., 1758 in the Middle East; it can be distinguished from all other congeners by its very large, pointed granules on the dorsoexternal surface of the chela manus, and an untypically elongated chela manus.

Comfrey (Symphytum officinale) is a very hardy perennial herb adaptable to most conditions. Comfrey grows best in partial to full sun in moist, fertile soil. Comfrey is propagated by root cuttings or crown divisions. Comfrey can be planted throughout the growing season but establishes best when planted in the spring. Maintain moderate soil moisture, especially when plants are young. Once established, comfrey outcompetes weeds and does not have any reported insect or disease issues in Utah. The changes in stream discharge extremes due to temperature and seasonality are key metrics in assessing the effects of climate change on the hydrological cycle. While scaling is commonly applied to temperature and precipitation due to the physical connections between temperature and moisture (i.e., Clausius–Clapeyron), the scaling rate of stream discharge extremes to air and dewpoint temperatures has not been evaluated. To address this challenge, we assess the scaling rates between stream discharge and air temperature and between stream discharge and dewpoint temperature in Utah using a well-designed statistical framework. Locations of 74,755 vascular plant vouchers representing 2,425 species were digitized into a spatial database from the Atlas of the Vascular Plants of Utah (Albee et al. 1988). Source maps consisted of 1:6,000,000-scale shaded relief maps of Utah with points representing collection locations by species. Location points, representing 1 or more specimens, were transposed onto these maps from the approximately 400,000 herbarium records of 3 major universities and federal land management agencies.
