Maternal RNAs are stored from minutes to decades in oocytes throughout meiosis I arrest in a transcriptionally quiescent state. Recent reports, however, propose a role for nascent transcription in arrested oocytes. Whether arrested oocytes launch nascent transcription in response to environmental or hormonal signals while maintaining the meiosis I arrest remains undetermined. We test this by integrating single-cell RNA sequencing, RNA velocity, and RNA fluorescence in situ hybridization on C. elegans meiosis I arrested oocytes. We identify transcripts that increase as the arrested meiosis I oocyte ages, but rule out extracellular signaling through ERK MAPK and nascent transcription as a …

The control of virulence two-component gene regulatory system (CovRS) is critical to the pathogenesis of many medically important streptococci. In emm1 group A streptococci (GAS), CovR directly binds the promoters of numerous GAS virulence factor-encoding genes. Elimination of CovS phosphatase activity increases CovR phosphorylation (CovR~P) levels and abrogates GAS virulence. Given the emm type-specific diversity of CovRS function, in this study we used chromatin immunoprecipitation sequencing (ChIP-seq) to define global CovR DNA occupancy in the wild-type emm3 strain MGAS10870 (medium CovR~P) and its CovS phosphatase-negative derivative 10870-CovS-T284A (high CovR~P). In the wild-type emm3 strain, 89% of the previously identified emm1 …

Shark-Diving Tourism In The Macaronesian Archipelagos: Challenges And Opportunities, Pedro Gonzáles Mantilla
ITSA 2022 Gran Canaria - 9th Biennial Conference: Corporate Entrepreneurship and Global Tourism Strategies After Covid 19
The Macaronesian region is typically heavily exploited by fisheries; however, in recent years, marine wildlife tourism has become popular and a shark-diving industry has emerged, potentially presenting an alternative for the sustainable use of sharks. Combining a literature review with interviews with dive operators conducting shark encounters in the Macaronesian archipelagos, we provide an overview of the challenges and conservation potential of shark-diving tourism for these territories. Owing to the regular presence of important shark species for tourism and the growth of the scuba-diving industry, shark-diving has potential to expand over the region. Yet, the overlap between European industrial fishing …
Does Chronic Low Back Pain Influence Breathing Mechanics And Diaphragm Positioning? A Pilot Study, Lindsey Wensel
Does Chronic Low Back Pain Influence Breathing Mechanics And Diaphragm Positioning? A Pilot Study, Lindsey Wensel
Theses and Dissertations
Background and objective: Chronic low back pain (CLBP) is a complex musculoskeletal condition and often the source of the pain is not clear. A correlation has been found to exist between incidences of low back pain (LBP) and those with respiratory diseases such as COPD. This could give reason to believe that the sequence of events could be reversed, and LBP could elicit changes in respiratory function. The purpose of this study is to investigate if CLBP has an influence on breathing mechanics and the positioning of the diaphragm in the trunk. Methods: Volunteers were recruited between the ages of …

Variability Among Individual Male Runners Influences Cumulative Loading More Than Foot Strike Type, Tamarie Trotter
Theses and Dissertations
Foot strike type affects running mechanics and may influence overuse injury occurrence. Measuring the interaction between cumulative load and foot strike type may provide additional information that could eventually help with understanding injury mechanisms. The purpose of this study was to determine how foot strike type affects cumulative loading in runners, and to determine if individual metrics change throughout a 5-kilometer sub-maximal run. 30 participants ran on an instrumented treadmill for 5 km at 3.15 m/s with their preferred foot strike type (14 rearfoot strike, 16 non-rearfoot strike). Stride rate, foot strike angle, loading rate, per stride and per km …

Introduction: Glyphosate is a widely used, non-selective herbicide. Glyphosate and glyphosate-based herbicides (GBHs) are considered safe for non-target organisms and environmentally benign at currently allowed environmental exposure levels. However, their increased use in recent years has triggered questions about possible adverse outcomes due to low dose chronic exposure in animals and humans. While the toxicity of GBHs has primarily been attributed to glyphosate, other largely unstudied components of GBHs may be inherently toxic or could act synergistically with glyphosate. Thus, comparative studies of glyphosate and GBHs are needed to parse out their respective toxicity. Methods: We performed such a comparative …

The notion that errors in protein synthesis are universally harmful to the cell has been questioned by findings that suggest such mistakes may sometimes be beneficial. However, how often these beneficial mistakes arise from programmed changes in gene expression as opposed to reduced accuracy of the translation machinery is still unclear. A new study published in JBC shows that some bacteria have beneficially evolved the ability to mistranslate specific parts of the genetic code, a trait that allows improved antibiotic resistance.

Coronavirus disease-19 (COVID-19), caused by Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2), is an ongoing pandemic-endemic. In the United States, it remains the third leading cause of death in 2022. SARS-CoV-2, an RNA virus, has a genome prone to rapid mutation, generating variant species capable of resisting therapies and vaccination efforts. However, it has been estimated that more than 40% of SARS-CoV-2 infections may be asymptomatic, which calls into question any biological differences between the asymptomatic and afflicted groups. Additionally, Respiratory Syncytial Virus (RSV) is the most significant cause of bronchiolitis and asthma in neonates, with an increasing burden on the …

The transcription factor ΔNp63 regulates epithelial stem cell function and maintains the integrity of stratified epithelial tissues by acting as transcriptional repressor or activator towards a distinct subset of protein-coding genes and microRNAs. However, our knowledge of the functional link between ∆Np63 transcriptional activity and long non-coding RNAs (lncRNAs) expression is quite limited. Here, we show that in proliferating human keratinocytes ∆Np63 represses the expression of the lncRNA NEAT1 by recruiting the histone deacetylase HDAC1 to the proximal promoter of NEAT1 genomic locus. Upon induction of differentiation, ∆Np63 down-regulation is associated by a marked increase of NEAT1 RNA levels, resulting …

The balance between bone-resorbing osteoclasts and bone-forming osteoblasts is essential for the bone remodeling process. This study aimed to investigate the effect of Ishophloroglucin A (IPA) isolated from Ishige okamurae on the function of osteoclasts and osteoblasts in vitro. First, we demonstrated the effect of IPA on osteoclastogenesis in receptor activator of nuclear factor κB ligand (RANKL)-induced RAW 264.7 cells. IPA inhibited the tartrate-resistant acid phosphatase (TRAP) activity and osteoclast differentiation in RANKL-induced RAW 264.7 cells. Moreover, it inhibited the RANKL-induced osteoclast-related factors, such as TRAP, matrix metalloproteinase-9 (MMP-9), and calcitonin receptor (CTR), and transcription factors, such as nuclear factor …

We present molecular data on Isometrus maculatus (DeGeer, 1778) for the first time from India, Japan, and Taiwan based on the COI gene and justify the cosmopolitan nature of this species. We also address the first population of I. maculatus found in Teliamura, Tripura, India and give its detailed description and comparison with the rest of the Isometrus species from India. Furthermore, we discuss the justification for considering Sri Lanka as the true home of I. maculatus.
Investigating The Activity Of Alternative Warheads For Targeted Covalent Inhibition Of The Inhibitor Vertebrate Lysozyme Protein From Pseudomonas Aeruginosa, Katie Hambrick
Master of Science in Chemical Sciences Theses
Pseudomonas aeruginosa (P. aeruginosa) is a Gram-negative bacterium that causes blood and lung infections in hospital environments due to its ability to survive on improperly sterilized medical equipment. P. aeruginosa has developed several multi-drug resistance mechanisms that make it very difficult to treat with current antibiotics.1 This presents the need for a new class of antibiotics that cannot be overcome by P. aeruginosa’s mechanisms of resistance.
The primary goal of this project was to develop a small library of inhibitors that could later be incorporated into lead compounds for novel antibiotic drug discovery. One of P. …

Historically, wildfire and tribal burning practices played important roles in shaping ecosystems throughout the Klamath Siskiyou Bioregion of northern California and southern Oregon. Over the past several decades, there has been increased interest in the application of fire for forest management through the implementation of prescribed fires within habitats that are used by a diversity of migrant and resident land birds. While many bird species may benefit from habitat enhancements associated with wildfires, cultural burning, and prescribed fire, individuals may face direct or indirect harm. In this study, we analyzed the timing of breeding and molting in 11 species of …

By knowing the nutritional status of a plant, one can better manage and use it for ethnobotanical purposes. Various nutrients are accumulated as osmotica in plants of stressful environments. The type and quantity of nutrients present in plants also determine the effectivity of medicine obtained from that plant. Hence, practical utilization of these plants as food or medicines needs to explore the exact nature and presence of the nutrient ions for discrimination of their toxic or medicinal nature. In the present study, some important nutrients in a number of xerophytic plants from Cholistan desert of Pakistan were quantified. Leave, stem …

Mutagenesis and genome instability are characteristics of cancer cells. People afflictedwith Bloom’s syndrome due to mutations in the BLM gene suffer from reduced life expectancy, accelerated aging, sensitivity to sun exposure, immune system abnormalities and increased risk of cancer. The predisposition to cancer is typically due to abnormal DNA repair that results in chromosome breakage and rearrangements. The homologous recombination factor Sgs1, which is a RecQ-like helicase and an ortholog of the human BLM helicase, functions primarily in DNA repair, but has also been implicated in other cellular functions like replication. Several functions of BLM such as its involvement in …

Toxoplasma gondii is a globally distributed zoonotic protozoan parasite. Infection with T. gondii can cause congenital toxoplasmosis in developing fetuses and acute outbreaks in the general population, and the disease burden is especially high in South America. Prior studies found that the environmental stage of T. gondii, oocysts, is an important source of infection in Brazil; however, no studies have quantified this risk relative to other parasite stages. We developed a Bayesian quantitative risk assessment (QRA) to estimate the relative attribution of the two primary parasite stages (bradyzoite and oocyst) that can be transmitted in foods to people in …

Candidatus Atelocyanobacterium thalassa (or UCYN-A) has emerged as a focal point due to its streamlined metabolism, intimate partnership with a haptophyte host, and broad distribution. Here, we explore the environmental parameters that govern UCYN-A’s presence at the San Pedro Ocean Time-series (SPOT), its host specificity, and statistically significant interactions with non-host eukaryotes from 2008-2018. 16S and 18S rRNA gene sequences were amplified by “universal primers” from monthly samples and resolved into Amplicon …

Freshwater systems are often breeding grounds for harmful algal blooms (HABs), although they are more dominant in ponds and lakes due to the prevailing conditions in those bodies of water. Therefore, the monitoring, modeling, and management of HABs requires knowledge of the complex interrelationship between factors that influence HABs and their detrimental effect on the ecosystem. High concentrations of chlorophyll a are often used to measure algal blooms in bodies of water. Generally, water samples are collected from the field and the concentration of chlorophyll a is measured in a laboratory and compared to water quality standards in order to …

Current microbiome studies have shown that the maintenance of homeostasis betweenmicrobial populations (e.g. bacteria, viruses) and the host immune system (e.g. innate immune molecules) is necessary for balancing health and disease outcomes within the host. These studies most often utilize vertebrate models; however, research in this field can benefit from diverse model systems that facilitate our ability to conduct experiments to identify phylogenically conserved rules influencing homeostasis in the gut of animals. The Dishaw has developed the use of a filter-feeding marine invertebrate chordate, Ciona robusta, to model such fundamental interactions[1]–[6]. While most biological diversity and functional contribution within microbiomes …
Utilization Of Detonation Nanodiamonds: Nanocarrier For Gene Therapy In Non-Small Cell Lung Cancer, Allan E. Gutierrez
Utilization Of Detonation Nanodiamonds: Nanocarrier For Gene Therapy In Non-Small Cell Lung Cancer, Allan E. Gutierrez
USF Tampa Graduate Theses and Dissertations
Lung cancer is one of the most prevalent and high mortality cancers in the world. Two different categories of lung cancer are non-small cell lung cancer which makes up 85% of cases, and small cell lung cancer which makes up the remaining 15% of cases. A significant risk factor that can lead to lung cancer is cigarette smoking (CS) however non-smoking cases i.e., secondhand CS, exposure to certain toxins and family history are on the rise. Both the main types of lung cancer have different system of stages, different ranges of treatment, and consequently low survival rates. Unfortunately, detection and …
Resource Recovery From Crew Metabolic Wastes In Lunar And Martian Habitats: Reuse Of Effluent From An Anaerobic Membrane Bioreactor Treating Fecal Organic Matter As Fertilizer Source For Plants, Alexandra A. Smith
USF Tampa Graduate Theses and Dissertations
As humans set their sights on exploration of the Moon and Mars human focused challenges like providing potable water, supplying nutritious food to the astronaut crew, and managing their waste present themselves. There are no readily available resources on the Moon and Mars, therefore nothing should be considered unusable including the organic wastes (i.e., feces and urine) produced by the crew. Fecal matter is not currently recycled and is treated as solid waste. After collection, it is discarded out of the International Space Station where it is incinerated upon re-entry. This approach will not work on the Moon’s surface. There …
